In an attempt to isolate and identify the target genes relevant to salt tolerance in a mangrove plant (Sesuvium portulacastrum L.), a subtracted cDNA library was constructed via suppressive subtractive hybridization (SSH), in which the poly(A)+RNA isolated from salt-tolerant S. portulacastrum leaves was used as a tester, whereas the driver was poly(A)+RNA, derived from salt-sensitive S. portulacastrum leaves. Screening of this subtracted cDNA library revealed five clones, of which the expression levels in the salt-tolerant plant were markedly higher than those observed in the salt-sensitive plant, indicating that these candidate clones may be involved in salt-tolerance pathways. Among the clones isolated, P66, P175, and P233 are novel because no significant similarity was obtained upon alignment with the GenBank database. Clone P89 demonstrated high homology with NADPH of Arabidopsis thaliana, whereas clone P152 was highly homologous with the gene encoding late embryogenesis abundant (LEA) protein of A. thaliana. The full-length gene of clone P152, with a predicated 344 amino acid residues, was shown to bear LEA-2 domains, a signature motif for proteins that have been enriched under salty and drought conditions. It is thus implied that clone P152 would be a salt-tolerance gene of S. portulacastrum. In addition, we have also developed a strategy for the extraction of total RNA from mangrove plants. 相似文献

In this work, severe acute respiratory syndrome associated coronavirus (SARS-CoV) genome BJ202 (AY864806) was completely sequenced. The genome was directly accessed from the stool sample of a patient in Beijing. Comparative genomics methods were used to analyze the sequence variations of 116 SARS-CoV genomes (including BJ202) available in the NCBI Gen-Bank. With the genome sequence of GZ02 as the reference, there were 41 polymorphic sites identified in BJ202 and a total of 278 polymorphic sites present in at least two of the 116 genomes. The distribution of the polymorphic sites was biased over the whole genome. Nearly half of the variations (50.4%, 140/278) clustered in the one third of the whole genome at the 3′ end (19.0 kb-29.7 kb). Regions encoding Orf10-11, Orf3/4, E, M and S protein had the highest mutation rates. A total of 15 PCR products (about 6.0 kb of the genome) including 11 fragments containing 12 known polymorphic sites and 4 fragments without identified polymorphic sites were cloned and sequenced. Results showed that 3 unique polymorphic sites of BJ202 (positions 13 804, 15 031 and 20 792) along with 3 other polymorphic sites (26 428, 26 477 and 27 243) all contained 2 kinds of nucleotides. It is interesting to find that position 18379 which has not been identified to be polymorphic in any of the other 115 published SARS-CoV genomes is actually a polymorphic site. The nucleotide composition of this site is A (8) to G (6). Among 116 SARS-CoV genomes, 18 types of deletions and 2 insertions were identified. Most of them were related to a 300 bp region (27 700-28 000) which encodes parts of the putative ORF9 and ORF10-11. A phylogenetic tree illustrating the divergence of whole BJ202 genome from 115 other completely sequenced SARS-CoVs was also constructed. BJ202 was phylogeneticly closer to BJ01 and LLJ-2004. 相似文献

Tissue culture techniques, medium composition, pH value and targeted tissues, agroinfection and co-culture conditions, selection process were optimized for efficient turfgrass transformation. A highly regenerable callus lines were produced in callus induction medium modified from N6 basal medium. Six-week-old calluses were cultured on Pre-regeneration medium I for 4 days and then subjected to Agrobacterium tumefaciens. After co-cultivation at 20±1 °C in a 16 h light/8 h darkness for 3 days, the calluses were cultured on non-selective Pre-regeneration medium II supplemented with 400 mg l−1 l-cysteine for 7 days. Plantlets were regenerated on the Regeneration medium without selection pressure. A selection pressure was given to the regenerated plantlets when they were rooted on the Plantlet rooting medium. Roots appeared within 8–12 days in putative transformed plantlets. Resistant plants obtained were phenotypically normal and fully fertile. Chemical and molecular analyses confirmed that foreign genes were successfully introduced into the genome of perennial ryegrass or tall fescue. Canopy exchanges of H^+ and N (NH4^+-N, NO3^--N) and other major ions were evaluated and quantified In twolayer canopies based on throughfall measurements in Shaoshan Forest during the period 2000-2002, central-south China, The collected annual rainfall, throughfall, and sub-throughfall were 1 401, 1 191, and 1 084 mm/year, respectively. Fifteen percent and 8% of rainfall (or 9% of throughfall) were intercepted by the top canopy and sub-canopy layers, respectively, The foliar leaching of base cations from the top canopy was significantly higher than that from the sub-canopy, and the latter accounted for 25% of the former. The uptake of H^+ and NH4^+ was significantly higher in the top canopy than in the sub-canopy, indicating that the canopy buffering capacity in the top canopy was stronger than the sub-canopy; Mg^2+ can be absorbed from water flux on the sub-canopy foliar surfaces to compensate for the Mg deficit in the forest soil during the growing season, 相似文献

