白音华矿区土壤重金属含量的空间异质性

为探讨开矿对白音华矿区土壤重金属空间分布的影响,本研究以内蒙古西乌珠穆沁旗白音华煤矿区周边土壤为对象,分析了距离矿区8 km内的重金属Cu、Cr、Pb和Mn含量的空间异质性.结果表明:土壤重金属Cu、Cr、Pb和Mn的平均含量分别为12.7、32.6、29.9和201.3 mg ? kg-1,其变异系数分别为26.8％...     

Increment of hFIX expression with endogenous intron 1 in Vitro

This paper probes into the feasibility of increasing expression level of hFIX gene with endogenous intron 1 sequence.hFIX minigene was obtained with middle sequence truncated intron 1 inserted into the relative site of hFIX cDNA,and plasmid vector pKG5i‘IX,retroviral vector G1NaCi‘IX were constructed.These vectors were transduced into target cells of PA317,C2C12,primary rabbit skin fibroblasts (RSF) and primary human skin fibroblasts (HSF).The expression level of mixed colonies are PA317/pKG5i‘IX,151 ng/10^6 cells/24h;PA317/G1NaCi‘IX,308 ng/10^6 cells/24 h;C2C12/G1NaCi‘IX,186 ng/10^6 cells/24 h;RSF/G1NaCi‘IX,1929 ng/10^6 cells/24 h;HSF/G1NaCi‘IX,1646 ng/10^6 cells/24 h.These results indicated that hFIX minigene with intron l is able to increase the expression level to about 3 times of that of hFIX cDNA.Meanwhile,in order to study the application of hFIX minigene in the retroviral-mediated gene transfer system and refrain from intron splicing during viral production,a retroviral vector G1NaCi‘IXR with reversely inserted hFIX minigene expression cassette was constructed.The expression level of reverse constructor in PA317 cells was 390 ng/10^6 cells/24 h with 79% of bioactivity.PCR detection of HT/G1NaCi‘IXR cells infected with PA317/G1NaCi‘IXR supernatant confirmed the existence of intron 1 sequence.These results suggested that expression vector with forward-inserted intronl-carrying hFIX expression cassette can be used in directed gene transfer,but when using the retroviral-mediated gene transfer system,reversely-inserted intronl-carrying hFIX expression cassette should be considered.     

Isolation of plasma membrane from protoplasts of Lolium multiflorum (ryegrass) endosperm cells.

Plasma membranes have been isolated from protoplasts of suspension-cultured ryegrass (Lolium multiflorum) endosperm cells. The protoplast membrane is coated before cell disruption with murine myeloma protein J539, a galactose-binding immunoglobulin A. The plasma membrane is labelled with 125I by using chemically or enzymically catalysed iodination techniques, or, more conveniently, by using 125I-labelled myeloma protein J539, which enables the membrane to be simultaneously coated and labelled. Protoplast lysis is effected by gentle mechanical means after swelling in hypo-osmotic medium. The plasma-membrane fraction is recovered at low centrifugal forces by fractionation of cell lysates on a discontinuous sucrose/sorbitol gradient. The plasma-membrane fraction is enriched 96-fold on a protein basis with respect to the specific radioactivity of 125I-labeled myeloma protein J539 in the homogenate. Electron microscopy showed long membrane profiles often associated with one another.     

Some virus and virus-like diseases of tobacco,tomato, papaya,and rubber tree in vietnam and cambodia

In Vietnam a green strain of tobacco mosaic virus was isolated having TIP 89°C (10 min) and causing systemic necrosis in tobacoo ‘Xanthi-nc’ and sometimes also inDatura stramonium. In symptomless tomato plants an elongated virus belonging apparently to the Carlavirus group (NL 630 nm) was found. In papaya trees showing severe symptoms of mosaic and/or ringspot elongated virus particles (NL 730 nm) were observed; this virus being apparently a member of the Potyvirus group, resembled as far as its symptoms in papaya are concerned, the papaya ringspot or the distortion ringspot. In Cambodia some young rubber trees showed malformed leaves (esp. edges and veins) with yellow discolorations along the veins. Such leaves contained elongated virus-like particles (rigid or slightly flexible) of various length (60 to 880 nm), so that their normal length (NL) could not be established precisely. Particles 120 to 150 nm long occurred very frequently.     

