LIVIN, a member of the inhibitor of apoptosis proteins (IAPs), is reported playing important roles in the development and progression of multiple human cancers. However, its underlined mechanisms in human renal cell carcinoma (RCC) are still needed to be clarified. In the present study, we reported that inhibition of miR-214 promoted the expression of LIVIN, then facilitated RCC cells growth and reduced the sensitivity of RCC cells to chemotherapeutic drugs. In constant, overexpression of miR-214 had contradictory effects. Further investigation showed that miR-214 was down-regulated in RCC because of abnormal methylation. In addition, DNA methyltransferase DNMT1, miR-214 and LIVIN are directly correlated in RCC patients. In conclusion, these results suggest that abnormal miR-214 methylation negatively regulates LIVIN, which may promote RCC cells growth and reduced the sensitivity of RCC cells to chemotherapeutic drugs. 相似文献

There is a concern of whether the structure and diversity of a microbial community can be effectively revealed by short-length
pyrosequencing reads. In this study, we performed a microbial community analysis on a sample from a high-efficiency denitrifying
quinoline-degrading bioreactor and compared the results generated by pyrosequencing with those generated by clone library
technology. By both technologies, 16S rRNA gene analysis indicated that the bacteria in the sample were closely related to,
for example, Proteobacteria, Actinobacteria, and Bacteroidetes. The sequences belonging to Rhodococcus were the most predominant, and Pseudomonas, Sphingomonas, Acidovorax, and Zoogloea were also abundant. Both methods revealed a similar overall bacterial community structure. However, the 622 pyrosequencing
reads of the hypervariable V3 region of the 16S rRNA gene revealed much higher bacterial diversity than the 130 sequences
from the full-length 16S rRNA gene clone library. The 92 operational taxonomic unit (OTUs) detected using pyrosequencing belonged
to 45 families, whereas the 37 OTUs found in the clone library belonged to 25 families. Most sequences obtained from the clone
library had equivalents in the pyrosequencing reads. However, 64 OTUs detected by pyrosequencing were not represented in the
clone library. Our results demonstrate that pyrosequencing of the V3 region of the 16S rRNA gene is not only a powerful tool
for discovering low-abundance bacterial populations but is also reliable for dissecting the bacterial community structure

Forty-eight 2-year-old Liaoning Cashmere goats (body weight = 38.0 ± 2.94 kg) were used to investigate the effects of dietary
iodine (I) and selenium (Se) supplementation on nutrient digestibility, serum thyroid hormones, and antioxidant status during
the cashmere telogen period to learn more about the effects of dietary I and Se on nutrition or health status of Cashmere
goats. The goats were equally divided into six groups of eight animals each that were treated with 0, 2, or 4 mg of supplemental
I/kg dry matter (DM) and 0 or 1 mg of supplemental Se/kg DM in a 2 × 3 factorial arrangement of treatments. The six treatments
were I0Se0, I2Se0, I4Se0, I0Se1, I2Se1, and I4Se1. The concentrations of I and Se in the basal diet were 0.67 and 0.09 mg/kg DM, respectively. The study started in March and
proceeded for 45 days. Supplemental I or Se alone had no effect on nutrient digestibility and nitrogen metabolism. However,
the interaction between I and Se was significant regarding the digestibility of acid detergent fiber (ADF; P < 0.05), and compared with group I4Se1, the digestibility of ADF was significantly increased in group I4Se0 (P < 0.05). Selenium supplementation did not affect serum triiodothyronine (T3) or thyroxine (T4) concentrations. However, the concentration of serum T4 but not that of T3 was significantly increased with I supplementation (P < 0.05). In addition, serum superoxide dismutase (SOD) activity was not affected (P > 0.05), but serum glutathione peroxidase (GSH-Px) activity was significantly decreased by I supplementation (P < 0.05). The antioxidant status was improved by Se supplementation, and the activities of SOD and GSH-Px were significantly

Background
Autophagy can either be protective and confer survival to stressed cells, or it can contribute to cell death. The antimitotic drug 2-ethyl-3-O-sulpamoyl-estra-1,3,5(10),15-tetraen-17-ol (ESE-15-ol) is an in silico-designed 17-β-estradiol analogue that induces both autophagy and apoptosis in cancer cells. The aim of the study was to determine the role of autophagy in ESE-15-ol-exposed human adenocarcinoma breast cancer cells; knowledge that will contribute to future clinical applications of this novel antimitotic compound. By inhibiting autophagy and determining the cytotoxic effects of ESE-15-ol-exposure, deductions could be made as to whether the process may confer resistance to the drug, or alternatively, contribute to the cell death process.Methods and results
Spectophometrical analysis via crystal violet staining was used to perform cytotoxicity studies. Morphology studies were done using microscopic techniques namely polarization-optical transmitted light differential interference light microscopy, fluorescent microscopy using monodansylcadaverine staining and transmission electron microscopy. Flow cytometry was used to quantify the autophagy inhibition and assess cell viability. Results obtained indicated that 3-methyladenine inhibited autophagy and increased cell survival in both MCF-7 and MDA-MB-231 cell lines.Conclusion
This in vitro study inferred that autophagy inhibition with 3-methyladenine does not confer increased effectiveness of ESE-15-ol in inducing cell death. Thus it may be concluded that the autophagic process induced by ESE-15-ol exposure in MCF-7 and MDA-MB-231 cells plays a more significant role in cell death than conferring survival.996.

