Nano neodymium oxide induces massive vacuolization and autophagic cell death in non-small cell lung cancer NCI-H460 cells 总被引:3,自引:0,他引:3
Neodymium, a rare earth element, was known to exhibit cytotoxic effects and induce apoptosis in certain cancer cells. Here we show that nano-sized neodymium oxide (Nano Nd2O3) induced massive vacuolization and cell death in non-small cell lung cancer NCI-H460 cells at micromolar equivalent concentration range. Cell death elicited by Nano Nd2O3 was not due to apoptosis and caspases were not involved. Electron microscopy and acridine orange staining revealed extensive autophagy in the cytoplasm of the cells treated by Nano Nd2O3. Autophagy induced by Nano Nd2O3 was accompanied by S-phase cell cycle arrest, mild disruption of mitochondrial membrane potential, and inhibition of proteasome activity. Bafilomycin A1, but not 3-MA, induced apoptosis while inhibiting autophagy. Our results revealed a novel biological function for Nano Nd2O3 and may have implications for the therapy of non-small cell lung cancer. 相似文献

Honey bees, Apis mellifera L., often thought to be extremely susceptible to insecticides in general, exhibit considerable variation in tolerance to pyrethroid insecticides. Although some pyrethroids, such as cyfluthrin and lambda-cyhalothrin, are highly toxic to honey bees, the toxicity of tau-fluvalinate is low enough to warrant its use to control parasitic mites inside honey bee colonies. Metabolic insecticide resistance in other insects is mediated by three major groups of detoxifying enzymes: the cytochrome P450 monooxygenases (P450s), the carboxylesterases (COEs), and the glutathione S-transferases (GSTs). To test the role of metabolic detoxification in mediating the relatively low toxicity of tau-fluvalinate compared with more toxic pyrethroid insecticides, we examined the effects of piperonyl butoxide (PBO), S,S,S-tributylphosphorotrithioate (DEF), and diethyl maleate (DEM) on the toxicity of these pyrethroids. The toxicity of the three pyrethroids to bees was greatly synergized by the P450 inhibitor PBO and synergized at low levels by the carboxylesterase inhibitor DEF. Little synergism was observed with DEM. These results suggest that metabolic detoxification, especially that mediated by P450s, contributes significantly to honey bee tolerance of pyrethroid insecticides. The potent synergism between tau-fluvalinate and PBO suggests that P450s are especially important in the detoxification of this pyrethroid and explains the ability of honey bees to tolerate its presence. 相似文献

An ideal expression algorithm should be able to tell truly different expression levels with small false positive errors and be robust to assay changes. We propose two algorithms. PQN is the non-central trimmed mean of perfect match intensities with quantile normalization. DQN is the non-central trimmed mean of differences between perfect match and mismatch intensities with quantile normalization. The quantiles for normalization can be either empirical or theoretical. When array types and/or assay change in a study, the normalization to common quantiles at the probe set level is essential. We compared DQN, PQN, RMA, GCRMA, DCHIP, PLIER and MAS5 for the Affymetrix Latin square data and our data of two sets of experiments using the same bone marrow but different types of microarrays and different assay. DEAD-box RNA解旋酶参与RNA转录、前体mRNA剪切、核糖体发生、核质运输、蛋白质翻译、RNA降解等重要的生命活动.根据本室在S-Mo17Rf3Rf3cDNA芯片研究中,检测到花粉发育后期RNA解旋酶上调表达的结果,应用RACE技术从S-Mo17Rf3Rf3花粉中克隆得到该RNA解旋酶基因全长cDNA,命名为ZmRH2并在GenBank注册登记 (DQ327709).序列分析表明:该cDNA全长1 652bp,从第163 bp开始到1 386bp含有一个开放阅读框,编码407个氨基酸.其编码的蛋白质具有DEAD-box RNA解旋酶特有的9个保守模体,与水稻、拟南芥和豌豆中的DEAD-box RNA解旋酶的氨基酸序列存在着很高的同源性.RT-PCR分析表明,该基因在近等基因系S-Mo17Rf3Rf3和S-Mo17rf3rf3的叶、根、和雌穗中的表达没有差异,但在花丝和花粉中有明显差异. 相似文献

