Angiogenesis is a multistep complex phenomenon critical for several inflammatory and neoplastic disorders. Basophils, normally confined to peripheral blood, can infiltrate the sites of chronic inflammation. In an attempt to obtain insights into the mechanism(s) underlying human basophil chemotaxis and its role in inflammation, we have characterized the expression and function of vascular endothelial growth factors (VEGFs) and their receptors in these cells. Basophils express mRNA for three isoforms of VEGF-A (121, 165, and 189) and two isoforms of VEGF-B (167 and 186). Peripheral blood and basophils in nasal polyps contain VEGF-A localized in secretory granules. The concentration of VEGF-A in basophils was 144.4 +/- 10.8 pg/10(6) cells. Immunologic activation of basophils induced the release of VEGF-A. VEGF-A (10-500 ng/ml) induced basophil chemotaxis. Supernatants of activated basophils induced an angiogenic response in the chick embryo chorioallantoic membrane that was inhibited by an anti-VEGF-A Ab. The tyrosine kinase VEGFR-2 (VEGFR-2/KDR) mRNA was expressed in basophils. These cells also expressed mRNA for the soluble form of VEGFR-1 and neuropilin (NRP)1 and NRP2. Flow cytometric analysis indicated that basophils express epitopes recognized by mAbs against the extracellular domains of VEGFR-2, NRP1, and NRP2. Our data suggest that basophils could play a role in angiogenesis and inflammation through the expression of several forms of VEGF and their receptors.  相似文献

Abstract

The public debate about GM crops that took place in Britain in 2002–03 was an unprecedented experiment for the UK in public engagement. The Government, which sponsored the exercise, made a pledge: to take into account lay perspectives, as well as the views of its expert advisers, when making a decision about the possible commercialisation of this controversial technology. As officially accredited but independent evaluators of the debate, we were given unique behind-the-scenes access to the organisation and execution of the debate process. In this paper we report on our findings, which reveal a debate process that, whilst innovative, suffered from a number of important flaws: in terms of both design and implementation. These shortcomings in part reflect constraints on time and budget, and the context of political pressure in which the debate took place. However, they also demonstrated a lack of design knowledge and availability of appropriate expertise: matters with an important bearing on future plans for such initiatives.  相似文献

The sphingolipid ceramide elicits several stress responses, however, organisms survive despite increased ceramide but how they do so is poorly understood. We demonstrate here that the AKT/FOXO pathway regulates survival in increased ceramide environment by metabolic adaptation involving changes in glycolysis and lipolysis through novel downstream targets. We show that ceramide kinase mutants accumulate ceramide and this leads to reduction in energy levels due to compromised oxidative phosphorylation. Mutants show increased activation of Akt and a consequent decrease in FOXO levels. These changes lead to enhanced glycolysis by upregulating the activity of phosphoglyceromutase, enolase, pyruvate kinase, and lactate dehydrogenase to provide energy. A second major consequence of AKT/FOXO reprogramming in the mutants is the increased mobilization of lipid from the gut through novel lipase targets, CG8093 and CG6277 for energy contribution. Ubiquitous reduction of these targets by knockdown experiments results in semi or total lethality of the mutants, demonstrating the importance of activating them. The efficiency of these adaptive mechanisms decreases with age and leads to reduction in adult life span of the mutants. In particular, mutants develop cardiac dysfunction with age, likely reflecting the high energy requirement of a well-functioning heart. The lipases also regulate physiological triacylglycerol homeostasis and are important for energy metabolism since midgut specific reduction of them in wild type flies results in increased sensitivity to starvation and accumulation of triglycerides leading to cardiac defects. The central findings of increased AKT activation, decreased FOXO level and activation of phosphoglyceromutase and pyruvate kinase are also observed in mice heterozygous for ceramide transfer protein suggesting a conserved role of this pathway in mammals. These data reveal novel glycolytic and non-autonomous lipolytic pathways in response to increased ceramide for sustenance of high energy demanding organ functions like the heart.  相似文献

