We have studied inflammatory reactions in the mesenteric tissue of mice infected with Schistosoma mansoni. Perivascular tissue contained diffuse infiltrates of macrophages, eosinophilic granulocytes and lymphocytes. Angiogenesis in the perivascular adipose tissue was associated with superficial plasmacytogenic foci. Polypoid structures were occasionally formed adjacent to inflammatory foci in the adipose tissue, organized around loops of capillaries, with terminal formation of a glomerular capillary network embedded in connective tissue, covered by plasmacytes. We conclude that these structures are specialized milky spots dedicated to active plasmacytogenesis and antibody secretion into the peritoneal cavity of schistosome-infected mice. 相似文献

Research and clinical observations during the past six decades have shown that: 1. Iron promotes cancer cell growth; 2. Hosts attempt to withhold or withdraw iron from cancer cells; and 3. Iron is a factor in prevention and in therapy of neoplastic disease. Although normal and neoplastic cells have similar qualitative requirements for iron, the neoplastic cells have more flexibility in acquisition of the metal. Excessive iron levels in animals and humans are associated with enhanced neoplastic cell growth. In invaded hosts, cytokine-activated macrophages increase intracellular ferritin retention of the metal, scavenge iron in areas of tumor growth, and secrete reactive nitrogen intermediates to effect efflux of nonheme iron from tumor cells. Procedures associated with lowering host intake of excess iron can assist in prevention and in management of neoplastic disease. Chemical methods for prevention of iron assimilation by neoplastic cells are being developed in experimental and clinical protocols. The antineoplastic activity of a considerable variety of chemicals, as well as of radiation, is modulated by iron. The present article focuses on recent findings and suggests directions for further cancer-iron research. 相似文献

A design is proposed for case-control studies in which selection of subjects for full variable ascertainment is based jointly on disease status and on easily obtained "screening" variables that may be related to the disease. Recruitment of subjects follows an independent Bernoulli sampling scheme, with recruitment probabilities set by the investigator in advance. In particular, the sampling can be set up to achieve, on average, frequency matching, provided prior estimates of the disease rates or odds ratios associated with screening variables such as age and sex are available. Alternatively--for example, when studying a rare exposure--one can enrich the sample with certain categories of subject. Following such a design, there are two valid approaches to logistic regression analysis, both of which allow for efficient estimation of effects associated with the screening variables that were allowed to bias the recruitment. The statistical properties of the estimators are compared, both for large samples, based on asymptotics, and for small samples, based on simulations. 相似文献

Increasing data suggest that mitochondrial dysfunction may be an important early component of nephrotoxin-induced changes in renal cell function and viability. This study was designed to obtain more detailed information about the effects on several basic bioenergetic parameters of the direct interaction of Hg2+ with renal cortical mitochondria in vitro as a necessary prelude to studies of mitochondrial functional changes after treatment with mercuric chloride in vivo. Beginning at a threshold level of 2 nmol of Hg2+/mg of mitochondrial protein, Hg2+ induced marked stimulation of State 4 respiration, mild inhibition of State 3 respiration, and 2,4-dinitrophenol uncoupled respiration, a striking increase in atractyloside-insensitive ADP uptake and stimulation of both basal- and Mg2+-activated oligomycin-sensitive mitochondrial ATPase activity. These effects of Hg2+ could be prevented and reversed by the sulfhydryl reagent dithioerythritol and by albumin but were not affected by Mg2+. Detailed studies on the addition of HgCl2 to the preparation at different stages of the mitochondrial isolation procedure demonstrated that the presence of other proteins decreased mitochondrial Hg2+ binding, that the Hg2+ was not readily washed off the mitochondria by nonprotein-containing solutions, and that prolonged exposure of mitochondria to Hg2+ during the isolation procedure did not markedly alter its functional effects or their reversibility as assessed on the final mitochondrial preparation. These data provide an important basis for critically assessing the changes in function of mitochondria isolated after in vivo treatment with mercuric chloride. 相似文献

The mRNA for alcohol dehydrogenase (ADH) in D. melanogaster has been identified by translation in a cell-free system. The in vitro synthesized polypeptide, specifically precipitated by anti-ADH antibody, has identical subunit molecular weight (25,000 daltons) and tryptic peptide profile to the in vivo synthesized ADH. The poly A containing ADH-mRNA has been purified by specific precipitation of ADH-polysomes using anti-ADH antibody and S. aureus. Transformation of E. coli with the dA-tailed ADH-mRNA-complementary DNA hybrid annealed to the dT-tailed pBR322 yielded one plasmid which has been identified as the ADH-cDNA clone. The identification involved hybridization selection of ADH-mRNA and in vitro translation, in situ hybridization to the Adh locus on salivary gland polytene chromosomes and DNA sequencing. This ADH-cDNA plasmid contains 349 bases of the C-terminal protein coding and 180 bases of the 3' untranslated region. 相似文献

Rat soleus muscles were ectopically innervated by implanting a foreign nerve in an endplate-free region of muscle and, 2–3 weeks later, cutting the original nerve. The junctional, 16 S form of acetylcholinesterase (AChE) and focal staining for AChE disappeared from the old endplate region within a few days after denervation. In muscles with an ectopic nerve, but not in paired control muscles, 16 S AChE and focal staining were restored in the old endplate region 1–2 weeks after denervation even though nerve fibers could not be detected in that region. These results suggest that the nerve exerts a local effect, specifying the site at which junctional AChE appears, and a nonlocal effect, perhaps mediated by muscle activity, regulating the amount of junctional AChE. 相似文献

A fetus with a 46,XX,r(17) karyotype identified initially by amniocentesis and confirmed by banding studies is described. No significant phenotypic abnormalities were identified despite isolation of the ring chromosome from multiple fetal tissues. Interactions of two tubicolous, deposit-feeding polychaetes, Clymenella torquata (Leidy) and Polydora ligni Webster, with a filter-feeding bivalve, Gemma gemma (Totten), were examined to test the functional group hypothesis that small, filter-feeding bivalves with large young should attain high densities with tube-builders of any trophic mode because the large young escape predation by tube-builders. The prediction was not supported by Polydora and Gemma, since the two species have not been reported to coexist in high densities in nature. In laboratory experiments, Polydora reduced recruitment and adult survival of Gemma relative to a control. Polydora preyed on juvenile clams and reduced the time which adult clams had their siphons in the feeding position, relative to a control.For Clymenella and Gemma, the functional group hypothesis made the correct prediction (i.e. the two species are reported to coexist in high densities in nature), but for the wrong reason. The size of Gemma's offspring is not related to the intensity of Clymenella predation. Clymenella does not ingest Gemma since the worm does not usually feed from the zone occupied by the clam. In contrast to Polydora, Clymenella did not reduce the time which clams fully extend and open their siphons relative to a control. In laboratory experiments Clymenella increased adult bivalve growth, survival after one year, and recruit size after one month. Clam life history characteristics were similar in the Clymenella and Clymenella plus Polydora treatments because the number of surviving Polydora was reduced by a negative interaction with Clymenella.The tube-builders had different effects on the filter-feeder, Gemma, because these tube-building species differ in feeding position, diet and behavior. Thus, the functional group hypothesis was too general to predict and explain the species interactions and distributions. 相似文献
