In the possum a marked sex difference has been found in the steroids in adrenal venous plasma. Four 5 beta-pregnane and four 5 alpha (beta) androstane derivatives together with ten 4-ene-3-keto steroids were isolated from the adrenal venous plasma of the female and definitively identified by gas chromatography-mass spectrometry. The major reduced steroids were: 5 beta-pregnane-3 alpha,17 alpha-diol-20-one and 5 beta-pregnane-3 alpha,17 alpha,20 alpha-triol, at concentrations of 52 +/- 12 micrograms/100 ml and 44 +/- 8 micrograms/100 ml mean +/- SEM respectively. The concentration of cortisol was 198 +/- 47 micrograms/100 ml. The concentration of the 2 reduced steroids in peripheral plasma were approx. 100 times less. In contrast the adrenal venous plasma of a male contained 14 steroids of which only three, found in trace amounts, were reduced. The results confirm previous in vitro observations that reduced steroids are produced by the adrenocortical special zone, which is only present in the female. The physiological significance of the presence of reduced steroids of adrenocortical origin in the circulation of the female possum is discussed.  相似文献

Nipple reconstruction is performed as a last stage in breast reconstruction following mastectomy. Various methods of nipple reconstruction have been described, most of them utilizing either free composite grafts or local flaps. The main problem encountered using either method is the gradual absorption and flattening of the nipple. The technique we used in reconstructing 22 nipples, in preference over the various methods accepted in breast reconstruction, achieves a long-standing, protruding nipple constructed from two large local flaps raised from an S-shape design. The technique is simple and permits freedom in choosing the height of the nipple, even in the presence of a mastectomy scar. The size of the nipple thereby constructed is in excess of what was expected. Shrinkage occurs during the first 2 months, and the resulting size is more than adequate. The areola is reconstructed by a full-thickness skin graft harvested from a nonhairy area of the upper inner thigh. The local flaps lack the necessary color, which is achieved by tattooing.  相似文献

Identification of the residues in human CD4 critical for the binding of HIV   总被引:52,自引:0,他引:52  
The CD4 molecule is a T cell surface glycoprotein that interacts with high affinity with the envelope glycoprotein of the human immunodeficiency virus, HIV, thus serving as a cellular receptor for this virus. To define the sites on CD4 essential for binding to gp120, we produced several truncated, soluble derivatives of CD4 and a series of 26 substitution mutants. Quantitative binding analyses with the truncated proteins demonstrate that the determinants for high affinity binding lie solely with the first 106 amino acids of CD4 (the V1 domain), a region having significant sequence homology to immunoglobulin variable regions. Analysis of the substitution mutants further defines a discrete binding site within this domain that overlaps a region structurally homologous to the second complementarity-determining region of antibody variable domains. Finally, we demonstrate that the inhibition of virus infection and virus-mediated cell fusion by soluble CD4 proteins depends on their association with gp120 at this binding site.  相似文献

Sequence analysis of a substantial part of the polymerase gene of the murine coronavirus MHV-A59 revealed the 3' end of an open reading frame (ORF1a) overlapping with a large ORF (ORF1b; 2733 amino acids) which covers the 3' half of the polymerase gene. The expression of ORF1b occurs by a ribosomal frameshifting mechanism since the ORF1a/ORF1b overlapping nucleotide sequence is capable of inducing ribosomal frameshifting in vitro as well as in vivo. A stem-loop structure and a pseudoknot are predicted in the nucleotide sequence involved in ribosomal frameshifting. Comparison of the predicted amino acid sequence of MHV ORF1b with the amino acid sequence deduced from the corresponding gene of the avian coronavirus IBV demonstrated that in contrast to the other viral genes this ORF is extremely conserved. Detailed analysis of the predicted amino acid sequence revealed sequence elements which are conserved in many DNA and RNA polymerases.  相似文献

