Apramycin is unique in the aminoglycoside family due to its octodiose moiety. However, either the biosynthesis process or the precursors involved are largely unknown. Addition of glycine, as well as serine or threonine, to the Streptomyces tenebrabrius UD2 fermentation medium substantially increases the production of apramycin with little effect on the growth of mycelia, indicating that glycine and/or serine might be involved in the biosynthesis of apramycin. The 13C-NMR analysis of [2-13C] glycine-fed (25% enrichment) apramycin showed that glycine specifically and efficiently incorporated into the only N-CH3 substituent of apramycin on the C7' of the octodiose moiety. We noticed that the in vivo concentration of S-adenosyl methionine increased in parallel with the addition of glycine, while the addition of methione in the fermentation medium significantly decreased the productivity of apramycin. Therefore, the methyl donor function of glycine is proposed to be involved in the methionine cycle but methionine itself was proposed to inhibit the methylation and methyl transfer processes as previously reported for the case of rapamycin. The 15N NMR spectra of [2-13C,15N]serine labeled apramycin indicated that serine may also act as a limiting precursor contributing to the -NH2 substituents of apramycin.  相似文献

目的筛选适合制备检测试剂盒的抗原组合。方法用 ELISA方法,将5株小鼠肝炎病毒(MHV)抗原分别组合作为包被抗原,研究MHV抗原不同组合与标准毒株抗体的反应性,分析MHV各毒株间的差别,进一步比较不同组台抗原的灵敏性、特异性。结果通过对1997~1999年送检238只普通级实验小鼠检测,结果表明,MHV的感染率虽有下降趋势,但感染率仍然很高(59%~87%)。结论我国分离的MHV-HU79株,在本次研究中表明能够用于MHV抗体检测试剂盒。  相似文献

Acipenseriformes is an endangered primitive fish group, which occupies a special place in the history of ideas concerning fish evolution, even in vertebrate evolution. However, the classification and evolution of the fishes have been debated. The mitochondrial DMA (mtDNA) ND4L and partial A7D4 genes were first sequenced in twelve species of the order Acipenseriformes, including endemic Chinese species. The following points were drawn from DNA sequences analysis: (i) the two species of Huso can be ascribed to Acipenser; (ii) A. dabryanus is the mostly closely related to A. sinensis, and most likely the landlocked form of A. sinensis; (iii) genus Acipenser in trans-Pacific region might have a common origin; (iv) mtDNA ND4L and ND4 genes are the ideal genetic markers for phylogenetic analysis of the order Acipenseriformes.  相似文献

ABSTRACT. The life history of the protozoan parasite Ascogregarina taiwanensis in mosquito larvae ( Aedes albopictus , collected in southern Taiwan) was shown to consist of two consecutive stages—intracellular and extracellular. Light microscopy showed that most trophozoites moved into the Malpighian tubules and developed into giant trophozoites during the first day pupa. The locomotion may be associated with bristle-like ridges of the trophozoite. The stage for sexual reproduction, i.e. the gamete, was then formed by segmentation of the giant trophozoite and twisting off the anucleate extremities of the body. Sexual reproduction occurred via fertilization by fusion of two resulting gametes, presumably two opposed sexes. The fused gametes finally generate the formation of the gametocyst, within which oocysts develop by budding from the cytoplasmic mass. This type of sexual reproduction has not been reported previously in any gregarine protozoa. We here proposed it as a new hypothesis for further elucidation of the protozoan reproduction.  相似文献

To investigate whether an erythropoietin (EPO) gene-based therapy could serve as an alternative to the repeated injection of rhEPO in treatment to renal anemia, the genetically modified myoblasts of rats, named Myo/ EPO, were implanted through intramuscular injection to model rats with renal anemia. The hemoglobin (Hb) and hematocrit (HCT) of the rats increased from (92. 5±3.0) g/L and 0.29 ±0.04 to the peak values of (103.8 ±5.0) g/L and 0. 32 ±0. 04 respectively 14 d after implantation, and sustained the pre-implantation level for 90 d. Otherwise, the control rats implanted with Myo/X, which carried the parent retroviral vector, gradually became severe in anemia. The PCR detection for hEPO cDNA in the rat muscle adjacent to injection sites indicated that the Myo/EPO cells survived for a long period in the muscle of rats. The results primarily demonstrate that myoblast gene transfer of EPO is effective for the treatment of rat renal anemia.  相似文献

在对目的DNA序列尤其是高GC含量的片段进行PCR扩增的时候,经常需要对一些试验条件进行优化。在一定条件下,二甲基亚砜、甲酰胺、甘油、NP-40和Tween20等可以在某种程度上提高PCR的特异性和效率。我们在对禾本科lea3基因进行分离克隆时发现了一种新的可以提高PCR产量和特异性的物质——极高热稳定单链结合蛋白(ETSSB),研究发现在每50μlPCR反应体系中加入200ng的ETSSB,可以有效地抑制DNA片段的非特异性条带的产生,并可以提高目的片段的产量。  相似文献

以金百合“普瑞头(Prato)”为试材,研究1-MCP对切花瓶插寿命及相关生理代谢的影响。结果表明:金百合“普瑞头(Prato)”为呼吸跃变型切花。30n1 L^-1的1-MCP能显著延长切花瓶插寿命2.15d、盛开天数增加0.43d,使其花径增大1.68倍(P〈0.01);有效延缓了水分代谢的失调与呼吸峰的出现时间;明显降低了呼吸强度与细胞膜透性,为供试百合品种最适应用浓度。  相似文献

运用邻接法(neibor-joining,NJ)、最大似然法(maximum likehood,ML)对国际病毒分类委员会(Intemational Committee on Taxonomy of Viruses,ICTV)第八次报告中所有已报道的呼肠孤病毒科外层衣壳蛋白序列进行了系统发育分析.结果表明:两种方法构建的系统树拓扑结构基本一致,尽管部分分支略有差异,但都能够反映出科内各属的系统发育关系,分析结果支持了该科的分类地位.同时还比较两种建树方法的异同点,并对建树产生的差异原因作了探讨.  相似文献
