A mathematical model based on a dynamic metabolic flux balance (DMFB) is developed for a process of fed‐batch fermentation of Bordetella pertussis. The model is based on the maximization of growth rate at each time interval subject to stoichiometric constraints. The model is calibrated and verified with experimental data obtained in two different bioreactor experimental systems. It was found that the model calibration was mostly sensitive to the consumption or production rates of tyrosine and, for high supplementation rates, to the consumption rate of glutamate. Following this calibration the model correctly predicts biomass and by‐products concentrations for different supplementation rates. Comparisons of model predictions to oxygen uptake and carbon emission rates measurements indicate that the TCA cycle is fully functional. © 2013 American Institute of Chemical Engineers Biotechnol. There is considerable evidence that host genetic factors are important in determining susceptibility to mycobacterial infections.
More recently, functional genetic mutations affecting IL-10 receptor 1 (IL-10R1) were described. In this study, we investigated
the relationship of IL-10R1 S138G loss-of-function polymorphism (A536G: rs3135932) with susceptibility to active tuberculosis
(TB) in Tunisian patients. A total of 168 patients with pulmonary TB, 55 with extrapulmonary TB, and 150 control subjects
were studied. Genomic DNA samples were extracted from leukocytes and used to investigate S138G polymorphism in IL-10R1 gene
by multiplex allele-specific polymerase chain reaction. Associations between G allele [odds ratio OR = 5.01; 95% confidence
intervals CI = 2.58–9.77; P = 10−7], GG genotypes [OR=9.06; 95% CI (1.58–67.33); correcting P-values using the Bonferroni method for multiple tests Pc=0.015] and AG genotype [OR=3.75; 95% CI (1.62–8.7); Pc=0.0012] with
the risk development of active extrapulmonary TB were found. In contrast, the AA genotype was found to be associated with
resistance to extrapulmonary TB [OR=0.19; 95% CI (0.09–0.42); Pc=6.10−6]. No association was found between S138G SNP and pulmonary TB. In conclusion, our study suggested the possible role of IL-10R1

BackgroundThe twenty first century can be called the genomic era referring to the rapid development of genetics, and the beginning of genomic medicine. An initial step towards genomic medicine is to evaluate the knowledge and attitude towards genetic testing among different populations. The aims of this study were to assess the genetic knowledge and attitude towards genetic testing among the Jordanian population and patients with immune diseases. In addition, we evaluated the association between knowledge, attitude and several demographic factors of the population.MethodsThis study was performed using an online questionnaire that was distributed to respondents from different regions of Jordan.ResultsA total of 1149 participants were recruited from the Jordanian population. Overall factual genetic knowledge of the participants was good (65.4%), with education level, working or studying in a health-related field and household average monthly income being significant predictors of factual knowledge scores (P = 0.03, P < 0.001 and P < 0.001, respectively). However, factual knowledge results revealed that scores of questions related to diseases were significantly higher than scores of gene-related scientific facts (P < 0.01). Participants of our study reported to have low perceived knowledge on medical uses (39.5%) and social consequences (23.9%) of genetic testing. Regarding the participants’ attitudes, favorable attitudes towards genetic testing were prevailing (91.5%). Favorable attitudes were more prominent among higher educated participants, and participants with higher scores of factual knowledge.ConclusionDespite the fact that our Jordanian-based study revealed a good level of genetic knowledge as well as a favorable attitude towards genetic testing, we realized an imbalance of knowledge between gene-related scientific facts and disease-related concepts as well as between factual and perceived genetic knowledge, which indicates the necessity of increasing the awareness about genetic testing in order to ensure that individuals can take informed decisions that help in the employment of personalized medicine. 相似文献

BackgroundAngiotensin-converting enzyme (ACE) stimulates angiogenesis that leads to the development of diabetic retinopathy (DR). Alu repetitive elements in ACE gene increase the expression of this enzyme. We investigated the frequency of Alu repetitive elements, insertion/deletion (I/D) polymorphism, in angiotensin-converting enzyme among diabetic retinopathy patients and whether this polymorphism is associated with the severity of retinopathy in Jordanians with type 2 diabetes.MethodsA total of 277 subjects participated in this case/ control study (100 diabetic patients without DR, 82 diabetic patients with DR, and 95 healthy control). Blood samples were withdrawn, followed by DNA extraction. Alu repetitive elements were examined by polymerase chain reaction followed by gel electrophoresis.ResultsThe genotype and allele frequencies among diabetic patients, were close to healthy controls (genotypes, II 44.4 vs. 44.7%, ID 44.4 vs. 42.6%, DD 12.2 vs. 12.8%, P = 0.402 and 0.677 respectively, alleles, I 65.6 vs. 66%, D 34.4 vs. 34%, P=0.863). Complicated diabetics with retinopathy showed similar genotype and allele frequency to those without complications. The severity of diabetic retinopathy in affected individuals was not correlated with I/D polymorphism (P=0.862).ConclusionsWe conclude that the presence of Alu repetitive elements did not increase the development or progression risk to retinopathy in Jordanian type 2 diabetic patients. Fungal β-glucosidases were extensively studied regarding their various potential biotechnology applications. Here, we report the selection of Fusarium solani strain producing high yield of β-glucosidase activity. The effect of some factors on β-glucosidase production was studied including: Initial pH, medium composition, concentration of carbon and nitrogen sources, and particle size of raw substrates. The optimal enzyme production was obtained with 4?units of pH. The highest β-glucosidase activity was produced on 4% wheat bran (WB) as raw carbon sources, reaching 5?U/mL. A positive correlation between WB particle size and the β-glucosidase production level was settled. The last one was enhanced to 13.60?U/mL in the presence of 0.5% (w/v) of ammonium sulfate. Interestingly, the activated charcoal was used as an inexpensive reagent enabling a rapid and efficient purification prior step that improved the enzyme-specific activity. Eventually, F. solani β-glucosidase acts efficiently during the bioconversion process of oleuropein. Indeed, 82.5% of oleuropein was deglycosylated after 1?hr at 40°C. Altogether, our data showed that the β-glucosidase of F. solani has a potential application to convert oleuropein to ameliorate food quality. 相似文献

