Summary It has shown that several characteristics of high-producing industrial strains of Penicillium chrysogenum tend to segregate in the course of cultivation (slant-to-slant transfer). Segregation includes a decrease in the yield of penicillin, mean conidial size, mean size of the nuclei, and an increase in the proportion of morphologically wild-type colonies. These lower-producing segregants also have a higher sensitivity against ultraviolet radiation and, as shown by cytofluorometric methods, a lower DNA content in the condia, a decrease in phosphate uptkae and in the activity of extracellular alkaline phosphatases compared to high-producing strains. Obviously, during mutagenesis/selection programmes ploidy mutants have been selected, which entails an increase in the number of genes coding enzymes responsible for penicillin biosynthesis. In the absence of selection pressure these high-producing strains segregate to lower-producing strains by chromosome losses in the course of slant-to-slant transfers.

Catabolism of equine haptoglobin (Hp), haptoglobin-haemoglobin complex (Hp-Hb) and haemoglobin (Hb) in the hen was studied in the liver reticuloendothelial system (RES) in which endocytosis of protein was inhibited by aggregated denatured albumin (Agr-Alb). Intravenous injection of Agr-Alb together with equine [125I]Hp, [125I]Hp-Hb or [125I]Hb partially inhibited elimination of these proteins from hen circulation. The decrease of clearance was less pronounced when the labelled proteins were introduced 30 min after Agr-Alb injection due to stimulation of phagocytosis in RES by Agr-Alb. Elimination of equine proteins (Hp, Hp-Hb and Hb) from hen circulation by RES is only one of the possible metabolic fates of these proteins. 相似文献

Breakage of human chromosomes was studied in vitro by treating leukocyte cultures for 8 hours with tretamine, tepa, hempa, hemel, and apholate. The aziridines tretamine, tepa, and apholate effectively induced aberrations; the dimethylamino compounds, hemel and hempa, induced few. Tretamine, the most effective compound, induced aberrations in 78% of the cells; hempa, the least effective, induced aberrations in 14%, even at a 1000-fold greater molar concentration. This study aimed to obtain and characterize an RU-β-CD complex in the context of investigating the possibility of changes in the solubility, stability, antioxidative and microbiological activity as well as permeability of complexated rutin as against its free form. The formation of the RU-β-CD complex via a co-grinding technique was confirmed by using DSC, SEM, FT-IR and Raman spectroscopy, and its geometry was assessed through molecular modeling. It was found that the stability and solubility of the so-obtained complex were greater compared to the free form; however, a slight decrease was observed inits antibacterial potency. An examination of changes in the EPR spectra of thecomplex excluded any reducing effect of complexation on the antioxidative activity of rutin. Considering the prospect of preformulation studies involving RU-β-CD complexes, of significance is also the observed possibility of prolongedly releasing rutin from the complex at a constant level over along period of 20 h, and the fact that twice as much complexated rutin was able topermeate compared to its free form. 相似文献
