Abstract Carriage of Neisseria meningitidis B:4:P1.15 was higher among non-secretors during a school outbreak of meningitis; non-secretors had lower levels of anti-meningococcal salivary IgM. Flow cytometry was used to assess effects of secretor and non-secretor saliva on binding of B:4:P1.15 to buccal epithelial cells: (1) to assess inhibition by IgA and IgM; and (2) to assess contributions of salivary antibodies to inhibitory activities. Greater inhibition was obtained with secretor saliva: pooled ( P = 0.049); fresh ( P = 0.0001). Purified IgA ( P = 0.02) and IgM ( P = 0.03) were equally inhibitory. After absorption of anti-meningococcal antibodies, there was still significant inhibitory activity in the pools: secretors ( P = 0.018); non-secretors ( P = 0.005). These results indicate that both secretory immunoglobulins and other factors contribute to protection against colonisation by meningococci and might explain the increased carriage of B:4:P1.15 in this population.  相似文献

The purpose of this study was to examine the effects of unilateral isometric leg extension strength training on the strength and integrated electromyogram (IEMG) of both the trained and untrained limbs at multiple joint angles. A training (TRN) group [nine women; mean (SD) age, 20(1) years] exercised for 6 weeks with isometric leg extensions at 80% of maximal isometric torque. A control (CTL) group [eight women; 21(1) years] did not exercise. The training was performed three times per week on a Cybex II isokinetic dynamometer at a joint angle where the lever arm was 0.79 rad below the horizontal plane. The subjects were tested pre- and posttraining for maximal unilateral isometric torque in both limbs at joint angles of zero, 0.26, 0.79,1.31, and 1.57 rad below the horizontal plane. Bipolar surface electrodes were used to record the IEMG of the vastus lateralis (VL) and vastus medialis (VM) during the isometric tests. Three univariate (torque, IEMG-VL, and IEMG-VM) four-way (group x time x limb x angle) mixed factorial ANOVAs were used to analyze the data. The results indicated joint angle specificity for isometric torque in the TRN group only, with significant increases in torque at 0.79 (P = 0.0004) and 1.31 (P = 0.0039) rad. No significant increases in torque were found in the untrained limb of the TRN group or in either limb of the CTL group. Similarly, there were no significant changes in IEMG as a result of the training for the VL or VM. The joint-angle-specific strength increases without concomitant increases in IEMG were hypothesized to result from joint-angle-specific decreases in antagonistic co-contraction and/or preferential hypertropy of the quadriceps femoris at specific levels of the muscle group.  相似文献

Using 5-[14C] methyltetrahydropteroylglutamate and [3H] pteroylglutamate it was found that vitamin B12 dependent methyltransferase can be inhibited in vivo in mice by nitrous oxide. Examination of the liver [3H] folates showed as expected that both controls and nitrous oxide treated mice had synthesised the reduced 5-methyl form. However, the controls had converted [3H] almost exclusively into a polyglutamate with virtually no monoglutamate present, while nitrous oxide treated mice had failed to add glutamates to over half of the incorporated folate. Thus in vivo demethylation may be necessary prior to polyglutamate biosynthesis and act as a method of controlling folate accumulation, or be used to create a concentration gradient with the circulating folate.  相似文献

A theory is given that allows inbreeding coefficients to be calculated exactly for populations with overlapping generations. Emphasis is placed on providing equations well suited for computer iteration. Both monoecious and dioecious populations are considered and family size is not restricted to being Poisson. One-locus and two-locus inbreeding coefficients are evaluated, although the reader may omit the two-locus sections. The exact treatment is shown to be preferable to approximate treatments in that it applies to both early and late generations for all populations sizes. Inbreeding effective numbers found by the exact treatment are compared to various approximate numbers, and the approximate values are found to be generally very good.  相似文献

A series of artificial fertilizations using rainbow smelt Osmerus mordax were conducted, each designed to assess the fertilization success of a female's eggs with milt from one to five males. Mean egg fertilization success increased and variance in fertilization success decreased when more males contributed to a constant volume of milt. This may indicate the presence of an Allee effect. The results suggest that potential changes in egg fertilization success due to a decrease in the number of males, in addition to changes in egg production due to a decrease in the number of females, can affect estimates of maximum sustainable harvest rates, and recovery of depleted populations.  相似文献

