The successful use of macroinvertebrates as indicators of stream condition in bioassessments has led to heightened interest throughout the scientific community in the prediction of stream condition. For example, predictive models are increasingly being developed that use measures of watershed disturbance, including urban and agricultural land-use, as explanatory variables to predict various metrics of biological condition such as richness, tolerance, percent predators, index of biotic integrity, functional species traits, or even ordination axes scores. Our primary intent was to determine if effective models could be developed using watershed characteristics of disturbance to predict macroinvertebrate metrics among disparate and widely separated ecoregions. We aggregated macroinvertebrate data from universities and state and federal agencies in order to assemble stream data sets of high enough density appropriate for modeling in three distinct ecoregions in Oregon and California. Extensive review and quality assurance of macroinvertebrate sampling protocols, laboratory subsample counts and taxonomic resolution was completed to assure data comparability. We used widely available digital coverages of land-use and land-cover data summarized at the watershed and riparian scale as explanatory variables to predict macroinvertebrate metrics commonly used by state resource managers to assess stream condition. The “best” multiple linear regression models from each region required only two or three explanatory variables to model macroinvertebrate metrics and explained 41–74% of the variation. In each region the best model contained some measure of urban and/or agricultural land-use, yet often the model was improved by including a natural explanatory variable such as mean annual precipitation or mean watershed slope. Two macroinvertebrate metrics were common among all three regions, the metric that summarizes the richness of tolerant macroinvertebrates (RICHTOL) and some form of EPT (Ephemeroptera, Plecoptera, and Trichoptera) richness. Best models were developed for the same two invertebrate metrics even though the geographic regions reflect distinct differences in precipitation, geology, elevation, slope, population density, and land-use. With further development, models like these can be used to elicit better causal linkages to stream biological attributes or condition and can be used by researchers or managers to predict biological indicators of stream condition at unsampled sites.  相似文献

Defining the design principles guiding the fabrication of superior biocomposite structures from an assemblage of ordinary molecules is a key goal of biomimetics. Considering their low degree of mineralization, Glycera jaws have been shown to be extraordinarily resistant to abrasion based on the metric hardness3/Young's modulus2. The jaws also exhibit an impressive chemical inertness withstanding boiling concentrated hydrochloric acid as well as boiling concentrated sodium hydroxide. A major organic component largely responsible for the chemical inertness of the jaws has been characterized using a spectrophotometric assay for melanin content, 13C solid state nuclear magnetic resonance, IR spectroscopy, and laser desorption ionization-time of flight mass spectrometry and is identified here as a melanin-like network. Although melanin is widely distributed as a pigment in tissues and other structural biomaterials, to our knowledge, Glycera jaws represent the first known integument to exploit melanin as a cohesive load- and shape-bearing material.  相似文献

Marigold (cv. Lemon Gem), castor bean, ornamental pepper (cv. Black Pearl and Purple Flash), gerbera daisy (cv. Festival), feverfew, and sunflower (cv. Choco Sun) were evaluated for their suitability as banker plants (BP) for Orius insidiosus (Hemiptera: Anthocoridae) in commercial greenhouses. Oviposition, egg hatch, nymphal development to adulthood, and population increase were quantified in laboratory trials. Assessments of oviposition and egg hatch indicated that all plants tested were equally accepted by O. insidiosus. Nymphal development to adulthood and survival tests indicated that gerbera may be a suitable BP as survival was the highest (58.1 %), whereas marigold would not be an acceptable BP as only 10.7 % of nymphs survived to adulthood. Nymphal development time differed by only one day among all plants. In greenhouse cage experiments, Purple Flash pepper supported the greatest population growth over a ten week period. Based on the combined results from all tests, Purple Flash pepper appears to have the greatest potential as a BP species for O. insidiosus.  相似文献

Regulation of epithelial barrier function requires targeted insertion of tight junction proteins that have distinct selectively permeable characteristics. The insertion of newly synthesized proteins and recycling of internalized tight junction components control both polarity and junction function. Here we show that the small GTPase Rab14 regulates tight junction structure. In Madin–Darby canine kidney (MDCK) II cells, Rab14 colocalizes with junctional proteins, and knockdown of Rab14 results in increased transepithelial resistance. In cells without Rab14, there are small changes in the trafficking of claudin-1 and occludin. In addition, there is substantial depletion of the leaky claudin, claudin-2, but not other tight junction components. The loss of claudin-2 is complemented by inhibition of lysosomal function, suggesting that Rab14 sorts claudin-2 out of the lysosome-directed pathway. MDCK I cells lack claudin-2 endogenously, and knockdown of Rab14 in these cells does not result in a change in transepithelial resistance, suggesting that the effect is specific to claudin-2 trafficking. Furthermore, leaky claudins have been shown to be required for epithelial morphogenesis, and knockdown of Rab14 results in failure to form normal single-lumen cysts in three-dimensional culture. These results implicate Rab14 in specialized trafficking of claudin-2 from the recycling endosome.  相似文献

