Recent observations confirm the rising temperatures of Atlantic waters transported into the Arctic Ocean via the West Spitsbergen Current (WSC). We studied the overall abundance and population structure of the North Atlantic keystone zooplankton copepod Calanus finmarchicus, which is the main prey for pelagic fish and some seabirds, in relation to selected environmental variables in this area between 2001 and 2011, when warming in the Arctic and Subarctic was particularly pronounced. Sampling within a 3‐week time window each summer demonstrated that trends in the overall abundance of C. finmarchicus varied between years, with the highest values in “extreme” years, due to high numbers of nauplii and early copepodite stages in colder years (2001, 2004, 2010), and contrary to that, the fifth copepodite stage (C5) peaking in warm years (2006, 2007, 2009). The most influential environmental variable driving C. finmarchicus life cycle was temperature, which promoted an increased C5 abundance when the temperature was above 6°C, indicating earlier spawning and/or accelerated development, and possibly leading to their development to adults later in the summer and spawning for the second time, given adequate food supply. Based on the presented high interannual and spatial variability, we hypothesize that under a warmer climate, C. finmarchicus may annually produce two generations in the southern part of the WSC, what in turn could lead to food web reorganization of important top predators, such as little auks, and induce northward migrations of fish, especially the Norwegian herring.  相似文献

Single infective juveniles of Heterorhabditis bacteriophora, H. megidis (Nematoda: Heterorhabditidae), Steinernema arenarium, S. carpocapsae and S. feltiae (Nematoda: Steinernematidae) were used to infect single Galleria mellonella (Lepidoptera: Pyralidae) larvae. Four parameters of entomopathogenic nematodes pathogenicity were assessed: the mortality of insects, infectivity of nematodes, number of nematodes established per single G. mellonella, and degree of infective juveniles colonization (percent of infective juveniles which intestine was colonized by symbiotic bacteria). The accuracy, repeatability, and versatility for different species of EPNs in bioassay arenas were compared. Our modifications of the original methods yielded ~ 50% higher efficiency of infective juveniles in cell culture plates and > 20% higher efficiency in centrifuge test tubes. The efficiency of nematodes in cell culture plates (39–77%) was relatively low, especially in the case of Heterorhabditis spp. In the bioassay arena, infective juveniles migrated between cells. The results of our studies indicate that the pathogenicity of EPNs should be assessed in centrifuge test tubes. In these arenas, the infectivity of single IJs was ~ 90% for Heterorhabditis spp. and ~ 95% for Steinernema spp. The degree of colonization of the EPN isolates by symbiotic bacteria was in the range of 96–98%.  相似文献

Listeria monocytogenes and other pathogenic bacteria modify their peptidoglycan to protect it against enzymatic attack through the host innate immune system, such as the cell wall hydrolase lysozyme. During our studies on GpsB, a late cell division protein that controls activity of the bi‐functional penicillin binding protein PBP A1, we discovered that GpsB influences lysozyme resistance of L. monocytogenes as mutant strains lacking gpsB showed an increased lysozyme resistance. Deletion of pbpA1 corrected this effect, demonstrating that PBP A1 is also involved in this. Susceptibility to lysozyme mainly depends on two peptidoglycan modifying enzymes: The peptidoglycan N‐deacetylase PgdA and the peptidoglycan O‐acetyltransferase OatA. Genetic and biochemical experiments consistently demonstrated that the increased lysozyme resistance of the ΔgpsB mutant was PgdA‐dependent and OatA‐independent. Protein‐protein interaction studies supported the idea that GpsB, PBP A1 and PgdA form a complex in L. monocytogenes and identified the regions in PBP A1 and PgdA required for complex formation. These results establish a physiological connection between GpsB, PBP A1 and the peptidoglycan modifying enzyme PgdA. To our knowledge, this is the first reported link between a GpsB‐like cell division protein and factors important for escape from the host immune system.  相似文献

Membrane-anchored lipoproteins have a broad range of functions and play key roles in several cellular processes in Gram-positive bacteria. BA0330 and BA0331 are the only lipoproteins among the 11 known or putative polysaccharide deacetylases of Bacillus anthracis. We found that both lipoproteins exhibit unique characteristics. BA0330 and BA0331 interact with peptidoglycan, and BA0330 is important for the adaptation of the bacterium to grow in the presence of a high concentration of salt, whereas BA0331 contributes to the maintenance of a uniform cell shape. They appear not to alter the peptidoglycan structure and do not contribute to lysozyme resistance. The high resolution x-ray structure of BA0330 revealed a C-terminal domain with the typical fold of a carbohydrate esterase 4 and an N-terminal domain unique for this family, composed of a two-layered (4 + 3) β-sandwich with structural similarity to fibronectin type 3 domains. Our data suggest that BA0330 and BA0331 have a structural role in stabilizing the cell wall of B. anthracis.  相似文献

The study was aimed to establish the contribution of the dorsal branch of the accessory nerve (DBXI) to the innervation of the porcine trapezius muscle (TRAP). Combined retrograde tracing using fluorescent tracer Fast blue (FB) and surgical denervation procedure (excision of DBXI segment) were applied . FB+ neurons supplying the cervical (c‐TRAP) and thoracic part (th‐TRAP) of TRAP were localized in following nerve centres: the ipsilateral ventral horn of the grey matter of cervical neuromers, ipsilateral spinal ganglia and bilateral sympathetic chain ganglia. After the excision of DBXI segment, no FB+ motoneurons supplying c‐TRAP were found while the mean number of those supplying th‐TRAP was significantly decreased. A slight decrease in average numbers of sensory and autonomic neurons implemented in the innervation of both parts of TRAP was also observed. This study has revealed that in the pig DBXI is the only motor pathway to c‐TRAP while the vast majority of motoneurons supplying porcine th‐TRAP send their axons via DBXI.  相似文献

