碱性蛋白酶生产菌——地衣芽孢杆菌经丝裂霉素C或紫外线处理,均可诱导释放噬菌体,电镜观察表明噬菌体头部外廓呈六边形,有不收缩的尾部(头部40nm×40nm,尾部107×5,7nm),噬菌体BLL1 DNA对限制酶Eco R Ⅰ,HindⅢ和Bam H Ⅰ敏感,分别切割成8,15,和9个片段,经电泳法测定。噬菌体DNA分子量约相当于23.4kb,根据解链温度计算出噬菌体的G+C含量约为31.2摩尔%,噬菌体提纯的外壳蛋白经SDS-聚丙烯酰胺凝胶电泳呈现5条主带,其分子量分别约为78000,72000,55000,39000,35000。  相似文献

A pilot-scale NiiMi system for river landscape water treatment has been steadily operated for more than one year. The aim of the study was to investigate the spatial distributions of biofilm properties and water flow pattern in NiiMi system. Results showed that spatial distributions regularities of volatile suspended solid (VSS), respiration intensity, protein, polysaccharide, viable cell number and dehydrogenase activity were consistent with the aqueous rate. In addition, the viable cell, protein and polysaccharide specific dehydrogenase decreased with depth. Moreover, the protein: polysaccharide ratio increased with depth. Fourier transform infrared (FT-IR) spectroscopy indicated that protein was one of the components of extracellular polymeric substance (EPS). Gel filtration chromatography (GFC) analysis indicated that EPS had a broader MW distribution than that in the effluent water. Trace studies indicated that the occurrence of a dead zone volume of 18.7%, the top left section was the dead zone of the NiiMi system.  相似文献

The accumulation and deposition of beta‐amyloid (Aβ) is a key neuropathological hallmark of Alzheimer's disease (AD). Histone deacetylases (HDACs) are promising therapeutic targets for the treatment of AD, while the specific HDAC isoforms associated with cognitive improvement are poorly understood. In this study, we investigate the role of HDAC3 in the pathogenesis of AD. Nuclear HDAC3 is significantly increased in the hippocampus of 6‐ and 9‐month‐old APPswe/PS1dE9 (APP/PS1) mice compared with that in age‐matched wild‐type C57BL/6 (B6) mice. Lentivirus ‐mediated inhibition or overexpression of HDAC3 was used in the hippocampus of APP/PS1 mice to investigate the role of HDAC3 in spatial memory, amyloid burden, dendritic spine density, glial activation and tau phosphorylation. Inhibition of HDAC3 in the hippocampus attenuates spatial memory deficits, as indicated in the Morris water maze test, and decreases amyloid plaque load and Aβ levels in the brains of APP/PS1 mice. Dendritic spine density is increased, while microglial activation is alleviated after HDAC3 inhibition in the hippocampus of 9‐month‐old APP/PS1 mice. Furthermore, HDAC3 overexpression in the hippocampus increases Aβ levels, activates microglia, and decreases dendritic spine density in 6‐month‐old APP/PS1 mice. In conclusion, our results indicate that HDAC3 negatively regulates spatial memory in APP/PS1 mice and HDAC3 inhibition might represent a potential therapy for the treatment of AD.  相似文献

Parental selection is crucial for hybrid breeding, but the methods available for such a selection are not very effective. In this study, a 6×6 incomplete diallel cross was designed using 12 rapeseed germplasms, and a total of 36 hybrids together with their parental lines were planted in 4 environments. Four yield-related traits and seed oil content (OC) were evaluated. Genetic distance (GD) was estimated with 359 simple sequence repeats (SSRs) markers. Heterosis levels, general combining ability (GCA) and specific combining ability (SCA) were evaluated. GD was found to have a significant correlation with better-parent heterosis (BPH) of thousand seed weight (TSW), SCA of seeds per silique (SS), TSW, and seed yield per plant (SY), while SCA showed a statistically significant correlation with heterosis levels of all traits at 1% significance level. Statistically significant correlations were also observed between GCA of maternal or paternal parents and heterosis levels of different traits except for SS. Interestingly, maternal (TSW, SS, and OC) and paternal (siliques per plant (SP) and SY) inheritance of traits was detected using contribution ratio of maternal and paternal GCA variance as well as correlations between GCA and heterosis levels. Phenotype and heterosis levels of all the traits except TSW of hybrids were significantly correlated with the average performance of parents. The correlations between SS and SP, SP and OC, and SY and OC were statistically significant in hybrids but not in parents. Potential applications of parental selection in hybrid breeding were discussed.  相似文献

从以富含纤维蛋白的血凝块为食物的棕尾别麻蝇幼虫肠道浸提液中分离纯化出3种具有溶纤活性的蛋白酶,分别命名为BPGFP1,BPGFP2和BPGFP3。其中,BPGFP1由两个分子量分别为32000和30000的亚基组成。BPGFP2和BPGFP3均为单体,分子量分别为40000和28000。这三种蛋白酶具有相似的底物特异性和抑制剂特性。三种蛋白酶均能降解溶纤活性蛋白酶的特异底物纤维蛋白,Chromzym,P,Chromzym UK和S-2288。三种酶还能够强烈降解类胰蛋白酶专一底物Bz-Phe-Val Arg NA,cBz Gly-Pro-Arg NA,Bz-Pro-Phe-Arg NA和Bz-Val-Gly-Arg NA.PMSF,STI,LBTI和SBBI能够对三种蛋白酶活怀有极强的抑制作用。三种溶纤活性蛋白酶均在pH9.0-10.0范围内表现出较高活性。  相似文献

