Background
Pterygium is a common ocular surface disease characterized by fibrovascular invasion of the cornea and is sight-threatening due to astigmatism, tear film disturbance, or occlusion of the visual axis. However, the mechanisms for formation and post-surgical recurrence of pterygium are not understood, and a valid animal model does not exist. Here, we investigated the possible mechanisms of pterygium pathogenesis and recurrence.Methods
First we performed a genome wide expression analysis (human Affymetrix Genechip, >22000 genes) with principal component analysis and clustering techniques, and validated expression of key molecules with PCR. The controls for this study were the un-involved conjunctival tissue of the same eye obtained during the surgical resection of the lesions. Interesting molecules were further investigated with immunohistochemistry, Western blots, and comparison with tear proteins from pterygium patients.Results
Principal component analysis in pterygium indicated a signature of matrix-related structural proteins, including fibronectin-1 (both splice-forms), collagen-1A2, keratin-12 and small proline rich protein-1. Immunofluorescence showed strong expression of keratin-6A in all layers, especially the superficial layers, of pterygium epithelium, but absent in the control, with up-regulation and nuclear accumulation of the cell adhesion molecule CD24 in the pterygium epithelium. Western blot shows increased protein expression of beta-microseminoprotein, a protein up-regulated in human cutaneous squamous cell carcinoma. Gene products of 22 up-regulated genes in pterygium have also been found by us in human tears using nano-electrospray-liquid chromatography/mass spectrometry after pterygium surgery. Recurrent disease was associated with up-regulation of sialophorin, a negative regulator of cell adhesion, and never in mitosis a-5, known to be involved in cell motility.Conclusion
Aberrant wound healing is therefore a key process in this disease, and strategies in wound remodeling may be appropriate in halting pterygium or its recurrence. For patients demonstrating a profile of 'recurrence', it may be necessary to manage as a poorer prognostic case and perhaps, more adjunctive treatment after resection of the primary lesion. 相似文献122.
In a 4-year study of Hamerkops in central Mali, territories of breeding pairs overlapped and most clutches were laid in the late rainy or early dry seasons. Clutches averaged 4–8 eggs. Different pairs laid eggs having, on average, different dimensions. Eggs laid in the late rainy and early dry seasons were larger than those laid at other times. Eggs weighed, at 27-8 g, about 6% of adult female weight. Hatching success was 79% with a fledging rate of eggs hatched of 53%. Successful nests fledged 27 young but for all completed clutches the fledging rate was 0'9. Young hatched early in the sequence had a higher survival rate than those hatched later. Stomatal conductances were measured in ageing leaves of Commelinacommunis L. as plants developed; stomatal responses to CO2 andabscisic acid (ABA) in epidermal strips of C. communis takenfrom ageing leaves of developing plants and in epidermal stripsfrom the same-aged leaves (the first fully-expanded leaf) ofdeveloping plants were also monitored. Stomatal behaviour wascorrelated with parallel measurements of photosynthesis andleaf ABA concentrations. Stomatal conductance in intact leavesdecreased from a maximum of 0-9 cm s 1 at full leaf expansionto zero about 30 d later when leaves were very senescent. Conductancesdeclined more slowly with age in unshaded leaves. Photosynthesisof leaf slices also declined with age from a maximum at fullleaf expansion until about 30 d later when no O2 exchange wasdetectable. Exogenously applied ABA (0.1 mol m 3) didnot affect respiration or photosynthesis. In epidermal stripstaken from ageing leaves the widest stomatal apertures occurredabout 10 d after full leaf expansion (just before floweringbegan) and then decreased with age; this decrease was less dramaticin unshaded leaves. The inhibitory effects of ABA on stomatalopening in epidermal strips decreased as leaves aged and wasgreater in the presence of CO2 than in its absence. When leaveswere almost fully-senescent stomata were still able to open.At this stage, guard cells remained healthy-looking with greenchloroplasts while mesophyll cells were senescing and theirchloroplasts were yellow. Similar data were obtained for stomatain epidermal strips taken from the same-aged leaves of ageingplants. The inhibitory effects of ABA on stomatal opening alsodecreased with plant age. In ageing leaves both free and conjugated ABA concentrationsremained low before increasing dramatically about 30 d afterfull leaf expansion when senescence was well advanced. Concentrationsof free and conjugated ABA remained similar to each other atall times. It is concluded that the restriction of stomatal movements inintact leaves as the leaves and plants age is due mainly toa fall in photosynthetic capacity of the leaves which affectsintracellular CO2 levels rather than to an inherent inabilityof the stomata to function normally. Since stomatal aperturein epidermal strips declines with plant and leaf age and stomatabecome less responsive to ABA (while endogenous leaf ABA levelsremain fairly constant until leaf senescence) it is suggestedthat some signal, other than ABA, is transmitted from the leafor other parts of the plant to the stomata and influences theirbehaviour. Although loss of genetic variation is frequently assumed to be associated with loss of adaptive potential, only few studies have examined adaptation in populations with little genetic variation. On the Swedish west coast, the northern fringe populations of the natterjack toad Bufo calamita inhabit an atypical habitat consisting of offshore rock islands. There are strong among‐population differences in the amount of neutral genetic variation, making this system suitable for studies on mechanisms of trait divergence along a gradient of within‐population genetic variation. In this study, we examined the mechanisms of population divergence using QST–FST comparisons and correlations between quantitative and neutral genetic variation. Our results suggest drift or weak stabilizing selection across the six populations included in this study, as indicated by low QST–FST values, lack of significant population × temperature interactions and lack of significant differences among the islands in breeding pond size. The six populations included in this study differed in both neutral and quantitative genetic variation. Also, the correlations between neutral and quantitative genetic variation tended to be positive, however, the relatively small number of populations prevents any strong conclusions based on these correlations. Contrary to the majority of QST–FST comparisons, our results suggest drift or weak stabilizing selection across the examined populations. Furthermore, the low heritability of fitness‐related traits may limit evolutionary responses in some of the populations. 相似文献

