Some aspects of the fine structure of the alimentary tract of the infective larva of Breinlia sergenti (Nematoda: Filarioidea).

The fine structure of the alimentary tract of the infective larva of Breinlia sergenti has been studied. The possible derivation of the oesophagus from the pharyngeal thread of the microfilaria is discussed.     

Stimulating effect of the low dose laser - a new hypothesis

Increase in the amount of secretary material (interreceptor matrix) surrounding the visual cells was observed after laser treatment. At the same time, an increase in labelled uridine uptake by the pigment epithelium was detected. The increase uptake was highest 4 h after laser treatment.     

Ultrastructure and fluorescence histochemistry of endocrine (APUD-type) cells in tracheal mucosa of human and various animal species

This study describes distrinctive cells with ultrastructural and histochemical features of APUD-type endocrine cells within the tracheal epithelium of human fetuses, newborns and children as well as different animal species. These cells referred to as Kultschitzky cells (K cells) were found to be argyrophilic, but not argentaffin, and are considered analogous to the same type of cells in lung and gastro-intestinal tract. Fluorescence histochemistry demonstrated the presence of intracellular amine within tracheal K cells, but only after in-vitro or in-vivo administration of amine precursor (L-DOPA). Ultrastructurally, these cells are characterized by the presence of numerous cytoplasmic granules (dense core vesicles) which show species related morphologic variations. Two different types of K cells were found in trachea of lamb and armadillo, each type possessing morphologically different dense core vesicles. In human and rabbit tracheas, only one type of K cell was identified. K cells in the trachea are distributed as single cells between other epithelial cells; neuroepithelial bodies such as those found in bronchial mucosa were not identified. Well differentiated K cells were found in tracheas of early human fetuses and throughout gestation, infancy, and childhood. Preservation of K cells in human autopsy material and widespread occurence of these cells in various laboratory animals will permit further studies into the nature and function of tracheobronchial endocrine cells.     

Effects of subunit V antibodies on the topology of the subunit and the activity of beef heart cytochrome-c oxidase

Redox-sensitive epitopes on subunit V of beef heart cytochrome-c oxidase were demonstrated previously using polyclonal subunit-specific antibodies raised in rabbits. The antibodies only slightly inhibited electron transfer, and the accessibility of their epitopes depended on the presence of a membrane and on the redox state of the oxidase. The present paper describes additional preparations of antibodies raised against subunit V. These antibodies have an even higher subunit specificity, they are more than three times as inhibitory against electron transfer, and their binding does not require a membrane. Moreover, the redox-sensitive nature of their binding to detergent-dispersed oxidase is sensitive to the method of its isolation. We discuss inferences that can be drawn from a detailed quantitative comparison of the interactions of the two antibody preparations with the antigen in different environments. The techniques used in the comparison can be used to examine other perturbants of the oxidase as to their effects on specific segments of the enzyme.     

The role of phosphorylation in the alpha-adrenergic-mediated inhibition of rat hepatic pyruvate kinase

Phenylephrine in the presence of 1-methyl-3-isobutylxanthine and propanolol caused a 40-50% inhibition of pyruvate kinase (type L) activity in isolated hepatocytes, which was accompanied by a 2-3-fold increase in the phosphate content of the enzyme. These changes were blocked by the alpha-adrenergic antagonist dihydroergocryptine and could not be accounted for by the slight increase in cyclic AMP-dependent protein kinase activity generated by the alpha-adrenergic agonist. It is concluded that a significant component of the inhibition of hepatic pyruvate kinase mediated by alpha-adrenergic agonists can be attributed to a cyclic AMP-independent alteration in the phosphorylation state of the enzyme.     

Estimating Arundo donax shoot biomass

We developed an equation for estimating Arundo donax shoot dry weight from shoot length. The equation, shoot dry weight (g) = 14.254 (standard error = ±0.275) × shoot height² (m), was as effective at explaining a high proportion of total variation in shoot dry weight (R² = 0.90) as more complicated equations containing additional morphometric parameters. Tested against two independent datasets, the equation provided accurate estimates of dry weight for shoots ranging from 0.3 to 7.06 m height (dataset 1, P < 0.0001, R² = 0.87, N = 29; dataset 2, P < 0.0001, R² = 0.82, N = 192). The equation provides aboveground biomass estimates from stem counts and heights more rapidly than harvest methods.