We build a model with large-scale demographic consequences for migratory species. The model operates where four elements co-occur, and we rely on empirical research using migratory birds to demonstrate them. First, breeding ranges have internal structure flowing from natal philopatry. Second, fecundity varies geographically. Third, populations of different breeding provenances commingle during winter. And fourth, a population-limiting carrying capacity operates during winter. In the absence of breeding season population-limitation, only the breeding population with maximum fecundity persists. Consequently, some potential breeding areas that offer suitable and productive habitat are bereft of breeding birds because of the interplay between the geographical fecundity gradient and the shared winter quarters. Where breeding season population-limitation also plays a role for at least one population, one (or more) breeding population becomes permanently depressed, resulting in a density well below the carrying capacity of the productive breeding habitat that is occupied. In either case, not all populations fare equally well, despite net positive breeding season productivity. Changes in winter carrying capacity, for example habitat degradation in winter quarters, can lead to uneven effects on geographically defined breeding populations, even though there has been no change in the circumstances of the breeding range.  相似文献

The feed profile of glucose during fedbatch cultivation could be used to influence the retention of the periplasmic product ZZ-cutinase. An increased feed rate led to a higher production rate but also to an increased specific leakage, which reduced the periplasmic retention. Three growth rates: 0.3, 0.2 and 0.1 h(-1) where studied and resulted in 20, 9 and 6%, respectively, of the total ZZ-cutinase accumulating in the medium. It was also shown that leakage during fedbatch production of a Fab fragment was also influenced by the feed rate in a similar manner to ZZ-cutinase. If intracellular product accumulation is desired the advantage of a high productivity, resulting from a high substrate feed rate, is diminished because of a reduced product retention. Biochemical analysis revealed that the growth rate, resulting from a glucose limited feed, influenced the outer membrane protein compositions with respect to OmpF and LamB, whilst OmpA was largely unaffected. As the feed rate increased the amount of total outer membrane protein decreased. When ZZ-cutinase was produced there were further reductions in outer membrane protein accumulation, by 82, 100 and 22% for OmpF, LamB and OmpA, respectively, and the total reduction was almost 60% with a high product formation rate. We suggest that the reduced titre of the outer membrane proteins, OmpF and LamB, may have contributed to a reduced ability for the cell to retain recombinant protein secreted to the periplasm.  相似文献

The correlation between alleles at a pair of genetic loci is a measure of linkage disequilibrium. The square of the sample correlation multiplied by sample size provides the usual test statistic for the hypothesis of no disequilibrium for loci with two alleles and this relation has proved useful for study design and marker selection. Nevertheless, this relation holds only in a diallelic case, and an extension to multiple alleles has not been made. Here we introduce a similar statistic, R(2), which leads to a correlation-based test for loci with multiple alleles: for a pair of loci with k and m alleles, and a sample of n individuals, the approximate distribution of n(k - 1)(m - 1)/(km)R(2) under independence between loci is chi((k-1)(m-1))(2). One advantage of this statistic is that it can be interpreted as the total correlation between a pair of loci. When the phase of two-locus genotypes is known, the approach is equivalent to a test for the overall correlation between rows and columns in a contingency table. In the phase-known case, R(2) is the sum of the squared sample correlations for all km 2 x 2 subtables formed by collapsing to one allele vs. the rest at each locus. We examine the approximate distribution under the null of independence for R(2) and report its close agreement with the exact distribution obtained by permutation. The test for independence using R(2) is a strong competitor to approaches such as Pearson's chi square, Fisher's exact test, and a test based on Cressie and Read's power divergence statistic. We combine this approach with our previous composite-disequilibrium measures to address the case when the genotypic phase is unknown. Calculation of the new multiallele test statistic and its P-value is very simple and utilizes the approximate distribution of R(2). We provide a computer program that evaluates approximate as well as "exact" permutational P-values.  相似文献

The New Zealand native legume flora are represented by four genera, Sophora, Carmichaelia, Clianthus, and Montigena. The adventive flora of New Zealand contains several legume species introduced in the 19th century and now established as serious invasive weeds. Until now, nothing has been reported on the identification of the associated rhizobia of native or introduced legumes in New Zealand. The success of the introduced species may be due, at least in part, to the nature of their rhizobial symbioses. This study set out to address this issue by identifying rhizobial strains isolated from species of the four native legume genera and from the introduced weeds: Acacia spp. (wattles), Cytisus scoparius (broom), and Ulex europaeus (gorse). The identities of the isolates and their relationship to known rhizobia were established by comparative analysis of 16S ribosomal DNA, atpD, glnII, and recA gene sequences. Maximum-likelihood analysis of the resultant data partitioned the bacteria into three genera. Most isolates from native legumes aligned with the genus Mesorhizobium, either as members of named species or as putative novel species. The widespread distribution of strains from individual native legume genera across Mesorhizobium spp. contrasts with previous reports implying that bacterial species are specific to limited numbers of legume genera. In addition, four isolates were identified as Rhizobium leguminosarum. In contrast, all sequences from isolates from introduced weeds aligned with Bradyrhizobium species but formed clusters distinct from existing named species. These results show that native legume genera and these introduced legume genera do not have the same rhizobial populations.  相似文献
