Hepatocellular cancer (HCC) has been reported to belong to one of the highly vascularized solid tumours accompanied with angiogenesis of human umbilical vein endothelial cells (HUVECs). KDM5A, an attractive drug target, plays a critical role in diverse physiological processes. Thus, this study aims to investigate its role in angiogenesis and underlying mechanisms in HCC. ChIP-qPCR was utilized to validate enrichment of H3K4me3 and KDM5A on the promotor region of miR-433, while dual luciferase assay was carried out to confirm the targeting relationship between miR-433 and FXYD3. Scratch assay, transwell assay, Edu assay, pseudo-tube formation assay and mice with xenografted tumours were conducted to investigate the physiological function of KDM5A-miR-433-FXYD3-PI3K-AKT axis in the progression of HCC after loss- and gain-function assays. KDM5A p-p85 and p-AKT were highly expressed but miR-433 was down-regulated in HCC tissues and cell lines. Depletion of KDM5A led to reduced migrative, invasive and proliferative capacities in HCC cells, including growth and a lowered HUVEC angiogenic capacity in vitro. Furthermore, KDM5A suppressed the expression of miR-433 by demethylating H3K4me3 on its promoterregion. miR-433 negatively targeted FXYD3. Depleting miR-433 or re-expressing FXYD3 restores the reduced migrative, invasive and proliferative capacities, and lowers the HUVEC angiogenic capacity caused by silencing KDM5A. Therefore, KDM5A silencing significantly suppresses HCC tumorigenesis in vivo, accompanied with down-regulated miR-433 and up-regulated FXYD3-PI3K-AKT axis in tumour tissues. Lastly, KDM5A activates the FXYD3-PI3K-AKT axis to enhance angiogenesis in HCC by suppressing miR-433. 相似文献
Psychostimulant (methamphetamine, cocaine) use disorders have a genetic component that remains mostly unknown. We conducted genome-wide quantitative trait locus (QTL) analysis of methamphetamine stimulant sensitivity. To facilitate gene identification, we employed a Reduced Complexity Cross between closely related C57BL/6 mouse substrains and examined maximum speed and distance traveled over 30 min following methamphetamine (2 mg/kg, i.p.). For maximum methamphetamine-induced speed following the second and third administration, we identified a single genome-wide significant QTL on chromosome 11 that peaked near the Cyfip2 locus (LOD = 3.5, 4.2; peak = 21 cM [36 Mb]). For methamphetamine-induced distance traveled following the first and second administration, we identified a genome-wide significant QTL on chromosome 5 that peaked near a functional intronic indel in Gabra2 coding for the alpha-2 subunit of the GABA-A receptor (LOD = 3.6–5.2; peak = 34–35 cM [66–67 Mb]). Striatal cis-expression QTL mapping corroborated Gabra2 as a functional candidate gene underlying methamphetamine-induced distance traveled. CRISPR/Cas9-mediated correction of the mutant intronic deletion on the C57BL/6J background to the wild-type C57BL/6NJ allele was sufficient to reduce methamphetamine-induced locomotor activity toward the wild-type C57BL/6NJ-like level, thus validating the quantitative trait variant (QTV). These studies show the power and efficiency of Reduced Complexity Crosses in identifying causal variants underlying complex traits. Functionally restoring Gabra2 expression decreased methamphetamine stimulant sensitivity and supports preclinical and human genetic studies implicating the GABA-A receptor in psychostimulant addiction-relevant traits. Importantly, our findings have major implications for studying psychostimulants in the C57BL/6J strain—the gold standard strain in biomedical research. 相似文献

Lizard beetles (Erotylidae, Languriinae, Languriini) are known as stem borers of plants and contain agricultural pests and endangered species, but their species–host plant associations have been poorly documented. Here we investigated the larval host plants of two species of the genus Tetraphala Strum, T. collaris (Crotch) and Tetraphala sp. occurring in Taiwan. Females of T. collaris excavated living leafstalks and stems of the herbaceous dicot, Sambucus chinensis (Adoxaceae) using their mandibles for oviposition. We observed the eggs and early-instar larvae inside and nearby oviposition holes and late-instar larvae inside stems, suggesting that T. collaris uses living leafstalks and stems of S. chinensis as oviposition substrate and the larvae tunnel into stems with feeding on the tissues. Similarly, females of Tetraphala sp. excavated living leafstalks of the fern, Pteris wallichiana (Pteridaceae) using their mandibles for oviposition. We observed the eggs and early-instar larvae inside and nearby oviposition holes. When reared in laboratory, a larva reached adulthood inside the leafstalk. These results indicated that Tetraphala sp. uses living leafstalks of Pt. wallichiana as oviposition substrate and the larvae complete their development within. This study revealed that the genus Tetraphala contains both fern- and dicot-users during larval period. Further study is needed to clarify the evolutionary process of host plant use of languriines. Non-native invasive plant species influence plant community composition and competitively eradicate native species. However, there is doubt regarding how global invasive species increase and explosively interfere with native plants. Invasive plants always have strong allelopathic potential. In this study, allelopathic effects of switchgrass on redroot pigweed and crabgrass growth were investigated by field and laboratory experiments. Within a 0.4-m distance of switchgrass, density and shoot biomass of native species were significantly suppressed in the field, with 95.1% and 93.0% inhibition on density of redroot pigweed and crabgrass and with 99.0% and 97.7% inhibition on shoot biomass, respectively, during the third growing season. Significant inhibitory effects on shoot and root biomass were observed at the 5:5 (switchgrass–native species) proportion in glass bottles, by 41.57% and 51.21% for shoot and root biomass of redroot pigweed and by 33.42% and 56.95% for shoot and root biomass of crabgrass, respectively. Results of a glass bottle experiment showed that shoot and root biomass of redroot pigweed and crabgrass could be significantly inhibited by contact with switchgrass root. Results of a Petri dish experiment showed that aqueous extracts of switchgrass significantly inhibited germination process of both species at high concentrations, with 90.74% and 18.62% inhibition on germination rate and plumule length of redroot pigweed and with 63.59%, 16.38%, and 19.92% inhibition on germination rate, plumule, and radicle lengths of crabgrass, respectively, at the concentration of 0.1 g·mL?1. This report demonstrated that switchgrass had allelopathic effects on redroot pigweed and crabgrass growth.
Infants often experience complementary food-induced diarrhoea (CFID), which occurs when infants switch from breast milk to solid foods. The relative abundances of Prevotella and Rothia were higher in stools of infants with CFID, while the relative abundances of Enterococcus and Escherichia were higher in healthy infants. The abundance of Lactobacillus spp. normally found in breast milk fed to infants with CFID was significantly reduced, and Enterococcus spp. were less abundant when diarrhoea occurred. Furthermore, Lactobacillus and Enterococcus were present as shared bacteria in both mother and infant, and they were considered potential anti-CFID probiotics as their relative abundances in breast milk were negatively correlated to infant CFID. Kyoto encyclopedia of genes and genomes (KEGG) functional analysis showed that the function of amino acid metabolism differed between infants with CFID and healthy infants. Therefore, CFID might be related to the decomposition of proteins in food supplements. The screening revealed seven hydrolytic casein and five hydrolytic casein and rice protein isolates from 320 suspected Lactobacillus and Enterococcus isolates. The animal experiments demonstrated that a mixture of five isolates effectively hydrolysed the casein and rice protein and prevented diarrhoea in young rats. Thus, the occurrence of CFID was found to be closely related to the intestinal and breast milk microbiota, and bacteria that could assist in the digestion of cereal proteins were involved in CFID. 相似文献