We aimed to measure platelet function and its relationship with β2-GPI in prolonged isolated thrombocytopenia(PT) after allogeneic hematopoietic stem cell transplantation(allo-HSCT). Fifty-six patients with PT and 60 allo-HSCT recipients without PT(non-PT controls) were enrolled. Platelet aggregation and activation, β2-GPI and anti-β2-GPI antibody levels, vWF antigen,and vWF activity were analyzed. The effect of β2-GPI on platelet aggregation was also measured ex vivo. Results showed that ADP-induced platelet aggregation significantly increased(39%±7.5% vs. 23%±8.5%, P=0.032), and the platelet expression of both CD62 p(33.6%±11.6% vs. 8.5%±3.5%, P0.001) and PAC-1(42.4%±7.6% vs. 6.8%±2.2%, P0.001) was significantly higher in patients with PT than in those without PT. Significantly lower β2-GPI levels(164.2±12 μg m L–1 vs. 234.2±16 μg mL–1,P0.001), higher anti-β2-GPI IgG levels(1.78±0.46 U mL–1 vs. 0.94±0.39 U mL–1, P0.001), and increased vWF activity(133.06%±30.50% vs. 102.17%±25.90%, P0.001) were observed in patients with PT than in those without PT. Both ADPinduced platelet aggregation(n=116, r~2=-0.5042, P0.001) and vWF activity(n=116, r~2=-0.2872, P0.001) were negatively correlated with β2-GPI levels. In summary, our data suggested that platelet aggregation and activation were significantly higher in patients with PT than in those without PT, which might be associated with reduced β2-GPI levels. This paper investigates the spatial heterogeneity of landowners’ willingness to supply three bioenergy crops: switchgrass, Miscanthus, and willow, in the northeastern United States. Spatial heterogeneity might arise for several reasons. For example, landowners closer to bioenergy processing plants might be more likely to be willing to supply bioenergy crops, and landowners who are more willing to supply bioenergy crops may be spatially clustered because they share similar land attributes, demographics, experiences, and/or values. Using high‐resolution GIS data related to the location of pellet plants utilizing bioenergy crops and survey data related to landowners’ characteristics including spatial location, we estimate a spatial probit model to explain the variation in individual‐specific reservation prices (RPs)—the feedstock price at which landowners become willing to supply a bioenergy crop. We find that respondents’ RP is lower the closer they live to their nearest pellet plant and spatial dependency is only present for switchgrass supply. We also identify three economic hotspots (areas with high potential supply and low RPs) for each bioenergy crop. We believe that bioenergy supply chains could be developed around these hotspots. 相似文献

Soil multitrophic network complexity enhances the link between biodiversity and multifunctionality in agricultural systems 总被引:1,自引:0,他引:1
Belowground biodiversity supports multiple ecosystem functions and services that humans rely on. However, there is a dearth of studies exploring the determinants of the biodiversity–ecosystem function (BEF) relationships, particularly in intensely managed agricultural ecosystems. Here, we reported significant and positive relationships between soil biodiversity of multiple organism groups and multiple ecosystem functions in 228 agricultural fields, relating to crop yield, nutrient provisioning, element cycling, and pathogen control. The relationships were influenced by the types of organisms that soil phylotypes with larger sizes or at higher trophic levels, for example, invertebrates or protist predators, appeared to exhibit weaker or no BEF relationships when compared to those with smaller sizes or at lower trophic levels, for example, archaea, bacteria, fungi, and protist phototrophs. Particularly, we highlighted the role of soil network complexity, reflected by co-occurrence patterns among multitrophic-level organisms, in enhancing the link between soil biodiversity and ecosystem functions. Our results represent a significant advance in forecasting the impacts of belowground multitrophic organisms on ecosystem functions in agricultural systems, and suggest that soil multitrophic network complexity should be considered a key factor in enhancing ecosystem productivity and sustainability under land-use intensification. 相似文献