Muscle spindle stretch responses (cat gastrocnemius muscles) were studied when bothsteady stretch andsmall near-threshold random stretch determined the Ia impulse sequence. Statistical properties of the inter-impulse-intervals gave some insight into the Ia encoder mechanism. Superimposed random stretch of mean velocities vel below 5 mm s–1 did not change the mean discharge rate, but the width of the Ia interspike interval distribution was clearly increased. Raising the stretch velocity further ( vel > 5 mm s–1) led to an additional increase in the distribution width, finally reaching values of 0.6 for the coefficient of variation. The shapes of the impulse interval histograms changed from symmetrical to positively skewed ones. The 1st order serial correlation coefficient of the interval sequence shifted to slightly more negative values with increasing vel; on the average, ther 1,2-value varied between zero and –0.2. The data were discussed in relation to current ideas on the mechanism of impulse initiation in the Ia terminal ending. They provide evidence that a combination of multiple encoder sites located in the myelinated terminal branches and a separate pathway for large static and small-amplitude dynamic stretch is not very likely. A model is proposed as to how the whole tree of myelinated axons functions as a single encoder site.  相似文献

There has recently been great interest in the demography of primitive human populations. This can often be dealt with successfully by the use of demographic theory and model life tables; however, small populations present problems not encountered in large national censuses. This paper discusses some of these problems and illustrates the determination of model life tables in anthropology with a particular population, the Yanomama Indians. The concern is for the methods to be used, and their applicability, and not with the specific population illustrating them. By applying theoretical models, one can derive information that is otherwise unavailable but is necessary in the important biomedical, social, and ecological understanding of primitive societies.  相似文献

Vitellogenin is the serum precursor of the yolk proteins -lipovitellin,rß-lipovitellin, and phosvitin. The precursor canbe dissociated to produce the yolk proteins only by proteolyticenzymatic action, to which it is very susceptible. Denaturationin sodium dodecyl sulfate, combined with reduction of disulfidebridges and blocking of thiols, yields a complex with a molecularweight of 200,000 to 250,000. -Lipovitellin contains three polypeptides,with molecular weights of about 135,000, 105,000, and 40,000,and rß-lipovitellin is composed of two polypeptidechains with molecular weights of 135,000 and 30,000. The 40,000subunit of -lipovitellin and both rß-lipovitellinsubunits are phosphopeptides We tested RNA isolated from the liver of estrogen-treated roostersfor mRNA activity in a cell-free reticulocyte system. The vitellogeninmRNA has a sedimentation coefficient greater than 28S and thuscontains enough information to code for a long polypeptide chain.Estrogen administration to roosters induces the appearance ofvitellogenin and a lowdensity lipoprotein, the syntheses ofwhich are not coordinated. The course of vitellogenin synthesiswas calculated from accumulation and turnover data, and it wasfound that from about 25 hr after estradiol-17rß administrationthe rate of vitellogenin synthesis increases linearly for severaldays, paralleling an increase in vitellogenin-synthesizing polysomes.Thus, we estimate a constant translation rate of about 8 aminoacids per ribosome per sec. A "memory" effect is observed when a second hormone dose isgiven some time after the vitellogenin induced by the firstdose has disappeared from the blood. After the second dose vitellogeninsynthesis is detected sooner, and its initial increase is morerapid, than after the first dose. Although the synthesis ofvitellogenin starts 3 to 4 hr after the second as well as afterthe first injection, the rate of synthesis after the first injectionincreases much more slowly during the first 15 hr than duringthe subsequent period of linear accumulation, whereas afterthe second injection the linear increase in the rate of synthesisbegins immediately after the lag period of 3 to 4 hr. The "memory"effect is undiminished even 50 days after the first hormonedose; thus, the causative factor either is very stable or issynthesized in great excess during the first stimulation. Whenthe second injection is given during the descending part ofthe turnover curve, an increase in vitellogenin synthesis isobserved within 3.5 hr. There are thus at least three different effects of estradiol;(i) the "memory" effect, which probably is due to commitmentor differentiation of vitellogenin-synthesizing cells; (ii)the effect that causes the committed cells to give full responseafter the 3- to 4-hr lag period; and (iii) the effect that causesthe immediate response. To explain these results we suggestthat committed cells can synthesize vitellogenin mRNA only duringa certain period of the cell cycle.  相似文献