The biochemistry of mussel adhesion has inspired the design of surface primers, adhesives, coatings and gels for technological applications. These mussel-inspired systems often focus on incorporating the amino acid 3,4-dihydroxyphenyl-L-alanine (Dopa) or a catecholic analog into a polymer. Unfortunately, effective use of Dopa is compromised by its susceptibility to auto-oxidation at neutral pH. Oxidation can lead to loss of adhesive function and undesired covalent cross-linking. Mussel foot protein 5 (Mfp-5), which contains ∼30 mole % Dopa, is a superb adhesive under reducing conditions but becomes nonadhesive after pH-induced oxidation. Here we report that the bidentate complexation of borate by Dopa to form a catecholato-boronate can be exploited to retard oxidation. Although exposure of Mfp-5 to neutral pH typically oxidizes Dopa, resulting in a>95% decrease in adhesion, inclusion of borate retards oxidation at the same pH. Remarkably, this Dopa-boronate complex dissociates upon contact with mica to allow for a reversible Dopa-mediated adhesion. The borate protection strategy allows for Dopa redox stability and maintained adhesive function in an otherwise oxidizing environment.  相似文献

Between 1991 and 1993, Alaska harbor porpoise ( Phocoena phocoena ) abundance was investigated during aerial surveys throughout much of the coastal and offshore waters from Bristol Bay in the eastern Bering Sea to Dixon Entrance in Southeast Alaska. Line-transect methodology was used, and only those observations made during optimal conditions were analyzed. Survey data indicated densities of 4.48 groups/100 km2, or approximately 3,531 harbor porpoises (95% C. I. 2,206-5,651) in Bristol Bay and 0.54 groups/100 km2, or 136 harbor porpoises (95% C. I. 11-1,645) for Cook Inlet. Efforts off Kodiak Island resulted in densities of 1.85 groups/100 km2, or an abundance estimate of 740 (95% C. I. 259-2,115). Surveys off the south side of the Alaska Peninsula found densities of 2.03 groups/100 km2 and an abundance estimate of 551 (95% C. I. 423-719). Surveys of offshore waters from Prince William Sound to Dixon Entrance yielded densities of 4.02 groups/100 km2 and an abundance estimate of 3,982 (95% C. I. 2,567-6,177). Combining all years and areas yielded an uncorrected density estimate of 3.82 porpoises per 100 km2, resulting in an abundance estimate of 8,940 porpoises (CV = 13.8%) with a 95% confidence interval of 6,746-11,848. Using correction factors from other studies to adjust for animals missed by observers, the total number of Alaska harbor porpoises is probably three times this number.  相似文献

Many stimulators of prostaglandin production are thought to activate the Ca2+- and phospholipid-dependent protein kinase first described by Nishizuka and his colleagues (Takai, Y., Kishimoto, A., Iwasa, Y., Kawahara, Y., Mori, T., and Nishizuka, Y. (1979) J. Biol. Chem. 254, 3692-3695. In this paper we report evidence that the activation of protein kinase C caused by 12-O-tetradecanoylphorbol-13-acetate (TPA) is involved in the increased prostaglandin production induced by 12-O-tetradecanoylphorbol-13-acetate in Madin-Darby canine kidney (MDCK) cells. We have shown that TPA activates protein kinase C in MDCK cells with similar dose response curve as observed for TPA induction of arachidonic acid release in MDCK cells. Activation of protein kinase C was associated with increased phosphorylation of proteins of 40,000 and 48,000 daltons. We used two compounds (1-O-octadecyl-2-O-methyl-rac-glycero-3-phosphocholine (ET-18-OMe) and 1-(5-isoquinolinesulfonyl)piperazine) known to inhibit protein kinase C by different mechanisms to further examine if activation of protein kinase C was involved in the increased synthesis of prostaglandins in TPA-treated MDCK cells. We found that both compounds inhibited protein kinase C partially purified from MDCK cells and that ET-18-OMe inhibited the phosphorylation of proteins by protein kinase C in the intact cells. Addition of either compound during or after TPA treatment decreased both release of arachidonic acid from phospholipids and prostaglandin synthesis. Release of [3H]arachidonic acid from phosphatidylethanolamine in TPA-treated cells was blocked by ET-18-OMe or 1-(5-isoquinolinesulfonyl)piperazine addition. However, arachidonic acid release stimulated by A23187 is not blocked by Et-18-OMe. When assayed in vitro, treatment of cells with Et-18-OMe did not prevent the enhanced conversion of arachidonic acid into prostaglandins induced by pretreatment of cells with TPA. Our results suggest that the stimulation of phospholipase A2 activity by TPA occurs via activation of protein kinase C by TPA.  相似文献

Intracellular concentrations of Na+ and K+ of various normal, transformed, and tumor cell cultures were analyzed by atomic absorption spectrophotometry. In all of the cultures analyzed there were markedly different concentrations in the transformed and tumor cells when compared to their normal counterparts. Although increased Na+ was often observed, there were no definitive correlations between absolute ion concentrations, or Na+:K+ ratios, and cell transformation.  相似文献
