Summary

Proteins synthesized and accumulated during oogenesis or Pseudopotamilla occelata were analyzed by two-dimensional Polyacrylamide gel electrophoresis and fluorography. Significant changes in the patterns of synthesis and accumulation occur during oogenesis, paticularly between previtellogenic and vitellogenic stages, although many of the proteins are represented throughout the process. The changes may be generally dependent upon the variation in the gene expression affecting autosynthesis of proteins. However, appearance of proteins, which occur only at the vitellogenic stages but apparently not synthesized within the oocytes, indicates that heterosynthetic proteins are stage-specifically transported into oocytes.  相似文献

The phylogeographical patterns of the endemic minnow, Candidia barbatus, were examined using mitochondrial (mt) cytochrome b sequences, allozyme electrophoresis, and morphology. The 23 mtDNA haplotypes recovered were grouped into six lineages (A–F). According to a molecular clock of 1.7% per million years, we estimated that the colonization of C. barbatus to Taiwan took place three million years ago with several subsequent vicariance events. In the allozyme analysis, the Bayesian approach partitioned different populations into three clusters, with clusters I, II, and III mainly found in northern, central, and southern Taiwan, respectively. Morphologically, fish from north and south of the Tropic of Cancer had distinct numbers of lateral line scales and were separated into two morphs. The cohabitation of different mtDNA lineages with high allozymic and morphological similarities suggests that interbreeding amongst them is possible. Therefore, in spite of highly differentiated mtDNA, samples from different collecting sites should be considered a single species. Based on the current population distribution and levels of divergence, we propose that both orogenic activities and marine regression during glacial epoch were the two main forces shaping population differentiation of this species. © 2011 The Linnean Society of London, Zoological Journal of the Linnean Society, 2011, 161 , 613–632.  相似文献

Cell and vacuole areas were measured by light microscopy inlongitudinal and transverse sections cut at 0.4-mm intervalsalong the apical 7.2 mm of the primary root of pea. The vacuolararea (or volume) fraction — that is, vacuole area (orvolume) divided by cell area (or volume) — increased fromabout 15 % in cells 0.4 mm from the distal boundary of the apicalmeristem (the cap /root junction), to about 85% in cells situated6.8–7.2 mm from that boundary. At each distance, vacuoledevelopment tended to be greater in the cortex than in the stele.Vacuoles occupied about 22% of the tissue volume in the first1 mm length of root (measured from the cap/root junction), about31 % of the tissue volume in the first 2 mm, and about 45% whensummed over the apical 5-mm length of root. Phosphorus supplyor deprivation produced only minor and non-significant changesin vacuole development. We re‐examine the higher level phylogeny and evolutionary affinities of the family Gelechiidae (Lepidoptera: Gelechioidea) based on DNA sequence data for one mitochondrial gene (cytochrome c oxidase subunit I) and seven nuclear genes (Elongation Factor‐1α, wingless, Ribosomal protein S5, Isocitrate dehydrogenase, Cytosolic malate dehydrogenase, Glyceraldehyde‐3‐phosphate dehydrogenase and Carbamoylphosphate synthase domain protein). Fifty‐two taxa representing nearly all established subfamilies and tribes of Gelechiidae, and about 10% of described gelechiid genera, in addition to five outgroup taxa were sequenced. Data matrices (6157 bp total) were analysed under model‐based evolutionary methods (Maximum Likelihood and Bayesian Inference), resulting in novel high‐level phylogenetic interrelationships. The best supported cladogram divided the Gelechiidae into six distinct clades corresponding to the subfamilies Anacampsinae, Dichomeridinae, Apatetrinae, Thiotrichinae, Anomologinae and Gelechiinae (+ Physoptilinae, which were not available for study). The results suggest the following adjustments in gelechiid interrelationships: Brachmini is nested within Dichomeridinae; Anarsiini is the sister group of Chelariini; Pexicopiinae is the sister group of Apatetrinae, here suggested to be treated as a tribe Pexicopiini of Apatetrinae. A new subfamily Thiotrichinae ( subfam.n. ) is proposed on the basis of the resurrected genus Thiotricha Meyrick ( gen.rev. ), which includes Macrenches Meyrick, Palumbina Rondani and Polyhymno Chambers. Gelechiidae display a wide array of life‐history strategies, but the diversity in patterns of larval mode of life has direct phylogenetic correlation only below subfamily level, suggesting multiple origins and/or frequent reversals for traits such as external or internal feeding and leaf mining within the family.  相似文献

