Phlebotomus papatasi is a proven vector of Leishmania major which is one of the causative agents of cutaneous leishmaniasis in the Old World. Although it has a wide geographical range, its population structure is not yet well understood. In an effort to better understand the population dynamics of this vector, we developed a panel of di‐ and trinucleotide microsatellite markers, using a magnetic bead hybridization enrichment protocol. These microsatellite loci showed three to seven alleles with an expected heterozygosity range between 0.702 and 0.876. The level of polymorphisms found in this study suggests that these microsatellite loci can be used for population analysis of P. papatasi.  相似文献

Endogenous pararetroviruses (EPRVs) represent a new class of dispersed repetitive DNA in plants. The genomes of many Nicotiana species and other solanaceous plants are rich in EPRVs. Distinct EPRV families are present in N. sylvestris ( Ns ) and in N. tomentosiformis ( Nto ), the two diploid progenitors of allotetraploid N. tabacum . Nicotiana EPRVs represent an interesting type of repetitive sequence to analyse in polyploids because of their potential impact on plant fitness and the epigenetic architecture of plant genomes. The Ns EPRV family appears identical in N. sylvestris and N. tabacum , indicating little change has occurred in either species since polyploid formation. By contrast, the Nto EPRV family is larger in N. tomentosiformis than in N. tabacum , suggesting either preferential elimination from the polyploid genome or specific accumulation in the diploid genome following polyploidization. The lability of Nto EPRVs might be enhanced by a frequent association with gypsy retrotransposons. Although some EPRVs are probably benign, others are potentially pathogenic or, conversely, determinants of virus resistance. Normally quiescent EPRVs can be reactivated and cause symptoms of infection in hybrids of species that differ in their EPRV content. EPRVs that furnish immunity to the free virus exemplify the selective value of so-called 'junk' DNA. In order to better assess the role of agriculture within the global climate‐vegetation system, we present a model of the managed planetary land surface, Lund–Potsdam–Jena managed Land (LPJmL), which simulates biophysical and biogeochemical processes as well as productivity and yield of the most important crops worldwide, using a concept of crop functional types (CFTs). Based on the LPJ‐Dynamic Global Vegetation Model, LPJmL simulates the transient changes in carbon and water cycles due to land use, the specific phenology and seasonal CO2 fluxes of agricultural‐dominated areas, and the production of crops and grazing land. It uses 13 CFTs (11 arable crops and two managed grass types), with specific parameterizations of phenology connected to leaf area development. Carbon is allocated daily towards four carbon pools, one being the yield‐bearing storage organs. Management (irrigation, treatment of residues, intercropping) can be considered in order to capture their effect on productivity, on soil organic carbon and on carbon extracted from the ecosystem. For transient simulations for the 20th century, a global historical land use data set was developed, providing the annual cover fraction of the 13 CFTs, rain‐fed and/or irrigated, within 0.5° grid cells for the period 1901–2000, using published data on land use, crop distributions and irrigated areas. Several key results are compared with observations. The simulated spatial distribution of sowing dates for temperate cereals is comparable with the reported crop calendars. The simulated seasonal canopy development agrees better with satellite observations when actual cropland distribution is taken into account. Simulated yields for temperate cereals and maize compare well with FAO statistics. Monthly carbon fluxes measured at three agricultural sites also compare well with simulations. Global simulations indicate a ∼24% (respectively ∼10%) reduction in global vegetation (respectively soil) carbon due to agriculture, and 6–9 Pg C of yearly harvested biomass in the 1990s. In contrast to simulations of the potential natural vegetation showing the land biosphere to be an increasing carbon sink during the 20th century, LPJmL simulates a net carbon source until the 1970s (due to land use), and a small sink (mostly due to changing climate and CO2) after 1970. This is comparable with earlier LPJ simulations using a more simple land use scheme, and within the uncertainty range of estimates in the 1980s and 1990s. The fluxes attributed to land use change compare well with Houghton's estimates on the land use related fluxes until the 1970s, but then they begin to diverge, probably due to the different rates of deforestation considered. The simulated impacts of agriculture on the global water cycle for the 1990s are∼5% (respectively∼20%) reduction in transpiration (respectively interception), and∼44% increase in evaporation. Global runoff, which includes a simple irrigation scheme, is practically not affected.  相似文献

Sorghum is a promising alternative to maize for bioenergy production in Europe; however, its use is currently limited by poor adaptation to low temperatures during and after germination. We collected multi‐trait phenotype data under optimal and suboptimal temperatures in a genetically diverse recombinant inbred line (RIL) mapping population showing contrasting segregation patterns for pre‐ and post‐emergence chilling tolerance. Germination, emergence, seedling development, root architecture and seedling survival were assessed in two different seedlots. Emergence and root establishment were found to be the key determinants of development and survival under chilling stress. Highly interactive epistatic quantitative trait loci (QTL) hotspots, including a previously unknown QTL on Sb06 with a significant effect on prolonged chilling survival, were found to regulate different physiological mechanisms contributing to maintenance of growth and development despite the chilling temperatures. The major QTL regions harbour promising candidate genes with known roles in abiotic stress tolerance. Identification of loci in the QTL hotspot regions conferring maintenance of cell division and growth under early chilling stress represents a promising step towards breeding for successful establishment of sorghum in temperate climates.  相似文献