From June 2004 to May 2006, the reproductive biology of the blue runner Caranx crysos was studied in the Gulf of Gabes (Southern Tunisia). Of 1668 individuals examined, 777 were females (46.6%) and 891 were males (53.4%). The sex ratio significantly deviated in favour of males (♀:♂ = 0.87 : 1). The difference in the numbers of females and males was significant among size‐classes. However, there was no significant difference between sexes over months and seasons. Monthly changes in the Gonado‐Somatic Index (GSI) showed a rapid increase during summer to very high levels in July (3.51% for males and 2.55% for females) and August (3.47% for males and 2.59% for females) before declining sharply in September (0.58% for females and 0.38% for males). The gametogenesis activity began with a pre‐maturation phase, from 20 May to the first 10 days of July, followed by a pre‐spawning phase, from 20 July to 20 August. From the last 10 days of August to 20 September the gonads were in the ripe and spawning stages. From the end of September to early May, gonads were in the post‐spawning and resting stages. The size at which 50% of the population reached sexual maturity was significantly different in males and females: males attained sexual maturity at fork length FL50 = 210.20, whereas females attained maturity at FL50 = 222.3 mm. Summary Bioflocculation of bacteria isolated from the marine biomass acclimatized to textile wastewater has been achieved. Pseudomonas was the main genus found in the marine biomass. This bacterium showed an important hydrophobicity but slightly weaker than
that of pilot plant genera adapted to the conventional sewage. This hydrophobicity provided the bacteria with hydrophobic
characters, especially those of the polymeric extracellular substances (ECS) released in the sewage. Whereas some factors
such as the divalent cations, calcium and magnesium had an effect on this bacterial hydrophobicity, glucose concentration
did not have any impact. Furthermore, the physiological state of the cells hatched in the sewage had an impact on the hydrophobicity.
Indeed, during the logarithmic growth phase, cell hydrophobicity increase enhanced floc formation, thus improving wastewater

MRI biomarkers of tumor edema, vascular permeability, blood volume, and average vessel caliber are increasingly being employed to assess the efficacy of tumor therapies. However, the dependence of these biomarkers on a number of physiological factors can compromise their sensitivity and complicate the assessment of therapeutic efficacy. Here we examine the response of these MRI tumor biomarkers to cediranib, a potent vascular endothelial growth factor receptor (VEGFR) inhibitor, in an orthotopic mouse glioma model. A significant increase in the tumor volume and relative vessel caliber index (rVCI) and a slight decrease in the water apparent diffusion coefficient (ADC) were observed for both control and cediranib treated animals. This contrasts with a clinical study that observed a significant decrease in tumor rVCI, ADC and volume with cediranib therapy. While the lack of a difference between control and cediranib treated animals in these biomarker responses might suggest that cediranib has no therapeutic benefit, cediranib treated mice had a significantly increased survival. The increased survival benefit of cediranib treated animals is consistent with the significant decrease observed for cediranib treated animals in the relative cerebral blood volume (rCBV), relative microvascular blood volume (rMBV), transverse relaxation time (T2), blood vessel permeability (K(trans)), and extravascular-extracellular space (ν(e)). The differential response of pre-clinical and clinical tumors to cediranib therapy, along with the lack of a positive response for some biomarkers, indicates the importance of evaluating the whole spectrum of different tumor biomarkers to properly assess the therapeutic response and identify and interpret the therapy-induced changes in the tumor physiology. 相似文献

A benzoylphenyl urea insect growth regulator with the common name noviflumuron was evaluated for efficacy and residual activity on the German cockroach, Blattella germanica (L.). In laboratory studies evaluating residual activity, 0.05% noviflumuron suspension concentrate produced 100% nymphal mortality 120 d after application to steel and masonite substrates. Residual activity of noviflumuron was more variable on painted plywood substrates compared with stainless steel and masonite. In bioassay arenas, population reductions caused by noviflumuron were significantly greater than Archer and the untreated populations. After 16 wk, populations exposed to 0.05, 0.1, and 0.2% noviflumuron were reduced by 51.9 +/- 19.8, 62.2 +/- 6.5, and 62.6 +/- 18.4%, respectively. Control cockroach populations and populations exposed to 1.3% pyriproxyfen at labeled rate (Archer, 0.61 g/m2) increased by 1286.3 +/- 125.1 and 937.2 +/- 137.1%, respectively, at the end of 16 wk. A field study in multifamily housing complexes showed noviflumuron (0.2 and 0.5%) to provide 73.3 +/- 8.0 and 90.6 +/- 3.6% trap catch reduction at 4 wk posttreatment, respectively. There were no significant differences in the performance of noviflumuron, Maxforce FC Roach Bait Stations (0.05% [AI] fipronil), and Avert dust bait (0.05% [AI] abamectin B1). Noviflumuron shows excellent potential for use in cockroach management programs. 相似文献