A recent article on sectarianism in Scotland in Ethnic and Racial Studies by Bruce et al. sought to undermine the conclusions of a previous paper of ours, also published in this journal. Bruce et al. contend that sectarianism is a myth, while we have provided new qualitative evidence for personal experience of anti-Catholic discrimination in employment, which clearly contradicts their thesis. To contextualize these papers, we have summarized some of the key points of the evidence, and of the increasing concern about sectarianism in Scotland. In an effort to ridicule this concern, Bruce has attacked us as he has previously attacked a number of others. He and his co-authors accuse us of relying on respondents’ fallible judgements about the actual and appropriate proportions of Catholics in workplaces; then they try to interpret some of our respondents’ statements and their own quantitative evidence as supporting their myth hypothesis. We document here the process of misrepresentation by which they have sought to support their allegations, and we re-affirm the actual argument which led to our conclusions. We suggest a more obvious alternative interpretation of their own quantitative data, while raising misgivings about their use of 2001 census data. Questions about sectarian discrimination in Glasgow in the period 1950–2000 can no longer be baulked.  相似文献

Gauging the potential impacts of environmental change on the geographic distributions of species is a central area of modern biogeographic analysis, often involving complex models of species–environment interactions. The geographic distribution of fossil species can also provide a framework to test the impact of environmental change on biogeography and ecological niches of species, yet few paleontological analyses have attacked this question in deep time. Herein we present a quantitative biogeographic analysis to examine the stability of ecological niches and geographic ranges of rhynchonelliform brachiopods during an interval of sea level change preserved in Upper Ordovician strata of the Cincinnati Arch.The intensive sampling, excellent preservation, and numerous prior paleoecological and sedimentological analyses within the tri-state region of Kentucky, Indiana, and Ohio provide a robust framework for detailed paleobiogeographic study. Quantitative biogeographic modeling methods incorporating GIS (Geographic Information Systems) are utilized in order to spatially analyze the geographic ranges of brachiopod species of the Corryville and Mt. Auburn Formations of the C3 (uppermost Maysvillian) depositional sequence.This study employs the ecological niche modeling program GARP (Genetic Algorithm using Rule-set Prediction) to predict the geographic distribution of eight brachiopod species during three time slices within the C3 sequence. This method estimates a species’ geographic range by modeling the ecological niche of the species based on a set of known species occurrence data coupled with environmental data inferred from sedimentologic proxies. Once environmental tolerances for a species are modeled; the species is predicted to occur wherever its preferred set of environmental conditions occurs within the study region.Distributional patterns were reconstructed for three time slices during the C3 sequence. Recovered range predictions were quantitatively analyzed for evidence of temporal range changes. Results indicate that average species range within the study area decreased and species tracked their preferred niche with high fidelity during the transition from the early to middle portions of the C3 depositional sequence, an interval of rapid relative sea level change. However, during the transition from the middle to late portions of the sequence, gradual shallowing within the basin and development of discontinuous habitat patches correlates with niche evolution of five of the eight species modeled. The average area a species occupied within the basin increased during this interval, but there is a mixed response including both increases and decreases in range size within the study group. In general, the species that exhibit niche evolution increased their geographic range size while those that continue to track their niche with high fidelity experience a decrease in geographic range size. During the latter half of the C3 sequence, previously continuous habitats become fragmented, thereby isolating individual populations and providing a mechanism for niche evolution. The rate of sea level change and the corresponding fragmentation of previously continuous habitats into isolated patches appear to be the primary controls on both mean geographic range size and relative degree of niche evolution.  相似文献