Effects of methyl jasmonate and excess copper on root and leaf growth   总被引:1,自引:0,他引:1  
A short time effects of 25 and 150 μM Cu2+ or 50 μM methyl jasmonate (MJ) on growth of roots and leaves of Phaseolus coccineus, Allium cepa and Zea mays were investigated. Both Cu2+ and MJ inhibited root growth. Jasmonate synthesis inhibitors (ibuprofen, IB, salicylhydroxamic acid, SHAM, and propylgallate, PG) partially reversed the inhibitory effect of Cu2+ in P. coccineus, but in A. cepa this effect was not clear. Pretreatment with NADPH oxidase inhibitor (20 mM imidazole, IM), and especially ethylene inhibitor (silver thiosulphate, STS) mostly weakened Cu2+ effect on root growth in P. coccineus and A. cepa. The growth of P. coccineus leaves also slowed down by Cu2+ and this effect was partially ameliorated by IB, PG and IM, and completely by SHAM and STS. In Z. mays the effect of STS was considerably lower than that of PG and SHAM which reversed the effect of Cu2+. These results indicate that jasmonate, ethylene and NADPH oxidase activity may be involved in Cu2+ inhibitory action on the roots of dicotyledon plants, but in A. cepa only ethylene and NADPH oxidase are involved. However, leaf growth inhibition induced by excess Cu2+ is connected in Z. mays especially with jasmonate, and in P. coccineus with ethylene, NADPH oxidase and, to a minor degree, with jasmonate.  相似文献

Familial hypertrophic cardiomyopathy (HCM) displays autosomal dominant inheritance with incomplete penetration of defective genes. Data concerning the familial occurrence of ventricular preexcitation, i.e. Wolff-Parkinson-White (WPW) syndrome, also indicate autosomal dominant inheritance. In the literature, only a gene mutation on chromosome 7q3 has been described in familial HCM coexisting with WPW syndrome to date. The present paper describes the case of a 7-year-old boy with HCM and coexisting WPW syndrome. On his chromosome 14, molecular diagnostics revealed a C 9123 mutation (arginine changed into cysteine in position 453) in exon 14 in a copy of the gene for beta-myosin heavy chain (MYH7). It is the first known case of mutation of the MYH7 gene in a child with both HCM and WPW. Since no linkage between MYH7 mutation and HCM with WPW syndrome has been reported to date, we cannot conclude whether the observed mutation is a common cause for both diseases, or this patient presents an incidental co-occurrence of HCM (caused by MYH7 mutation) and WPW syndrome.  相似文献

In this work we studied the reaction of four quinones, 1,4-benzoquinone (1,4-BQ), 2,5-dimethyl-1,4-benzoquinone (2,5-DM-1,4-BQ), tetrachloro-1,4-benzoquinone (TC-1,4-BQ) and 1,4-naphthoquinone (1,4-NQ) with jack bean urease in phosphate buffer, pH 7.8. The enzyme was allowed to react with different concentrations of the quinones during different incubation times in aerobic conditions. Upon incubation the samples had their residual activities assayed and their thiol content titrated. The titration carried out with use of 5,5'-di-thiobis(2-nitrobenzoic) acid was done to examine the involvement of urease thiol groups in the quinone-induced inhibition. The quinones under investigation showed two distinct patterns of behaviour, one by 1,4-BQ, 2,5-DM-1,4-BQ and TC-1,4-BQ, and the other by 1,4-NQ. The former consisted of a concentration-dependent inactivation of urease where the enzyme-inhibitor equilibrium was achieved in no longer than 10min, and of the residual activity of the enzyme being linearly correlated with the number of modified thiols in urease. We concluded that arylation of the thiols in urease by these quinones resulting in conformational changes in the enzyme molecule is responsible for the inhibition. The other pattern of behaviour observed for 1,4-NQ consisted of time- and concentration-dependent inactivation of urease with a nonlinear residual activity-modified thiols dependence. This suggests that in 1,4-NQ inhibition, in addition to the arylation of thiols, operative are other reactions, most likely oxidations of thiols provoked by 1,4-NQ-catalyzed redox cycling. Various environmental and physiological factors that affect sex expression in plants have been identified. We made the presumption that in perennials, age may be a key factor that significantly diversifies sex expression over consecutive years of an individual's life. To test this hypothesis, we chose the sedge Carex secalina, a plant that reproduces only sexually and exhibits different sex expression patterns. These patterns had been previously observed in natural populations. In a four‐year experiment, the sex of spikes formed on reproductive tillers of 100 individuals originating from three populations was monitored. A significant association between individuals’ age and number of female/male spikes was found for each population. In addition, significant differences between the populations in the ratio of shoots with bisexual spikes to shoots with unisexual spikes were revealed. We also showed that the proportion of shoots with bisexual spikes in the individual populations changed significantly with the age of an individual and that in the successive years of the individual's life, the production level of female and male structures changed. Moreover, an age‐dependent decrease in both the number and length of female and male spikes was observed.  相似文献
