Objectives
The frequency of common oncogenic mutations and TP53 was determined in Asian oral squamous cell carcinoma (OSCC).Materials and Methods
The OncoCarta™ panel v1.0 assay was used to characterize oncogenic mutations. In addition, exons 4-11 of the TP53 gene were sequenced. Statistical analyses were conducted to identify associations between mutations and selected clinico-pathological characteristics and risk habits.Results
Oncogenic mutations were detected in PIK3CA (5.7%) and HRAS (2.4%). Mutations in TP53 were observed in 27.7% (31/112) of the OSCC specimens. Oncogenic mutations were found more frequently in non-smokers (p = 0.049) and TP53 truncating mutations were more common in patients with no risk habits (p = 0.019). Patients with mutations had worse overall survival compared to those with absence of mutations; and patients who harbored DNA binding domain (DBD) and L2/L3/LSH mutations showed a worse survival probability compared to those patients with wild type TP53. The majority of the oncogenic and TP53 mutations were G:C > A:T and A:T > G:C base transitions, regardless of the different risk habits.Conclusion
Hotspot oncogenic mutations which are frequently present in common solid tumors are exceedingly rare in OSCC. Despite differences in risk habit exposure, the mutation frequency of PIK3CA and HRAS in Asian OSCC were similar to that reported in OSCC among Caucasians, whereas TP53 mutations rates were significantly lower. The lack of actionable hotspot mutations argue strongly for the need to comprehensively characterize gene mutations associated with OSCC for the development of new diagnostic and therapeutic tools. 相似文献32.

Although cervical cancer is preventable with early detection, it remains the second most common malignancy among women. An understanding of how proteins change in their expression during a particular diseased state such as cervical cancer will contribute to an understanding of how the disease develops and progresses. Potentially, it may also lead to the ability to predict the occurrence of the disease. With this in mind, we aimed to identify differentially expressed proteins in the plasma of cervical cancer patients. Plasma from control, cervical intraepithelial neoplasia (CIN) grade 3 and squamous cell carcinoma (SCC) stage IV subjects was resolved by two-dimensional gel electrophoresis and the resulting proteome profiles compared. Differentially expressed protein spots were then identified by mass spectrometry. Eighteen proteins were found to be differentially expressed in the plasma of CIN 3 and SCC stage IV samples when compared with that of controls. Competitive ELISA further validated the expression of cytokeratin 19 and tetranectin. Functional analyses of these differentially expressed proteins will provide further insight into their potential role(s) in cervical cancer-specific monitoring and therapeutics. 相似文献

Microbial transformation of sulfate minerals plays an important role in controlling the behavior of heavy metals in mining areas. Here, the anaerobic reduction of Cr (VI)-loaded schwertmannite by Shewanella oneidensis MR-1 (S. oneidensis MR-1) was investigated. The release of ferrous iron (Fe(II)) to the solution demonstrated the microbial reduction of structural Fe(III) from the schwertmannite to Fe(II). The concentration of Cr in solution decreased in all treatments, indicating that no Cr was released to the solution during this bio-reduction process of schwertmannite. The incorporation of chromate into the mineral structure of schwertmannite increased the microbial stability of the mineral, retarding the formation of secondary phases during bio-reduction process. Analysis of the XRD, SEM and fourier transform infrared spectroscopy (FT-IR) results further showed that goethite formed after 3 or 7 days with a lower content (0.22% or 0.37%) of Cr in schwertmannite, while no secondary mineral was observed with a higher concentration of Cr (0.6 wt%) incorporated in schwertmannite until 22 days. These results imply that microbial reduction of Cr(VI)-loaded schwertmannite does not lead to the release of Cr to the solution, and the microbial stability of schwertmannite will be increased by the incorporation of chromate. 相似文献

Studies on some plant species have shown that increasing the growth temperature gradually or pretreating with high temperature can lead to obvious photosynthetic acclimation to high temperature. To test whether this acclimation arises from heat adaptation of ribulose 1,5‐bisphosphate carboxylase/oxygenase (Rubisco, EC 4.1.1.39) activation mediated by Rubisco activase (RCA), gene expression of RCA large isoform (RCAL) and RCA small isoform (RCAS) in rice was determined using a 4‐day heat stress treatment [40/30°C (day/night)] followed by a 3‐day recovery under control conditions [30/22°C (day/night)]. The heat stress significantly induced the expression of RCAL as determined by both mRNA and protein levels. Correlative analysis indicated that RCAS protein content was extremely significantly related to Rubisco initial activity and net photosynthetic rate (Pn) under both heat stress and normal conditions. Immunoblot analysis of the Rubisco–RCA complex revealed that the ratio of RCAL to Rubisco increased markedly in heat‐acclimated rice leaves. Furthermore, transgenic rice plants expressing enhanced amounts of RCAL exhibited higher thermotolerance in Pn and Rubisco initial activity and grew better at high temperature than wild‐type (WT) plants and transgenic rice plants expressing enhanced amounts of RCAS. Under normal conditions, the transgenic rice plants expressing enhanced amounts of RCAS showed higher Pn and produced more biomass than transgenic rice plants expressing enhanced amounts of RCAL and wild‐type plants. Together, these suggest that the heat‐induced RCAL may play an important role in photosynthetic acclimation to moderate heat stress in vivo, while RCAS plays a major role in maintaining Rubisco initial activity under normal conditions. 相似文献

