Abstract: The palaeohistological study of the calcified internal organ of Axelrodichthys araripensis Maisey, 1986, a coelacanthiform from the Lower Cretaceous of Brazil (Crato (Aptian) and Santana (Albian) formations of the Araripe Basin), shows that the walls of this organ consist of osseous blades of variable thickness separated from each other by the matrix. This indicates that, in the living individuals, the walls were reinforced by ossified plates, probably separated by conjunctive tissue. This calcified sheath present in Axelrodichthys, as well as in other fossil coelacanths, lies in ventral position relative to the gut and its single anterior opening is located under the opercle, suggesting a direct connection with the pharynx or the oesophagus. The calcified organ of Axelrodichthys, like that of other fossil coelacanths, is here regarded as an ‘ossified lung’ and compared with the ‘fatty lung’ of the extant coelacanth Latimeria. The reinforcement of the pulmonary walls by the overlying osseous blades could be interpreted as a means of adapting volumetric changes in the manner of bellows, a necessary function for ventilation in pulmonary respiration. Quantitative analysis of the distribution of dispersed spores and pollen (sporomorphs) has been used to assess temporal floral variation through the Middle Jurassic Ravenscar Group (Aalenian–Bathonian), North Yorkshire, UK. Aalenian, Bajocian and Bathonian strata possess relatively distinct sporomorph and palynofacies assemblages, which potentially reflect a dynamic history regarding the nature of parent vegetation. Specifically, Aalenian palynofloras are composed of a heterogeneous mixture of conifers, ferns, simple monosulcate pollen producers, sphenophytes and Caytoniales; Bajocian palynofloras are codominated by conifers and ferns; and Bathonian palynofloras are highly rich and contain assemblages of abundant ferns, conifers, lycophytes, pteridosperms/conifers and Caytoniales. Individual‐ and sample‐based rarefaction demonstrates that Bathonian samples are richer than Aalenian and Bajocian samples. Temporal variations in assemblages are a result of long‐term depositional and possible climatic fluctuations through the Middle Jurassic. Ordinations of sporomorph data using non‐metric multidimensional scaling (NMDS) demonstrate that short‐term variations between samples are largely governed by taphonomic biases as a result of slight changes in depositional processes, which give rise to highly variable catchment areas that supply deposits with sporomorphs. Long‐term compositional changes are apparent in sporomorph assemblages regardless of lithology/local depositional environments, suggesting that long‐term variations are more substantial than short‐term variations and potentially include genuine regional temporal changes in parent vegetation. Relating sporomorph assemblages with their respective depositional environments and relative catchment area sizes using lithological and palynofacies information suggests that the basin interior was occupied by mostly low‐standing species and extrabasinal vegetation was dominated by coniferous taxa. Comparisons of the dispersed sporomorph and plant megafossil records indicate that both fossil assemblages reflect different aspects of the palaeoflora due to a multitude of taphonomic and ecological biases. Genetic analyses are an important contribution to wildlife reintroductions, particularly in the modern context of extirpations and ecological destruction. To address the complex historical ecology of the sea otter (Enhydra lutris) and its failed 1970s reintroduction to coastal Oregon, we compared mitochondrial genomes of pre-extirpation Oregon sea otters to extant and historical populations across the range. We sequenced, to our knowledge, the first complete ancient mitogenomes from archaeological Oregon sea otter dentine and historical sea otter dental calculus. Archaeological Oregon sea otters (n = 20) represent 10 haplotypes, which cluster with haplotypes from Alaska, Washington and British Columbia, and exhibit a clear division from California haplotypes. Our results suggest that extant northern populations are appropriate for future reintroduction efforts. This project demonstrates the feasibility of mitogenome capture and sequencing from non-human dental calculus and the diverse applications of ancient DNA analyses to pressing ecological and conservation topics and the management of at-risk/extirpated species. 相似文献

Neurospora crassa on agar slants in ampules were frozen by direct immersion into liquid nitrogen and stored at −196 °C for 0–5.5 years. The survival, estimated as percentage germination of conidia, after warming, varied between 81.1 and 97.6%. Although there was a slight decline in survival immediately after the freezing treatment, no further significant decline has been detected during 5.5 years storage. The regression coefficients, (0.43, control, and 0.97, liquid nitrogen storage for 5.5 years), do not predict extinction of viability. 相似文献

Cerebral artery vasospasm is a major cause of death and disability in patients experiencing subarachnoid hemorrhage (SAH). Currently, little is known regarding the impact of SAH on small diameter (100-200 microm) cerebral arteries, which play an important role in the autoregulation of cerebral blood flow. With the use of a rabbit SAH model and in vitro video microscopy, cerebral artery diameter was measured in response to elevations in intravascular pressure. Cerebral arteries from SAH animals constricted more (approximately twofold) to pressure within the physiological range of 60-100 mmHg compared with control or sham-operated animals. Pressure-induced constriction (myogenic tone) was also enhanced in arteries from control animals organ cultured in the presence of oxyhemoglobin, an effect independent of the vascular endothelium or nitric oxide synthesis. Finally, arteries from both control and SAH animals dilated as intravascular pressure was elevated above 140 mmHg. This study provides evidence for a role of oxyhemoglobin in impaired autoregulation (i.e., enhanced myogenic tone) in small diameter cerebral arteries during SAH. Furthermore, therapeutic strategies that improve clinical outcome in SAH patients (e.g., supraphysiological intravascular pressure) are effective in dilating small diameter cerebral arteries isolated from SAH animals. 相似文献

Adult male rats received s.c. injections of either corticosterone (10 mg in 250 microL sesame oil; n = 8) or vehicle (250 microL; n = 8) daily for 3 weeks. A third group of rats served as intact controls (n = 4). Brains were stained using a Golgi-Cox procedure and pyramidal neurons in layer II-III of medial prefrontal cortex were drawn; dendritic morphology was quantified in three dimensions. Sholl analyses demonstrated a significant redistribution of apical dendrites in corticosterone-treated animals: the amount of dendritic material proximal to the soma was increased relative to intact rats, while distal dendritic material was decreased relative to intact animals. Thus, chronic glucocorticoid administration dramatically reorganized apical arbors in medial prefrontal cortex. Enhanced extinction of aversive memories by high-frequency stimulation of the rat infralimbic cortex
Electrical stimulation of the rodent medial prefrontal cortex (mPFC), including the infralimbic cortex (IL), immediately prior to or during fear extinction training facilitates extinction memory. Here we examined the effects of high-frequency stimulation (HFS) of the rat IL either prior to conditioning or following retrieval of the conditioned memory, on extinction of Pavlovian fear and conditioned taste aversion (CTA). IL-HFS applied immediately after fear memory retrieval, but not three hours after retrieval or prior to conditioning, subsequently reduced freezing during fear extinction. Similarly, IL-HFS given immediately, but not three hours after, retrieval of a CTA memory reduced aversion during extinction. These data indicate that HFS of the IL may be an effective method for reducing both learned fear and learned aversion. 相似文献
