Several studies have found genetic variation in plant resistance to herbivory. One of the explanations suggested for the observed intermediate levels of resistance are the costs of resistance, i.e., negative genetic correlations between resistance and other fitness components that may constrain the evolution of resistance. We studied the cost of herbivore resistance by investigating the genetic correlations between resistance traits and plant growth traits, and between resistance to insect and mammalian herbivores in cloned saplings of silver birch, Betula pendula. We used the performance of a geometrid moth, Epirrita autumnata, as an indicator of insect resistance. The numbers of resin droplets at the base and at the tip of the saplings correlate with mammalian resistance, and were thus used here as indicators of vole and hare resistance, respectively. We have previously observed genetic variation in these resistance traits. Further, we examined the correlations between several groups of secondary chemicals and plant growth traits. Finally, to reveal the effect of environmental factors on the trade-offs mentioned above, we investigated the correlations in saplings that were grown at two nutrient levels. We found significant negative correlations between indices of constitutive insect resistance and relative height growth in non-fertilized saplings, indicating cost of constitutive insect resistance. The two groups of secondary chemicals that have been shown to correlate strongly with constitutive insect resistance, i.e., condensed tannins and flavonol glycosides (especially myricetin glycosides), had different genetic correlations with plant traits; the concentration of condensed tannins did not correlate negatively with any of the plant traits, whereas the concentration of flavonol glycosides correlated negatively with plant height. Insect and mammalian resistance did not correlate negatively, indicating no ecological trade-offs.  相似文献

This study examined the overall impact of simulated herbivory on tillering and reproductive performance of an annual ryegrass,Lolium remotum. The interaction between herbivore damage and intraspecific competition and the effect of the timing of damage were also studied. The experimental plants were sown at two densities and were randomly assigned to eight different damage treatments consisting of artificial leaf area removal by clipping with scissors or removal of one-third of the ripening seeds. The treatments were executed at two flowering stages. The pattern of tiller development differed significantly among treatments and between densities. At the lower density, earlier treatments delayed tiller development more than the same treatments executed later. At the higher density, all treatments delayed tiller development. The density effect was significant for all reproductive traits measured. The reproductive output of plants grown at the higher density was lower and the negative treatment effects were stronger than at the lower density. The treatment effect was significant for seed dry weight per plant and individual seed weight but not for number of seeds per plant. There were no statistically significant interaction effects between the damage treatments and density, suggesting that the plants responded to the damage similarly, irrespective of the density. The plants did not totally compensate for losses due to damage at either density, even though they slightly increased their resource allocation to sexual reproduction at the higher density.  相似文献

The cDNA encoding human preprorenin has been introduced into the adenovirus-transformed human kidney cell line 293. The recombinant 293 cells expressed and secreted prorenin; trypsin was used to activate the secreted prorenin to renin in vitro. The recombinant protein was purified to homogeneity by a single affinity chromatographic step. Using synthetic tetradecapeptide, the Km was 57.1 +/- 9.3 microM and the kcat was (7.48 +/- 1.57) x 10(3)/hr. Activation with trypsin resulted in a secondary cleavage between Arg53 and Leu54 generating a two chain form held together via a disulfide between Cys51 and Cys58. This secondary cleavage did not affect enzyme activity as determined by the ability of renin to degrade a synthetic tetradecapeptide substrate. Our paper demonstrates the potential for producing large quantities of renin from human kidney cells and also suggests that the use of trypsin, which has been widely used to convert prorenin to renin in vitro, causes a secondary cleavage in the renin peptide chain.  相似文献

Evidence is presented that incubation of intact microsomal vesicles with ATP and the catalytic subunit of cAMP-dependent protein kinase does not stimulate glucose-6-phosphatase. Our analyses show that commercial preparations of ATP, phosphoenol pyruvate, pyruvate kinase and protein kinase contain free phosphate that complicates interpretation of experimental data obtained using colorimetric assays of enzymic activity. The marked inhibition of enzymic activity by dithiothreitol, present in reconstituted preparations of protein kinase, also is a confounding factor. We recommend the use of glucose-6-phosphatase assays employing32P-labelled substrate in future studies of this mechanism.  相似文献

Basophils, which are normally confined to the circulation, can migrate to sites of allergic inflammation. Using the specific mAb, BB1, we detected basophil infiltration of the gastric mucosa of Helicobacter pylori-infected patients affected by moderate and severe gastritis. Basophils were not found in H. pylori-free individuals or in subjects with mild gastritis. The H. pylori-derived peptide, Hp(2-20), was a potent basophil chemoattractant in vitro, whereas the control peptide, Hp1, was ineffective. Basophils from peripheral blood of healthy volunteers expressed mRNA for the formyl peptide receptors, N-formyl-peptide receptor (FPR), FPR-like (FPRL)1, and FPRL2. Preincubation of basophils with FMLP or Hp(2-20) caused complete desensitization to a subsequent challenge with homologous stimulus. Incubation of basophils with a low concentration of FMLP, which binds with high affinity to FPR, but not to FPRL1 or FPRL2, did not affect the chemotactic response to Hp(2-20). In contrast, a high concentration of FMLP, which binds to FPRL1 and FPRL2, reduced the chemotactic response to Hp(2-20). The FPR antagonist, cyclosporin H, prevented chemotaxis induced by FMLP, but not by Hp(2-20). Hp(2-20) could be responsible, at least in part, for basophil infiltration of the gastric mucosa of H. pylori-infected patients presumably through the interaction with FPRL1 and FPRL2.  相似文献

Checkpoint pathways inhibit cyclin-dependent kinases (Cdks) to arrest cell cycles when DNA is damaged or unreplicated. Early embryonic cell cycles of Xenopus laevis lack these checkpoints. Completion of 12 divisions marks the midblastula transition (MBT), when the cell cycle lengthens, acquiring gap phases and checkpoints of a somatic cell cycle. Although Xenopus embryos lack checkpoints prior to the MBT, checkpoints are observed in cell-free egg extracts supplemented with sperm nuclei. These checkpoints depend upon the Xenopus Chk1 (XChk1)-signaling pathway. To understand why Xenopus embryos lack checkpoints, xchk1 was cloned, and its expression was examined and manipulated in Xenopus embryos. Although XChk1 mRNA is degraded at the MBT, XChk1 protein persists throughout development, including pre-MBT cell cycles that lack checkpoints. However, when DNA replication is blocked, XChk1 is activated only after stage 7, two cell cycles prior to the MBT. Likewise, DNA damage activates XChk1 only after the MBT. Furthermore, overexpression of XChk1 in Xenopus embryos creates a checkpoint in which cell division arrests, and both Cdc2 and Cdk2 are phosphorylated on tyrosine 15 and inhibited in catalytic activity. These data indicate that XChk1 signaling is intact but blocked upstream of XChk1 until the MBT.  相似文献
