The incorporation of radiolabeled arachidonic acid and saturated fatty acids into choline-linked phosphoglycerides (PC) of rabbit and human neutrophils was investigated by resolving the individual molecular species by reversed-phase high performance liquid chromatography. PC from neutrophils incubated with a mixture of [3H]arachidonic acid and [14C]stearic or [14C]palmitic acid contains both radiolabels; however, double labeling of individual molecular species is minimal. After labeling for 2 h, the [3H]arachidonate is distributed almost equally between diacyl and 1-O-alkyl-2-acyl species, but it is incorporated into diacyl species containing unlabeled stearate or palmitate at the sn-1 position. In contrast, labeled saturated fatty acids are incorporated only into diacyl species and contain predominantly oleate and linoleate at the sn-2 position. Labeled linoleate is not incorporated into ether-linked species, but is found in the same species as labeled stearate. The findings suggest that mechanisms exist in neutrophils for specific shunting of exogenous arachidonic acid into certain phospholipid molecular species and support the concept that the 1-O-alkyl-2-arachidonoyl species may be a functionally segregated pool of arachidonic acid within the PC of neutrophils.  相似文献

Aim To assess the distribution, group size, seasonal occurrence and annual trends of cetaceans. Location The study area included all major inland waters of Southeast Alaska. Methods Between 1991 and 2007, cetacean surveys were conducted by observers who kept a constant watch when the vessel was underway and recorded all cetaceans encountered. For each species, we examined distributional patterns, group size, seasonal occurrence and annual trends. Analysis of variance (anova F) was used to test for differences in group sizes between multiple means, and Student’s t‐test was used to detect differences between pairwise means. Cetacean seasonal occurrence and annual trends were investigated using a generalized linear model framework. Results Humpback whales (Megaptera novaeangliae) were seen throughout the region, with numbers lowest in spring and highest in the fall. Fin whale (Balaenoptera physalus) and minke whale (Balaenoptera acutorostrata) distributions were more restricted than that reported for humpback whales, and the low number of sightings precluded evaluating seasonal trends. Three killer whale (Orcinus orca) eco‐types were documented with distributions occurring throughout inland waters. Seasonal patterns were not detected or could not be evaluated for resident and offshore killer whales, respectively; however, the transient eco‐type was more abundant in the summer. Dall’s porpoise (Phocoenoides dalli) were distributed throughout the region, with more sightings in spring and summer than in fall. Harbour porpoise (Phocoena phocoena) distribution was clumped, with concentrations occurring in the Icy Strait/Glacier Bay and Wrangell areas and with no evidence of seasonality. Pacific white‐sided dolphins (Lagenorhynchus obliquidens) were observed only occasionally, with more sightings in the spring. For most species, group size varied on both an annual and seasonal basis. Main conclusions Seven cetacean species occupy the inland waters of Southeast Alaska, with distribution, group size, seasonal occurrence and annual trends varying by species. Future studies that compare spatial and temporal patterns with other features (e.g. oceanography, prey resources) may help in identifying the key factors that support the high density and biodiversity of cetaceans found in this region. An increased understanding of the region’s marine ecology is an essential step towards ensuring the long‐term conservation of cetaceans in Southeast Alaska.  相似文献

The addition of physiological concentrations of zinc (25-200 (Μg/dL) to Dulbecco’s Modified Eagle’s Medium containing tibiae from 19-d chick embryos resulted in a concentration-dependent increase in tibial content of tartrate-resistant acid phosphatase (TRAP) and an increase in bone resorption, as measured by tibial calcium release. This increase in bone resorption was additive to the resorptive effect resulting from the addition of 10-9-10-7 M parathyroid hormone (PTH), but was not additive to similar effects produced by the addition of 10-9-10-7 M prostaglandin E2 (PGE2). An inhibitor of prostaglandin synthesis, flurbiprofen (10-6 M), did not influence the effect of zinc on bone resorption. However, the addition of 2,6-pyridinedicarboxylic acid (10-3 M, 2,6-PDCA), a chelator of zinc, did attenuate the effects of zinc, as did the addition of an inhibitor of DNA replication (hydroxyurea, 10-3 M). Hydroxyurea also attenuated the bone resorptive response to PGE2, but had no influence on the effects of PTH. These results indicate that physiological concentrations of zinc alter bone resorptive rates in vitro by a mechanism that is dependent on DNA replication.  相似文献

The native phytoseiidAmblyseius womersleyi Schicha controlledTetranychus urticae Koch in two separate commercial strawberry plantings near Nambour, Queensland, in 1981 and 1982.Phytoseiulus persimilis Athias-Henriot, either mass-released or naturally occurring, effectively controlledT. urticae in eleven commercial strawberry plantings during 1982–1984. Chemical controls for insect pests and diseases were successfully integrated with the predator. Birds combat ectoparasites with many defences but the first line of defence is grooming behaviour, which includes preening with the bill and scratching with the feet. Preening has been shown to be very effective against ectoparasites. However, most tests have been with feather lice, which are relatively slow moving. Less is known about the effectiveness of preening as a defence against more mobile and evasive ectoparasites such as hippoboscid flies. Hippoboscids, which feed on blood, have direct effects on the host such asanaemia, as well as indirect effects as vectors of pathogens. Hence, effective defence against hippoboscid flies is important. We used captive Rock Pigeons (Columba livia) to test whether preening behaviour helps to control pigeon flies (Pseudolynchia canariensis). We found that pigeons responded to fly infestation by preening twice as much as pigeons without flies. Preening birds killed twice as many flies over the course of our week-long experiment as birds with impaired preening; however, preening did not kill all of the flies. We also tested the role of the bill overhang, which is critical for effective preening against feather lice, by experimentally removing the overhang and re-measuring the effectiveness of preening against flies. Birds without overhangs were as effective at controlling flies as were birds with overhangs. Overall, we found that preening is effective against mobile hippoboscid flies, yet it does not eliminate them. We discuss the potential impact of preening on the transmission dynamics of blood parasites vectored by hippoboscid flies.  相似文献