In 2009 the WPA President established a Task Force that was to examine available evidence about the stigmatization of psychiatry and psychiatrists and to make recommendations about action that national psychiatric societies and psychiatrists as professionals could do to reduce or prevent the stigmatization of their discipline as well as to prevent its nefarious consequences. This paper presents a summary of the Task Force’s findings and recommendations. The Task Force reviewed the literature concerning the image of psychiatry and psychiatrists in the media and the opinions about psychiatry and psychiatrists of the general public, of students of medicine, of health professionals other than psychiatrists and of persons with mental illness and their families. It also reviewed the evidence about the interventions that have been undertaken to combat stigma and consequent discrimination and made a series of recommendations to the national psychiatric societies and to individual psychiatrists. The Task Force laid emphasis on the formulation of best practices of psychiatry and their application in health services and on the revision of curricula for the training of health personnel. It also recommended that national psychiatric societies establish links with other professional associations, with organizations of patients and their relatives and with the media in order to approach the problems of stigma on a broad front. The Task Force also underlined the role that psychiatrists can play in the prevention of stigmatization of psychiatry, stressing the need to develop a respectful relationship with patients, to strictly observe ethical rules in the practice of psychiatry and to maintain professional competence.  相似文献

Differentiation among the European populations of the six-toothed spruce bark beetle Pityogenes chalcographus was observed for the first time in the 1970s as mating males from Northern Europe with females from Central Europe led to a significant decline in fecundity. Morphological examination revealed that P. chalcographus can be separated into two European races. Here, we investigated the genetic background of this separation by analysing 39 populations ( n  = 695), sequencing almost the complete cytochrome oxidase I gene (1543 bp) and applying single-strand conformation polymorphism. Phylogenetic analysis of 58 haplotypes yielded three major clades with a maximum sequence divergence of 2.33%, indicating that the demographic events took place in the late Pleistocene. These results support the hypothesis of allopatric divergence of the mtDNA lineages, which postglacially came into sympatric existence in Europe. Zygogramma bicolorata Pallister (Coleoptera: Chrysomelidae), a biological control agent of Parthenium hysterophorus L. (Asteraceae), was feeding on leaves of Helianthus annuus in India, raising concerns of its host range. Based on the age grading technique, it was shown that the majority of Z. bicolorata on sunflower plants at any time were reproductively immature. This establishes that Z. bicolorata does not pose any risk to sunflower in India.  相似文献

Four isoforms of GIRK channels (GIRK1–4) have been described in humans. In addition, several splice variants of more or less unknown function have been identified from several tissues and species. In our study, we investigated the structure and function of a new variant of GIRK1 that has been isolated from rat brain. Because of wide similarities with a previously described variant, we also named it GIRK1d. This variant lacks a region corresponding to exon 2 of full-length GIRK1, leading to a truncated GIRK1 that lacks the main part of the C-terminus. To study GIRK1d we used the Xenopus laevis expression system, the two-electrode voltage clamp method, and confocal laser scan microscopy. We found that our GIRK1d variant preferentially binds GIRK2 or GIRK4 over GIRK1. Furthermore, it largely reduces conductances mediated by GIRK1/2 or GIRK1/4 hetero-multimeric channels when coexpressed and nearly totally abolishes currents when replacing GIRK1 in hetero-multimeric channels.  相似文献

The focus of the article is on a generic approach to prediction of a healing process based on automatic measurement and modelling of mechanical disease indicators. As a specific example the healing process after treatment of advanced periodontitis is characterised and predicted based on measurement of tooth mobility (TM). For the measurement a new mechatronic instrument was developed that includes a pneumatically driven probe with incorporated sensors of tooth displacement and loading force. TM is measured by a computer controlled data acquisition system and predicted by a non-parametric regression. Similarly as in the characterisation of technical processes, a measured sample is described by TM time series detected on a particular patient, while the regression is determined by a set of samples obtained from a group of patients following periodontal therapy. The influence of surgical access therapy and systemic administration of metronidazole as an adjunct to the mechanical treatment is demonstrated. For this purpose the average changes of tooth mobility are determined on variously treated subgroups. A new method of healing process prediction is demonstrated and its performance is described by a prediction quality.  相似文献

In accordance with the peculiarities of the sun's azimuthal path near the equator, African honey bees observed in their night-time dances apparently allowed for the azimuth moving backwards at night relative to its day-time movement. In several cases the bees evidently inferred the actual azimuthal position of the sun, rather than the one that would result from extrapolating a uniform movement of the sun.  相似文献