This study was conducted to examine the effects of clipping and nutrient addition on plant traits of a dominant perennial forb species, Heteropappus altaicus (Willd.) Novopokr. (Compositae), at both the individual and population levels in a temperate steppe in northern China. A nested experimental design was used with clipping as the main factor and nutrient, including nitrogen (N), phosphorus (P) and both, addition as the second factor. The main effect of clipping reduced plant height, aboveground biomass (AGB) per plant, and pollen production per floret by 15.8, 34.3, 28.0% (all p < 0.05), respectively, but enhanced reproductive allocation and population density by 8 and 28.2% (both p < 0.05), respectively, suggesting contrary effects of clipping on H. altaicus traits at the individual and population levels. N addition significantly stimulated plant height, AGB per plant, reproductive allocation, pollen diameter, and pistil length, but decreased population density. The main effects of P addition also stimulated the plant traits at individual level, but did not change population traits. The significant interactions of clipping and nitrogen addition were observed on AGB per plant, pollen production, and population density. The differential responses of H. altaicus at the individual and population levels to clipping and nutrient addition indicate that the future dynamics of H. altaicus in the temperate steppe are uncertain and need long-term research to demonstrate. 相似文献

We examined the endothelial gap junctions in diabetic hyperlipidemic mice. Male apolipoprotein E (apoE)-deficient mice were made diabetic by streptozotocin. Three weeks later, the animals were treated with simvastatin for 2 weeks. The expression of aortic gap junctions in the non-diabetic (n=10), untreated diabetic (n=10), and simvastatin-treated diabetic animals (n=6) was analyzed. There was a >4-fold increase in serum cholesterol level and >50% increase in plaque areas in the diabetic mice, regardless of simvastatin treatment. Western blotting of aortae showed reduced expression of connexin37 (Cx37) and Cx40 in the diabetic mice, which were further decreased in the simvastatin-treated diabetic mice. Immunoconfocal microscopy showed that endothelial gap junctions made of Cx37 and Cx40 were both reduced in the untreated diabetic mice compared with the non-diabetic mice (decrease: Cx37, 41%; Cx40, 42%; both p<0.01). The reduction was greater in the simvastatin-treated mice (decrease in treated diabetic vs non-diabetic: Cx37, 61%; Cx40, 79%; both p<0.01; decrease in treated diabetic vs untreated diabetic: Cx37, 34%; Cx40, 63%; both p<0.01). Cx37 and Cx40 were decreased in the endothelium of plaque surface. Cx43 appeared in the medial layer and inner layer of the intima. All three connexins were rarely expressed in monocytes/macrophages inside the plaques. In conclusion, in apoE-deficient mice, streptozotocin-induced diabetes is associated with downregulation of endothelial Cx37 and Cx40 gap junctions. Subtropical East Asia harbours a large plant diversity that is often attributed to allopatric speciation in this topographically complex region characterized by a relative climate stability. Here, we use observations of Platycarya, a widespread subtropical Asian tree genus, to explore the consequences of past climate stability on species’ evolutionary history in subtropical China. This genus has a controversial taxonomy: while it is now prevailingly treated as monotypic, two species have been originally described, Platycarya strobilacea and P. longipes. Previous information from species distribution models, fossil pollen data and genetic data based on chloroplast DNA (cpDNA) were integrated with newly obtained genetic data from the two putative species. We used both cpDNA (psbA-trnH and trnL-F intergenic spacers, including a partial trnL gene sequence) and nuclear markers. The latter included sequences of the internal transcribed spacer region (ITS1–5.8S–ITS2) of the nuclear ribosomal DNA and random genomic single nucleotide polymorphisms. Using these nuclear genetic markers, we found interspecific genetic divergence fitting with the ‘two species’ scenario and geographically structured intraspecific variation. Using cpDNA markers, we also found geographically structured intraspecific variation. Despite deep inter- and intraspecific genetic divergence, we detected genetic admixture in southwest China. Overall, our findings of genetic divergence within Platycarya support the hypothesis of allopatric speciation. However, episodes of population interconnection were identified, at least in southwest China, suggesting that the genus has had a dynamic population history. 相似文献
