Two pachyosteomorph arthrodires are described from the Upper Devonian of North America, Dunkleosteus raveri sp. nov. and Dunkleosteus amblyodoratus sp. nov. (from the Appalachian and the Michigan basins, respectively). Dunkleosteus raveri sp. nov. is found stratigraphically below the only other species within the Ohio Shale Formation (Famennian), and is characterized by the retention of a primitive dermal ornamentation, and shares with Dunkleosteus terrelli ( Newberry, 1873 ) the possession of articular facets on the parasphenoid. Dunkleosteus amblyodoratus sp. nov. (Kettle Point Formation, Upper Devonian), known only from incomplete nuchal, paranuchal, and parasphenoid plates, also retains dermal tuberculation, and is characterized by tapering articular facets on the parasphenoid. The current study reviews three North American species of Dunkleosteus (Du. terrelli, Du. raveri sp. nov. , and Du. amblyodoratus sp. nov. ). Formerly, two subgroups were recognized within the Dinichthyidae, the terrelli and herzeri groups, which are shown here to be members of distinct clades. Dinichthys herzeri Newberry, 1868 (the namesake member of the herzeri group) is now the sole member of the family Dinichthyidae comb. nov. , and is assigned to the Aspinothoracidi sensu Miles & Dennis, 1979 . Heintzichthys Whitley, 1933 and Gorgonichthys Claypole, 1892 (additional members of the herzeri group) were previously placed among the aspinothoracid arthrodires. Likewise, Holdenius holdeni Dunkle & Bungart, 1942 (the final member of the herzeri group) and Hadrosteus rapax Gross, 1932 are now placed within the Aspinothoracidi. This study investigated the interspecific differences in vulnerability to xylem embolism of four phreatophytes – two facultative phreatophytes ( Banksia attenuata and B. menziesii ) and two obligate phreatophytes ( B. ilicifolia and B. littoralis ). Species differences at the same position along an ecohydrological gradient on the Gnangara Groundwater Mound, Western Australia were determined in addition to intraspecific differences to water stress between populations in contrasting ecohydrological habitats. Stem- and leaf-specific hydraulic conductivity, as well as Huber values (ratio of stem to leaf area), were also determined to support these findings. We found that where water is readily accessible, there were no interspecific differences in vulnerability to water stress. In contrast both facultative phreatophyte species were more resistant to xylem embolism at the more xeric dune crest site than at the wetter bottom slope site. B. ilicifolia did not differ in vulnerability to embolism, supporting its classification as an obligate phreatophyte. Other measured hydraulic traits ( K S, K L and Huber value) showed no adaptive responses, although there was a tendency for plants at the wetter site to have higher K S and K L. This study highlights the influence site hydrological attributes can have on plant hydraulic architecture across species and environmental gradients.  相似文献

The endemic fauna of the Comoro Archipelago is composed of a mixture of taxa originating from Africa and Madagascar. Bats are the only native land dwelling mammals on this archipelago, but the biogeographical origins for the vast majority of species within this group are ambiguous. We report here genetic analyses based on two mitochondrial DNA markers to infer the origin of Comorian bats belonging to a reputed species complex of Miniopterus that is further distributed across Africa and Madagascar. Phylogenetic reconstructions show that east African M. minor are not closely related to the insular Miniopterus of Madagascar and the Comoros (Grande Comore and Anjouan). The latter cluster into two distinct, monophyletic clades (Clade 1 and Clade 2). Representatives of these clades occur sympatrically both on the Comoros and on Madagascar, and are distinguished by a large genetic distance (K2P: 9.9% for cytochrome b). No haplotypes are shared between any islands, suggesting the absence of contemporary gene flow. Populations of the widespread Clade 1 are furthermore characterized by a significant inter‐island structure (ΦCT = 0.249), and by high haplotype and nucleotide diversities (h = 0.90–0.98, π = 0.04–0.06). Demographic analyses of Clade 1 suggest secondary contact between two distinct phylogroups (Subclade 1 A and 1B) that reached Grande Comore and Anjouan, and a large, stable population with a long evolutionary history on Madagascar. These results and the current distribution of related lineages suggest that the Comoros were colonized independently at least two or three times by ancestors from Madagascar.  相似文献

Several house gecko species of the genus Hemidactylus are almost cosmopolitan lizards, with distributions that have probably been shaped by natural transoceanic dispersal as well as by more recent human introductions. Here we revise the Hemidactylus populations of Madagascar and compare them genetically with populations from other sites in the Indian Ocean region. Morphological data strongly confirm the occurrence of three Hemidactylus species on Madagascar: Hemidactylus frenatus , distributed along the western coast of Madagascar; H. platycephalus , restricted to the north-west and the widespread H. mercatorius that occurs throughout the island, including coastal areas at sea level as well as big cities (Antananarivo, Fianarantsoa) at altitudes of 1200–1300 m above sea level. Analyses of partial sequences of the 16S rRNA gene in 46 Hemidactylus specimens from Madagascar, East Africa, South Asia, and the Comoro and Mascarene archipelagos demonstrated the presence of a fourth species, H. brooki, on the Mascarenes (Réunion, Rodrigues, and Mauritius) and Comoros (Moheli). The Malagasy populations of H. platycephalus were genetically uniform and differentiated from the African and Comoroan specimens studied. H. frenatus had a relatively low genetic differentiation over the whole region with no recognizable phylogeographical structure, indicating more recent colonizations or introductions. In contrast, H. mercatorius showed a strong phylogeographical structure of haplotypes, with two distinctly different lineages in Madagascar. Moreover, all Malagasy specimens differed strongly from the single African specimen included. pPLA‐I is the evolutionarily oldest patatin‐related phospholipase A (pPLA) in plants, which have previously been implicated to function in auxin and defence signalling. Molecular and physiological analysis of two allelic null mutants for pPLA‐I [ppla‐I‐1 in Wassilewskija (Ws) and ppla‐I‐3 in Columbia (Col) ] revealed pPLA‐I functions in auxin and light signalling. The enzyme is localized in the cytosol and to membranes. After auxin application expression of early auxin‐induced genes is significantly slower compared with wild type and both alleles show a slower gravitropic response of hypocotyls, indicating compromised auxin signalling. Additionally, phytochrome‐modulated responses like abrogation of gravitropism, enhancement of phototropism and growth in far red‐enriched light are decreased in both alleles. While early flowering, root coils and delayed phototropism are only observed in the Ws mutant devoid of phyD, the light‐related phenotypes observed in both alleles point to an involvement of pPLA‐I in phytochrome signalling.  相似文献