以秋华柳为试验材料,采用水培试验方式,设置CK(0 mg·L-1 Cd2+)、T1(2 mg·L-1 Cd2+)、T2(10 mg·L-1Cd2+)、T3(20 mg·L-1Cd2+)、T4(50 mg·L-1Cd2+)5种镉处理浓度,通过对秋华柳根系活力,叶、韧皮部、木质部和根部的镉含量,以及Ca、Mg、Mn、Zn、Fe 5种常规金属元素含量的测定,研究了不同浓度镉胁迫下秋华柳根系活力及Ca、Mg、Mn、Zn、Fe 等金属元素含量的变化.结果表明:1)秋华柳根系活力随着镉处理浓度的增加而逐渐下降,当镉浓度≥10 mg·L-1时,根系活力与对照相比显著下降.2)随着镉处理浓度的增加,秋华柳叶中Fe的积累受到显著抑制;韧皮部Mg、Mn、Fe的积累受到显著抑制;木质部Ca、Mg、Mn、Zn、Fe 等5种常规金属元素的积累无显著性差异;根部5种常规金属元素的吸收和积累受到显著抑制,表现出镉对其他金属元素积累的拮抗作用.其中,50 mg·L-1的镉胁迫下,根部Zn的积累量降幅最大,受Cd的抑制最明显.3)各处理组Fe的转移系数与对照相比均无显著性差异;Ca、Mg、Mn、Zn的转移系数均高于对照,且在一定的镉处理水平上差异显著.4)镉胁迫下,秋华柳根部镉的积累量与Ca的积累量呈显著负相关,与其他4种常规金属元素的积累量呈极显著负相关,说明根部常规金属元素的变化可作为秋华柳受镉毒害程度的指示之一. 相似文献

根毛是植物体吸收养分的重要器官, 自然条件下根毛的寿命很短, 仅能存活2–3周, 随即脱落死亡。以模式植物拟南芥(Arabidopsis thaliana)根毛为材料, 对根毛死亡的细胞学特征进行了报道。结果发现, 根毛衰老死亡后细胞内的原生质体发生了收缩, 并在胞质中观察到凝集物的出现; 通过原位末端标记(TUNEL)检测, 发现幼根上的根毛细胞核DNA发生了片段化。上述结果表明, 拟南芥根毛的衰老死亡很可能是植物体自主调控的程序性细胞死亡(PCD)。另外, 当根毛衰老死亡后,细胞核大多会迁移到靠近根毛基部的位置, 且正常的长管状根毛发生旋转扭曲。 相似文献

A comparative evaluation of five different cell-disruption methods for the release of recombinant hepatitis B core antigen
(HBcAg) from Escherichia coli was investigated. The cell disruption techniques evaluated in this study were high-pressure homogenization, batch-mode bead
milling, continuous-recycling bead milling, ultrasonication, and enzymatic lysis. Continuous-recycling bead milling was found
to be the most effective method in terms of operating cost and time. However, the highest degree of cell disruption and amounts
of HBcAg were obtained from the high-pressure homogenization process. The direct purification of HBcAg from the unclarified
cell disruptate derived from high-pressure homogenization and bead milling techniques, using batch anion-exchange adsorption

The fish gill is a multifunctional organ responsible for gas exchange and ionic regulation. It is hypothesized that both morphological
and functional differentiation can be found in the gills of the aquatic air-breathing fish, Trichogaster leeri. To test this, we used the air-breathing fish, Trichogaster leeri, to investigate various morphological/functional parameters. First, we evaluated the importance of performing the aquatic
surface respiration behavior in T. leeri. A reduced survival rate was observed when fish were kept in the restrained cages in hypoxic conditions. On the gross anatomy
of gills, we found evidence of both morphological and functional modification in the first and the second gills and are responsible
for ionic regulation. There were large-bore arterioarterial shunts in the fourth gill arch. It is specialized for the transport
of oxygenated blood and is less responsive to environmental stress. In addition, the anterior and the posterior gills differed
in the Na+, K+-ATPase activity upon ionic stresses. That is, only the Na+, K+-ATPase activity of the anterior two gills was up-regulated significantly in the deionized water. Lastly, we found that the
number of mitochondria-rich cells in the first and the second gills increased following ionic stress and no difference was
found in the third and the fourth gills following such an exposure. These results supported the hypothesis that there are
morphological and functional differences between anterior and posterior gill arches within the air-breathing Trichogaster leeri. In contrast, no significant difference was found among gills in gross anatomy, filament density and Na+, K+-ATPase activity in the non-air-breather, Barbodes schwanenfeldi. 相似文献

Glycerol (30%) inhibited or delayed the adsorption of Shigella bacteriophage on its host organism, S. flexneri II; glycerol also inhibited or delayed the burst of phage, whether or not adsorption was carried out in the presence of glycerol. Studies of the mechanisms of these effects showed that viscosity and osmotic shock probably were not responsible for either phenomenon. The inhibition of adsorption, however, was proportional to the concentration of glycerol, and appeared to be a function of the hydroxyl groups on the glycerol molecule. The inhibition of burst seemed to be related to the osmotic pressure outside the bacterial cells. 相似文献
