Amylin is a pancreatic hormone involved in the regulation of glucose metabolism and homeostasis. Restoration of the post-prandial and basal levels of human amylin in diabetic individuals is a key in controlling glycemia, controlling glucagon, reducing the insulin dose and increasing satiety, among other physiologic functions. Human amylin has a high propensity to aggregate. We have addressed this issue by designing a liposomal human amylin formulation. Nanoparticles of multilamellar liposomes comprising human amylin were obtained with 53% encapsulation efficiency. The in vitro kinetic release assay shows a biphasic profile. The stabilization of the lipidic nanoparticle against freeze-drying was achieved by using mannitol as a cryoprotectant, as evidenced by morphological characterization. The effectiveness of the human amylin entrapped in lipidic nanoparticles was tested by the measurement of its pharmacological effect in vivo after subcutaneous administration in mice. Collectively these results demonstrate the compatibility of human amylin with the lipidic interface as an effective pharmaceutical delivery system.  相似文献

Here we aimed to investigate the association between poor quality of sleep, sleep debt during weekdays and the performance on an interval timing task, using the method of production of time and consisting on five sessions a day for nine days. The sleep of our volunteers was particularly restricted on weekdays. Moreover, later chronotype was associated with the need for compensation on weekends. Regarding the production of time, produced durations were consistently over-produced and there was an evident increase in the mean coefficient of variation (CV) from the wake-up morning to afternoon sessions, followed by a sharp decrease in the evening sessions. Importantly, CV was mainly determined by the interaction between MSFdiff, sleep duration on weekdays and age. In conclusion, our results indicate that when university students are under the effects of social jet lag and reduced sleep duration during weekdays, their performance in interval timing tasks may be compromised.  相似文献

Organoseleno-compounds have been investigated for its beneficial effects against methylmercury toxicity. In this way, diphenyl diselenide (PhSe)2 was demonstrated to decrease Hg accumulation in mice, protect against MeHg-induced mitochondrial dysfunction, and protect against the overall toxicity of this metal. In the present study we aimed to investigate if co-treatment with (PhSe)2 and MeHg could decrease accumulation of Hg in liver slices of rats. Rat liver slices were co-treated with (PhSe)2 (0.5; 5 µM) and/or MeHg (25 µM) for 30 min at 37 °C and Se and Hg levels were measured by inductively coupled plasma mass spectrometry (ICP-MS) in the slices homogenate, P1 fraction, mitochondria and incubation medium. Co-treatment with (PhSe)2 and MeHg did not significantly alter Se levels in any of the samples when compared with compounds alone. In addition, co-treatment with (PhSe)2 and MeHg did not decrease Hg levels in any of the samples tested, although, co-incubation significantly increased Hg levels in homogenate. We suggest here that (PhSe)2 could exert its previously demonstrated protective effects not by reducing MeHg levels, but forming a complex with MeHg avoiding it to bind to critical molecules in cell.  相似文献

Intensive land use of the Brazilian Atlantic Forest accelerated with the rise of sugar cane plantations in the northeastern part of Brazil. Consequently, many ecosystems were destroyed, including riparian forests. The number of studies of riparian restoration has increased but comparative studies on the belowground effects of common reforestation strategies are rare. Here, we compared soil microbial properties among four different land use types: native rainforest, sugar cane plantation, single species reforestation, and mixed species reforestation, each replicated at two spatially independent sites. Soil samples were taken in 2013 and 2014, that is 2 and 3 years after reforestation, respectively. In both years, land use types had a significant effect on basal respiration, microbial biomass, and specific respiration (whereas specific respiration was marginally affected in 2014). In 2013, basal respiration in sugar cane plantations was significantly lower (?65%) when compared to native forests. In 2014, basal respiration (+60%) and soil microbial biomass (+90%) were significantly higher in mixed species reforestation compared to sugar cane, whereas single species reforestation had comparable values as in sugar cane plantations. Our results indicate that soil microbial biomass and activity respond rapidly to land use change when mixed species reforestation is used. Thus, using mixed species reforestation may enhance the provisioning of ecosystem services already in the short term.  相似文献