Abstract Surveys of mating-system parameters in populations of the annual, self-compatible, tristylous, emergent aquatic, Eichhornia paniculata (Pontederiaceae) from N.E. Brazil and Jamaica have indicated that the species exhibits a wide range of outcrossing rates. To investigate whether temporal variation in outcrossing rate was also a feature of populations, open-pollinated families were sampled from five populations of contrasting style morph structure from N.E. Brazil over three consecutive years (1987–1989). Multilocus estimates of outcrossing rate ( t ) were obtained from assays of isozyme polymorphisms using starch gel electrophoresis. There was significant variation both among populations and between years in the frequency of outcrossing. Outcrossing in three tristylous populations was high ( t > 0.80), with relatively small fluctuations occurring over the three-year sampling period. In contrast, in a dimorphic and monomorphic population considerable self-fertilization occurred and the frequency of outcrossing declined significantly from 1987 to 1989 in both populations. In the dimorphic population, increased selfing was associated with a marked reduction in population size and an increase in the frequency of selfing variants of the mid-styled morph. The significance of temporal variation in outcrossing frequency in plant populations is discussed in relation to its effect on population genetic structure and recent models of mating-system evolution.  相似文献

Summary

We present the results of a variety of studies showing that activation of protein kinase C (PKC) in oocytes of Chaetopterus pergamentaceus results in germinal vesicle breakdown (GVBD). Phorbol esters and diacylglycerol can initiate a morphologically normal GVBD accompanied by a spectrum of associated biochemical processes, including increased protein phosphorylation, a shift in protein synthesis and activation of a protein kinase, maturation promoting factor (MPF). MPF activation is essential for GVBD in response to phorbol esters. In addition, inhibitors of PKC can block naturally-induced GVBD. We also present evidence that PKC can phosphorylate p34cde2, the catalytic subunit of MPF and that phosphorylation by PKC increases the histone H1 kinase activity of immunoprecipitated MPF. Immunoblot studies show that Chaetopterus oocyte p34cdc2 is not tyrosine phosphorylated prior to the initiation of GVBD, indicating that activation of MPF at GVBD in this species does not require p80cdc25, the activator of MPF at mitosis. These results suggest that PKC is an essential regulator of GVBD which can directly phosphorylate and regulate p34cdc2. Since PKC is the intracellular receptor for and is directly activated by tumor-promoters, tumor promotion might involve acceleration of the cell cycle through modification of the enzymatic activity of MPF by PKC.  相似文献

Background

Osteosarcoma (OS) is the most frequent pediatric malignant bone tumor. OS patients have not seen any major therapeutic progress in the last 30 years, in particular in the case of metastatic disease, which requires new therapeutic strategies. The pro-apoptotic cytokine Tumor necrosis factor (TNF)–Related Apoptosis Inducing Ligand (TRAIL) can selectively kill tumor cells while sparing normal cells, making it a promising therapeutic tool in several types of cancer. However, many OS cell lines appear resistant to recombinant human (rh)TRAIL-induced apoptosis. We, therefore, hypothesized that TRAIL presentation at the membrane level of carrier cells might overcome this resistance and trigger apoptosis.

Methods

To address this, human adipose mesenchymal stromal cells (MSCs) transfected in a stable manner to express membrane-bound full-length human TRAIL (mbTRAIL) were co-cultured with several human OS cell lines.

Results

This induced apoptosis by cell-to-cell contact even in cell lines initially resistant to rhTRAIL. In contrast, mbTRAIL delivered by MSCs was not able to counteract tumor progression in this OS orthotopic model.

Discussion

This was partly due to the fact that MSCs showed a potential to support tumor development. Moreover, the expression of mbTRAIL did not show caspase activation in adjacent tumor cells.  相似文献

A 2-year-old pine plantation was selected to receive treatments of fertilizers and herbicides to evaluate effects on Nantucket pine tip moth infestations and the tree growth parameters of height, diameter and volume increment. Nitrogen and phosphorus fertilizers, and hexazinone and sulfometuron methyl herbicides were used in creating six treatments: (i) control; (ii) phosphorus; (iii) nitrogen and phosphorus; (iv) phosphorus and herbicide; (v) nitrogen, phosphorus and herbicide; and (vi) herbicide. Treatments were applied in 1987 and 1988. In 1987, trees treated with nitrogen, phosphorus and herbicide had significantly greater height, diameter and volume growth than trees not receiving fertilizer treatments, but did not have significantly higher tip moth infestations than control trees. Treatments receiving phosphorus only had much lower tip moth infestation rates than other treatments except nitrogen and phosphorus. In 1988, tip moth infestations were uniformly low, with no differences in treatment effects observed.  相似文献
