Embryonic cell number in miniature pigs inbred for specific SLA haplotypes (a, c, and d) was determined on Day 6 by nuclear staining and, on Days 9 and 11, by DNA analyses (first day of oestrus = Day 0). Pigs exhibiting first behavioural oestrus at 08:00 h were hand-mated to an SLA homozygous boar 12 and 24 h later. Numbers of embryos flushed from uteri at 08:00-10:00 h on Days 6, 9 and 11 were greater (P less than 0.05) for SLAd females than for SLAa or SLAc females, which did not differ (8.2 vs 6.8 and 6.2, respectively). Recovery rates (embryos recovered/CL number) were similar, averaging 75.8% for all three SLA haplotypes. Embryos from SLAd dams contained fewer blastomeres (23 cells) on Day 6 than did embryos from SLAa (89 cells) or SLAc (79 cells) females. The reduced cell numbers of SLAd vs SLAa or SLAc embryos continued to Day 9 (28 vs 107 and 67 ng DNA/embryo) and Day 11 (167 vs 674 and 586 ng DNA/embryo). Factors both intrinsic and extrinsic to the lung may cause inhomogeneity of alveolar pressures during deflation. Wilson et al. (J. Appl. Physiol. 59: 1924-1928, 1985) predicted that any such inhomogeneity would be limited by interdependence of regional expiratory flows. To test this hypothesis and to explore how the pleural pressure gradient might affect inhomogeneity of alveolar pressures, we deflated at submaximal flows excised canine lobes that first were suspended in air and then were immersed in foams that simulated the vertical gradient of pleural pressure. Interregional inhomogeneity of regional transpulmonary pressures was measured with use of an alveolar capsule technique. Flow-dependent inhomogeneity of alveolar pressures was present, with differences in alveolar pressure quickly relaxing to a constant limiting value at each flow. Foam immersion increased inhomogeneity at a given flow. We conclude that factors intrinsic to the lung cause significant inhomogeneity of alveolar pressures at submaximal expiratory flows and that this inhomogeneity is enhanced by the extrinsic gradient of pleural pressure. Summary Uroporphyrinogen III synthase [UROS; hydroxymethylbilane hydro-lyase (cyclizing), EC 4.2.1.75] is the fourth enzyme in the human heme biosynthetic pathway. The recent isolation of the cDNA encoding human UROS facilitated its chromosomal localization. Human UROS sequences were specifically amplified by the polymerase chain reaction (PCR) from genomic DNA of two independent panels of human-rodent somatic cell hybrids. There was 100% concordance for the presence of the human UROS PCR product and human chromosome 10. For each of the other chromosomes, there was 19%–53% discordance with human UROS. The chromosomal assignment was confirmed by Southern hybridization analysis of DNA from somatic cell hybrids with the full-length UROS cDNA.