GABA is synthesized from glutamate by glutamate decarboxylase (GAD), which exists in two isoforms, that is, GAD65 and GAD67. In line with GAD65 being located in the GABAergic synapse, several studies have demonstrated that this isoform is important during sustained synaptic transmission. In contrast, the functional significance of GAD65 in the maintenance of GABA destined for extrasynaptic tonic inhibition is less well studied. Using GAD65-/- and wild type GAD65+/+ mice, this was examined employing the cortical wedge preparation, a model suitable for investigating extrasynaptic GABA(A) receptor activity. An impaired tonic inhibition in GAD65-/- mice was revealed demonstrating a significant role of GAD65 in the synthesis of GABA acting extrasynaptically. The correlation between an altered tonic inhibition and metabolic events as well as the functional and metabolic role of GABA synthesized by GAD65 was further investigated in vivo. Tonic inhibition and the demand for biosynthesis of GABA were augmented by injection of kainate into GAD65-/- and GAD65+/+ mice. Moreover, [1-(13) C]glucose and [1,2-(13) C]acetate were administered to study neuronal and astrocytic metabolism concomitantly. Subsequently, cortical and hippocampal extracts were analyzed by NMR spectroscopy and mass spectrometry, respectively. Although seizure activity was induced by kainate, neuronal hypometabolism was observed in GAD65+/+ mice. In contrast, kainate evoked hypermetabolism in GAD65-/- mice exhibiting deficiencies in tonic inhibition. These findings underline the importance of GAD65 for synthesis of GABA destined for extrasynaptic tonic inhibition, regulating epileptiform activity.  相似文献

We examined diet-dependent plasticity in head shape in larvae of the eastern long-toed salamander, Ambystoma macrodactylum columbianum. Larvae in some populations of this species exhibit trophic polymorphism, with some individuals possessing exaggerated trophic features characteristic of a cannibalistic morphology in larval Ambystoma; e.g. a disproportionately broad head and hypertrophied vomerine teeth. We hypothesized that 1) head shape variation results from feeding upon different types of prey and that 2) cannibal morphs are induced by consumption of conspecifics. To induce variation, we fed three groups of larvae different diets: 1) brine shrimp nauplii only; 2) nauplii plus anuran tadpoles; 3) nauplii, tadpoles and conspecific larval salamanders. Comparisons of size (mass)-adjusted means revealed that this manipulation of diet induced significant variation in six measures of head shape, but not in the area of the vomerine tooth patch. For five of the six head traits, larvae that ate tadpoles and brine shrimp nauplii developed significantly broader, longer and deeper heads than did larvae that only ate brine shrimp nauplii. The ingestion of conspecifics, in addition to nauplii and tadpoles, significantly altered two head traits (interocular-width and head depth), compared to larvae only fed nauplii and tadpoles. Canonical discriminant function analysis detected two statistically reliable canonical variables: head depth was most highly associated with the first canonical variable, whereas three measures of head width (at the jaws, gills and eyes) and interocular width were most highly associated with the second canonical variable. Despite this diet-enhanced morphological variation, there was no indication that any of the three types of diet (including conspecific prey) induced the exaggerated trophic features of the cannibal morph in this species. These results illustrate that ingestion of different types of prey contributes to plasticity in head shape, but that some other proximate cue(s), either alone or in combination with diet variation, is essential to induce the extremes of trophic polymorphism in this species.  相似文献

Numerous studies have demonstrated the association of childhood socioeconomic position and adult height. Many have suggested the use of adult height as a marker of overall childhood well-being. However, few studies have examined the relationship between child/adolescent socioeconomic position and adult height in a racially/ethnically diverse cohort. Using the National Longitudinal Study of Adolescent Health, we examined the association of child/adolescent SEP (maternal education and maternal report of household income) and measured adult height in a diverse cohort of US adolescents/young adults. We found a positive gradient effect of maternal education on height in the overall population and in White and Mixed race males and females; no such gradient existed in Hispanic, Asian, or Black males or females. Only in Mixed race females was household income positively associated with height. These findings emphasize the need to recognize differential effects of socioeconomic status on height in different racial/ethnic and gender subpopulations.  相似文献