蛇足石杉[Huperzia serrata(Thunb.)Trey.]是石杉科石杉属多年生小型草本植物,土生、附生或生于苔藓层中,喜阴凉潮湿环境。蛇足石杉含石杉碱甲,为目前治疗老年痴呆等疾病的特效药。对蛇足石杉的分类学、生物学特性(形态学、生态学、生理学与共生真菌研究)、繁殖与栽培、药用成分及含量测定等方面的研究新进展进行综述。并对其应用与保护前景进行展望。  相似文献

目的:探讨并比较血浆脑钠肽(BNP)对不同年龄组急性冠脉综合征(ACS)患者经皮冠脉介入(PCI)术后临床主要不良心脏事件(MACE)的预测价值。方法:回顾分析2007年12月至2010年3月因ACS在我院行PCI的患者,分为260岁和〈60岁两组,入院常规检验BNP,随访住院期间及出院后6个月内MACE(包括死亡、心源性休克、再发心绞痛、非致命性再次心肌梗死、非致命性心力衰竭、心律失常、靶血管血运重建、再次入院复合终点)的发生情况。结果:随访269例260岁患者,失访7例(2.6%),118例〈60岁患者,失访3例(2.5%),262例≥60岁患者的年龄与BNP水平显著正相关(r=0.368,P〈0.01),而115例〈60岁患者无显著相关性(r=-0.014,P〉0.05)。年龄≥60岁患者BNP水平与〈60岁患者比较,差异显著(P〈0.01),年龄260岁患者MACE与〈60岁患者比较,差异显著(P〈0.01),两组患者BNP水平与LVEF呈显著负相关(P均〈0.01)。多因素logistic回归分析显示:BNP可独立预测两组患者住院期间及出院后6个月内MACE(≥60岁OR=4.369P〈0.01;〈60岁OR=7.773P〈0.01)发生率。两组患者BNP水平对MACE预测的ROC曲线结果比较,差异无统计学意义(P〉0.05)。结论:不同年龄患者BNP水平及MACE发生率存在差异,但年龄并不影响BNP对ACS患者临床预后的预测价值。  相似文献

观察曲古抑菌素A(TSA)对骨肉瘤细胞株143B增殖及凋亡的作用,并探讨其机制。TSA与p38抑制剂(SB203580,3μmol/L)及JNK抑制剂(SP600125,0.5μmol/L)单独或同时处理143B细胞,分别以MTT、台盼蓝染色、流式细胞术和JC—1(测定线粒体跨膜电位)法检,TSA对143B细胞的增殖、存活、周期以及凋亡的影响。应用RT-PCR、Westernblot检测Bax、Bcl.2、p38/JNK表达。结果显示,TsA能够以时间和剂量依赖方式抑制143B细胞增殖,使细胞周期阻滞于GdG。与G2/M期,并能诱导143B细胞凋亡,引起线粒体膜电位降低,促凋亡蛋白Bax表达上调,抑凋亡蛋白Bcl.2表达下调,同时使p38/ⅢK活化增加。p38/JNK4检测剂则能逆转TSA对Bax/Bcl.2的上调及抑制作用。研究结果揭示,TSA可以时间剂量依赖方式抑制143BN胞增殖,阻滞细胞周期,诱导细胞凋亡;其诱导细胞凋亡的机制可能与活化MAPK通路中p38和JNK的活性从而激发线粒体凋亡通路有关。  相似文献

As a polyphenolic compound, resveratrol (Res) is widely present in a variety of plants. Previous studies have shown that Res can inhibit various tumors. However, its role in c remains largely unexplored. In the present study, we first demonstrated that Res inhibited cell viability and induced apoptosis of glioblastoma A172 cell. Further experiments showed that Res induced mitochondrial dysfunction and activated the activity of caspase-9. Functional studies have found that Res treatment is associated with an increase in the expression of Pak2. Interestingly, inhibition of Pak2 could further augment the proapoptotic effect of Res. Mechanistically, Pak2 inhibition induced reactive oxygen species overproduction, mitochondria-JNK pathway activation, and AMPK-YAP axis suppression. However, overexpression of YAP could abolish the anticancer effects of Res and Pak2 inhibition, suggesting a necessary role played by the AMPK-YAP pathway in regulating cancer-suppressive actions of Res and Pak2 inhibition. Altogether, our results indicated that Res in combination with Pak2 inhibition could further enhance the anticancer property of Res and this effect is mediated via the AMPK-YAP pathway.  相似文献

目的:观察复方扁咽宁对急性咽炎大鼠的治疗作用及其作用机理,为其临床应用提供依据。方法:采用氨水直接喷雾法复制大鼠急性咽炎模型,将其随机分为模型组、复方扁咽宁高、中、低剂量组,另取正常大鼠作为对照,每组10只,高、中、低剂量组剂量(g/kg)分别按28.8、14.4、7.2(生药量计)灌胃药物水提液,正常组与模型组给予等量蒸馏水,观察并记录各组大鼠的症状表现;于末次给药24 h后,腹主动脉采血,ELISA法检测血清中IL-1β及TNF-α的含量,采用HE染色法观察各组大鼠肺组织及肺支气管组织形态,透射电镜法观察气管纤毛形态。结果:模型组急性咽炎表现明显:咽部红肿、充血,气管及肺组织出现病理改变,血清中IL-1β及TNF-α的含量显著升高(与正常组比较,P<0.05),提示造模成功;与模型组相比,复方扁咽宁低、中、高剂量组急性咽炎的症状如咽部红肿、充血等现象得到明显缓解,肺组织及肺支气管组织形态得到明显的改善,血清中IL-1β及TNF-α的含量显著下降,中剂量组和高剂量组疗效更显著。结论:复方扁咽宁可加速急性咽炎的康复进程,改善支气管及肺的组织形态,其作用机制可能与抑制血清IL-1β和 TNF-α的释放等有关。  相似文献