Endogenous levels of the polyamines putrescine, spermidine andspermine were quantified in the primary leaves of five cultivarsof bean (Phaseolus sp.) differing in their wilting responseto a chilling exposure of 5 ?C for 24 h. Levels of polyamines prior to chilling treatment did not appearto be correlated with chill-tolerance as levels in the non-chilledcontrols were highest in cultivars of medium chill-sensitivity.Plants grown under a vapour saturation deficit (VSD) of 8?4gm3 day/6?1 g m3 night exhibited a mild hardeningas compared to plants grown under a VSD of 5?7 gm3 day/4?1gm3 night, as the former showed less wilting on chilling.Hardening at high VSD had the effect of slightly lowering theputrescine content of non-chilled tissue but total polyaminecontent remained unchanged. However, on chilling, the largestrelative increase in polyamine levels, in particular that ofputrescine, occurred in hardened plants. There was also a significantrelative increase in putrescine titre in response to chillingin non-hardened genotypes of high chill-tolerance, whereas morechill-sensitive genotypes remained unchanged or slightly declinedin putrescine content on chilling. Relative changes in putrescine content rather than absolutelevels appears to be correlated with chill-tolerance. The effects of an auxin herbicide, 2,4-D, at a concentration of 0.01 mM, on the K+ uptake and efflux of excised roots of wheat (Triticum aestivum L. cv. Rannaya) were investigated at different pH values. The K+ movement was monitored with a K+ (86Rb) tracer. In parallel experiments the ATPase activities of microsomal fractions were determined by the inorganic phosphate liberation method. 2,4-D inhibited the K+ uptake especially at low pH, irrespective of whether Ca2+ was present or not. No marked changes were observed in the K+ efflux properties at pH values above 4. The inhibitory effect on K+ uptake exhibited a correlation with the hydrocarbon solubility of the herbicide, but not with the 2,4-D-induced decrease of the ATPase activity. It is suggested that 2,4-D exerts a non-specific effect on the lipid-protein interactions, giving rise to a generalized alteration of the transport barrier properties of the plasma membrane even at as low a concentration as 0.01 mM. 相似文献

Comparative studies of mammalian lysozymes and their genes have contributed to knowledge of how new functions arise during evolution. The recruitment of lysozymes for functioning in the stomach fluid of ruminants has occurred in response to selection pressures that are partly known and on a time-scale that is known. A semiquantitative analysis of adaptive evolution is thus made possible by the ruminant lysozyme system. Large-scale production of lysozyme by the stomach lining entailed gene duplication as well as a change in gene expression. Remoulding of the lysozyme for working and lasting in the stomach fluid involved accelerated amino acid replacements, which may have been facilitated by intergenic recombination. The possibility that multigene families can accelerate adaptive evolution, by virtue of their capacity for bringing together functionally coupled substitutions, receives emphasis in this review. 相似文献

Callus cultures have been initiated from stem explants of youngplants of Hevea brasiliensis and maintained over long periodsat 30 ?C by serial subculture in Murashige and Skoog mediumcontaining 2 mg 11 2,4-D and 0.5 mg 11 kinetin.Newly-initiated cultures spontaneously initiated roots but,on serial subculture, this property was lost and the culturesbecame heterogeneous (consisting of proliferating light segmentsand darker compact non-growing segments). Serially propagatedcultures continued to differentiate a few scattered latex vesselscontaining particulate material similar to that in the rootlaticifers. This callus (O callus) did not yield a growing cellsuspension when transferred to agitated liquid medium. However,the large cell aggregates which could be recovered after twopassages in liquid medium, when again grown on solid mediumyielded a highly friable light-coloured fast-growing homogeneouscallus (R callus) which retained its distinctive character onsubculture. This callus when transferred back to agitated liquidmedium yielded a fine rapidly growing cell suspension culturewhich could be serially propagated at 30 ?C in the same mediumas that used for callus culture. Both the O and R cultures were2,4-D dependent, but differed in their responses to 2,4-D. Bothretained their diploid character when serially propagated. Serially-propagatedsuspensions came to contain a proportion of polyploid cells.When the suspensions were maintained for several months withoutsubculture the larger cell aggregates which developed gave riseto embryo-like structures.