Nonylphenol, the most abundant environmental pollutant with endocrine disrupting activity, is also toxic to plants and microorganisms,
but its actual impact in the field is unknown. In this study, diversity of culturable soil microfungal and plant communities
was assessed in a disused industrial estate, at three sites featuring different nonylphenol pollution. Although soil microfungal
assemblages varied widely among the sites, no significant correlation was found with point pollutant concentrations, thus
suggesting indirect effects of soil contamination on microfungal assemblages. The potential of indigenous fungi and plants
to remove nonylphenol was assessed in mesocosm experiments. Poplar plants and a fungal consortium consisting of the most abundant
strains in the nonylphenol-polluted soil samples were tested alone or in combination for their ability to reduce, under greenhouse
conditions, nonylphenol levels either in a sterile, artificially contaminated sand substrate, or in two non-sterile soils
from the original industrial area. Introduction of indigenous fungi consistently reduced nonylphenol levels in all substrates,
up to ca. 70% depletion, whereas introduction of the plant proved to be effective only with high initial pollutant levels.
In native non-sterile soil, nonylphenol depletion following fungal inoculation correlated with biostimulation of indigenous
fungi, suggesting positive interactions between introduced and resident fungi.

Phenylketonuria (PKU) is a common metabolic disorder among Chinese, with a prevalence of about 1 in 16,500 births. This frequency is very similar to that among Caucasians. Individual exons of the phenylalanine hydroxylase (PAH) gene with flanking introns were amplified by polymerase chain reaction and cloned into M13 for sequence analysis. An Arg111-to-Ter111 mutation has been identified in exon 3 of the PAH gene in a Chinese PKU patient. The mutation is in linkage disequilibrium with the mutant haplotype 4 alleles which are the most prevalent haplotype among the Orientals. The mutation accounts for about 10% of the Chinese PKU alleles and is absent from the Caucasians, demonstrating that independent mutational events have occurred in the PAH locus after racial divergence. 相似文献

Glutathione S-transferases (GST) are multifunctional detoxification enzymes that participate in a variety of cellular processes, including stress responses. In this study, we report the molecular characteristics of five GST genes (MaGSTU1, MaGSTU2, MaGSTU3, MaGSTF1 and MaGSTL1) cloned from banana (Musa acuminate L. AAA group, cv. Cavendish) using a RACE-PCR-based strategy. The predicted molecular masses of these GSTs range from 23.4 to 27.7 kDa and their pIs are acidic. At the amino acid level, they share high sequence similarity with GSTs in the banana DH-Pahang (AA group) genome. Phylogenetic analysis showed that the deduced amino acid sequences of MaGSTs also have high similarity to GSTs of other plant species. Expression analysis by semi-quantitative RT-PCR revealed that these genes are differentially expressed in various tissues. In addition, their expression is regulated by various stress conditions, including exposure to signaling molecules, cold, salinity, drought and Fusarium oxysporum f specialis(f. Sp) cubense Tropical Race 4 (Foc TR4) infection. The expression of the tau class MaGSTs (MaGSTU1, MaGSTU2 and MaGSTU3) mainly responded to cold, salinity and drought while MaGSTF1 and MaGSTL1 expressions were upregulated by signaling molecules. 于1988年春,对广西南宁、柳州、桂林市区的园林植物作了全面调查。结果如下:1)三市区园林植物较丰富,其中南宁市351种隶属245属108科;柳州76科152属198种;桂林76科144属172种。2)园林植物的性状组成中,乔木占50—60%;灌木占20—30%,草本和藤本分别占15%和5%。3)园林植物的主要科是桑科、大戟科、棕榈科、木兰科、桃金娘科、苏木科、蔷薇科、樟科等;优势种是扁桃、人面子、柠檬桉、羊蹄甲、芒果、阴香、樟树、榕树、桂花、银桦、大红花、夹竹桃、九里香、正木等。4)园林植物的区系成分以热带—亚热带为主。5)园林植物的分布以公园最为丰富,其种数占总种数的67—87%,街道和阳台分别为43%和30%。6)不同市区园林植物的优势科、优势种、区系成分、生态外貌有明显差异,反映出各自的特点。与当地自然景观相一致。最后,在调查分析的基础上,对城市园林绿化的发展问题讨论。 相似文献