Pollen from chasmogamous flowers of Salpiglossis sinuata L. could not be induced to germinate in vitro unless stigmatal extract was applied to the culture medium. The substance that induces pollen germination in the stigmatal extract is water-soluble and heat-stable. Crosses could not be achieved between chasmogamous and cleistogamous flowers because of structural incompatibility. Pollinated pistils of chasmogamous flowers release a large amount of ethylene. The burst of ethylene release is due to an interaction between pollen tubes and stylar tissue and is directly proportional to the quantity of pollen placed on the stigma. Cleistogamous flower buds also produce a burst of ethylene at the time of pollination within the closed flower. The ethylene release may be a cause of reduced corolla development associated with cleistogamous flowers.  相似文献

The introduction and repression of CAM in Sedurn telephiunmL, a temperate succulent, was investigated in watered, progressivelydrouglited and rewatered plants in growth chambers. Measurementswere made of water vapour and CO2 exchange, titratable acidity(TA) and xylem sap tension. Effects of photoperiod were alsostudied. CAM was induced by drought under long or short days,although when watered no CAM activity was expressed. C3-CAM intermediate plants were used for the investigation ofwater supply. Those which had received water and those drought-stressedboth displayed a similar nocturnal increase in TA, with a day-nightmaximum (H+) of 69 µmol g–1 fr. wt. The wateredplants took up CO2 at a maximum rate of 2?2 µmol m–2s–1 only in the light period, while the droughted plantsshowed a maximum nocturnal CO2 uptake rate of 0?69 µmolm–2 s–1. Subsequently, as CAM was repressed, thewatered S. telephiwn displayed little variation in TA, withconstant levels at 42 µmol g–1 fr. wt. (day 10).After 10 d of drought stress, the CAM characteristics of S.telephiurn were aLso affected, with reduced net CO2 uptake andH+. The transition between C3 and CAM in S. telephium can be describedas a progression in terms of the proportion of respiratory CO2which is recycled and refixed at night as malic acid, in comparisonwith net CO2 uptake. Recycling increased from 20% (day 1) to44% (day 10) as a result of the drought stress and was highin both the CAM-C3 stage (no net CO2 uptake at night) and alsoin the drought-stressed CAM stage (reduced net CO2 uptake atnight). The complete C3-CAM transition occurred in less than8 d, and the stages could be characterized by xylem sap tensionmeasurements: CAM = 0?50 MPa C3-CAM = 0?36 MPa C3 = 0?29 MPa. Several lines of evidence suggest a novel regulatory mechanism for diapause regulation in the gypsy moth. We propose that ecdysteroids play a role in the induction and maintenance of the pharate first instar larval diapause in this species. A 55 kDa gut protein that is indicative of diapause is expressed in intact and neck-ligated pharate larvae but is not expressed when a ligature is placed posterior to the prothorax, site of the prothoracic gland. Guts cultured in vitro for 12 h cease to synthesize the 55 kDa protein, but synthesis of the protein resumes if the culture medium is enriched with a prothorax extract from pharate larvae or a prothoracic gland extract from fifth instar larvae. Injection of 20-hydroxyecdysone or the ecdysteroid agonist, RH-5992, into isolated abdomens stimulates synthesis of the diapause-specific 55 kDa protein, suggesting that the essential factor from the prothorax is an ecdysteroid. KK-42, an imidazole derivative known to inhibit ecdysteroid biosynthesis, averts diapause when applied to prediapausing pharate first instar larvae, but this effect can be countered by application of 20-hydroxyecdysone or RH-5992, i.e. KK-42 treated pharate larvae that are exposed to an ecdysteroid or RH-5992 readily enter diapause. A chilling period (120 days at 5 degrees C) is normally adequate to prompt an immediate termination of diapause when pharate larvae are transferred to 25 degrees C, but if such larvae are held in hanging drop cultures with ecdysteroids they fail to terminate diapause. Together, these results suggest that ecdysteroids are essential for the induction and maintenance of diapause and imply that a drop in the ecdysteroid titer is essential for diapause termination.
