BAR superfamily domains shape membranes through poorly understood mechanisms. We solved structures of F-BAR modules bound to flat and curved bilayers using electron (cryo)microscopy. We show that membrane tubules form when F-BARs polymerize into helical coats that are held together by lateral and tip-to-tip interactions. On gel-state membranes or after mutation of residues along the lateral interaction surface, F-BARs adsorb onto bilayers via surfaces other than their concave face. We conclude that membrane binding is separable from membrane bending, and that imposition of the module's concave surface forces fluid-phase bilayers to bend locally. Furthermore, exposure of the domain's lateral interaction surface through a change in orientation serves as the crucial trigger for assembly of the helical coat and propagation of bilayer bending. The geometric constraints and sequential assembly of the helical lattice explain how F-BAR and classical BAR domains segregate into distinct microdomains, and provide insight into the spatial regulation of membrane invagination. 相似文献

Satellite-linked depth recorders (SDRs) were attached to 47 harbor seals in Prince William Sound, Alaska, during 1992–1996. Parameters describing diving effort, diving focus, and focal depth (depth bin to which diving was focused) were calculated from binned data on maximum dive depth and time spent at depth, and analyzed using repeated-measures mixed models. This analysis method accounted for individual variability, temporal autocorrelation, and the binned nature of SDR data, which are often ignored using standard statistical techniques. Results indicated that diving effort remained steady from September to April, when seals spent 68%-75% of their overall time in the water. Time spent in the water declined to 60% in May and to about 40% in July. Seals spent the most time in the water at night and the least in the morning. The diving of all seals in all months was highly focused. Overall, diving was focused to one depth bin approximately 75% of the time. Diving was more focused for females than for males and subadults. Focal dive depth was deepest in winter and shallowest during May-July. Focal depth and diving focus varied by region. Collinearity between month and region in the focal depth model suggests that seals move in winter to regions where prey are found deeper in the water column. Variations in diving behavior presumably result from combinations of regional bathymetry, seasonal cycles in type or depth distribution of prey, and seal life-cycle events such as reproduction and molting. 相似文献

The use of a backward (false) step to initiate forward movement has been regarded as an inferior starting technique and detrimental to sprinting performance over short distances as it requires additional time to be completed, but little evidence exists to support or refute this claim. Therefore, we recruited 27 men to examine the temporal differences among three standing starts that employed either a step forward (F) or a step backward (B) to initiate movement. An audio cue was used to mark the commencement of each start and to activate the subsequent timing gates. Three trials of each starting style were performed, and movement (0 m), 2.5 m, and 5 m times were recorded. Despite similar performances to the first timing gate (0.80 and 0.81 s for F and B, respectively), utilizing a step forward to initiate movement resulted in significantly slower sprint times to both 2.5 and 5 m (6.4% and 5.3%, respectively). Furthermore, when the movement times were removed and performances were compared between gates 1 and 2, and 2 and 3, all significant differences were seen before reaching a distance of only 2.5 m. The results from this investigation question the advocacy of removing the false step to improve an athlete's sprint performance over short distances. In fact, if the distance to be traveled is as little as 0.5 m in the forward direction, adopting a starting technique in which a step backward is employed may result in superior performance. 相似文献

The Mexican axolotl (Ambystoma mexicanum) provides a well-defined set of color genes which are useful for various types of analyses. These include the a (albino), m (melanoid), ax (axanthic), and d (white) genes. In addition, various combinations of these genes and a number of as yet undescribed mutants also exist. Three of these mutants (a, ax, and m) have defects associated with specific neural-crest-derived pigment cell types. The fourth mutant (d) appears to provide an unsuitable environment for the migration and maintenance of pigment cells. In one case (m), detailed information concerning the specific nature of the genetic defect is available. The goal of this article is to demonstrate ways in which the existing information on the axolotl color genes can best be utilized in terms of understanding not only the mutant phenotypes, but basic concepts in the cell and developmental biology of pigmentation as well. Thus, an attempt has been made to sort through the genetic and biochemical data relevant to these mutants in order to stimulate renewed interest in a more detailed pursuit of such studies. 相似文献

The hypoglycemic agent, 2-tetradecylglycidic acid (TDGA), administered in vivo lowered the concentration of plasma glucose and ketone bodies but raised the concentration of liver and plasma triglycerides in 10-day-old suckling rats. Phospholipid and cholesterol content of the plasma and liver were unaffected by drug treatment. TDGA inhibited the in vivo oxidation of [1-14C]palmitate but not that of [1-14C]decanoate. In suckling rat liver perfusion, TDGA totally inhibited ketone body formation from palmitate and depressed ketone body production from decanoate by 20%. Liver ATP and ADP content in the presence of TDGA decreased although this was probably a reflection of the increased triglyceride content of the liver since the was the same as control livers. The results are discussed in relation to the diet and to the inhibition of carnitine acyl transferase in suckling rats. 相似文献

Research was conducted in Bristol Bay, Alaska, to determine the applicability of radiotagging to studies of behavior, distribution and movements of belukha whales. Backpack-style VHF transmitters were attached to two belukhas by pinning through the dorsal ridge. Both packages were shed after about 2 wk due to migration of the pin through the tissue. Movements of radio-tagged whales were essentially local within Kvichak Bay. Three basic respiration patterns were identified: surfacings that were grouped into breathing periods separated by longer dives; surfacings that did not occur during restricted breathing periods; and long-to-very-long surfacings separated by short-to-very-short dives. These patterns were interpreted as representing traveling, feeding and feeding or resting in very shallow water. Surface and dive interval data were used to calculate a correction factor of 2.75, which could then be applied to aerial survey counts to estimate the total number of belukha whales in the study area.