Exploiting plant somatic radiation hybrids for physical mapping of expressed sequence tags

Methods are described for the optimisation of the generation of radiation hybrids suitable for physical mapping of a plant (barley) genome. A combination of PCR-based technologies, involving the use of whole genome, mixed primer and hemi-nested primer amplifications, can greatly extend their utility for the physical mapping of expressed sequence tags (ESTs). Using panels of hybrids and ESTs, donor DNA retention and individual marker retention frequencies for the expressed portion of the barley genome in the hybrids were estimated.Electronic Supplementary Material Supplementary material is available in the online version of this article at Communicated by P. Fixing a Match or Two: Cricket,Public Confession and Moral Regeneration

‘In a moment of stupidity and weakness I allowed Satan and the world to dictate terms to me. The moment I took my eyes off Jesus my whole world turned dark.’ With these words, the former South African cricket captain, Hansie Cronje, publicly confessed that he had been a cheat, attempted to make peace with the United Cricket Board, located his fall from grace within a peculiarly fundamentalist Christian frame of guilt and atonement, and subtly connected himself and his corruption with a set of understandings about the simultaneous rigidity and vulnerability of the moral order that have grown over the past two centuries to inform, underpin and idealise Afrikaner collective notions of self. That moral order and its vulnerabilities, I will suggest here, have framed a collective identity and a collective sense of both victimhood and entitlement, within which any act becomes acceptable and possible if it can be deemed necessary.     

Multimodal signals in male European treefrog (Hyla arborea) and the influence of population isolation on signal expression

In nocturnal treefrogs, mate choice implies the use of acoustic and visual signals. Multimodality is suspected to have evolved for either information redundancy or information complementariness. It is essential to explore multimodality in a natural context to understand the selection pressures operating on the signals. In the present study, we investigated calling and coloration in relation to male biometry and condition in four populations of European treefrog (Hyla arborea) varying in size and genetic isolation. We compared the signal intensity between core and satellite populations to estimate the impact of genetic diversity on male secondary sexual traits. The results obtained show important regional variations in both traits, likely as a result of local adaptations. Call and coloration are weakly correlated within an individual, implying that these traits likely convey different information about the signaller's identity or quality, thus supporting the hypothesis of complementariness of multiple messages. By contrast to the experimental evidence, we find that call and coloration are not related to male condition (as estimated by the residual of mass over size), suggesting that the condition‐dependence of these traits may be mediated by complex mechanisms not accurately reflected by the chosen estimator. Finally, male call and colour phenotypes present no robust pattern of variation with isolation status, probably because of variation in local selective pressures and in history of population dynamics. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 633–647.     

苜蓿银纹夜蛾核多角体病毒iap1和iap2基因功能分析

苜蓿银纹夜蛾核多角体病毒（Autographa californica nucleopolyhedrovirus,AcMNPV）基因组含有3个细胞凋亡抑制基因,即p35,iap1和iap2.其中,p35作为一个有效的依赖于天冬氨酸的半胱氨酸蛋白酶（caspase）抑制因子,能够抑制多种因素诱发细胞凋亡,而iap1和iap2的功能仍未完全明晰,本研究对IAP1和IAP2的功能进行了详细分析.缺失了p35的AcMNPV仍可抑制棉铃虫核多角体病毒（Helicoverpa armigera single nucleocapsid NPV,HearNPV）诱导的BTI-Tn-5B1-4（Tn-Hi5）细胞凋亡并挽救HearNPV在Tn-Hi5细胞中复制及HearNPV出芽型病毒粒子的产生.进一步构建了瞬时表达质粒以及分别表达AcMNPV的p35,iap1和iap2基因的重组HearNPV,转染瞬时表达的IAP1和IAP2对HearNPV感染诱导的Tn-Hi5细胞凋亡有抑制效果,而重组病毒感染Tn-Hi5细胞也可抑制其凋亡并在其中复制,然而重组HearNPV表达的p35,iap1和iap2并未能挽救出芽型病毒粒子的产生.结果表明,AcMNPV的iap1和iap2基因表达产物作为细胞凋亡抑制因子是有功能的。     

Male preponderance in early diagnosed type 2 diabetes is associated with the ARE insertion/deletion polymorphism in the <Emphasis Type="Italic">PPP1R3A</Emphasis> locus

Background  

The ARE insertion/deletion polymorphism of PPP1R3A has been associated with variation in glycaemic parameters and prevalence of diabetes. We have investigated its role in age of diagnosis, body weight and glycaemic control in 1,950 individuals with type 2 diabetes in Tayside, Scotland, and compared the ARE2 allele frequencies with 1,014 local schoolchildren.
