SYNOPSIS. Nosema parkeri sp. n. is described from nymphs and adults of the argasid tick, Ornithodoros parkeri Cooley, from a laboratory colony. Schizogonic and sporogonic stages are described from various tick tissues. Spores are binucleate, measuring 3.2 (3–4) × 1.9 (1.8–2.5) μm. Transmission is transovarial and transstadial. The parasite does not appear to affect adversely the development or reproduction of the tick. Dermacentor andersoni Stiles was experimentally infected. Attempts to infect Swiss mice by tick feeding or by injection of infected tick suspensions were unsuccessful. The microsporidan differs in structure from Encephalitozoon ixodis Weiser) and Nosema slovaca Weiser & Reháček, the only other microsporidans known from ticks.  相似文献

The differentiation of Trypanosoma cruzi epimastigotes into trypomastigotes was studied in diffusion chambers sub-cutaneously implanted in mice. Using epimastigotes of the Tulahuén strain, transformation was first evident at 16 h after implantation and reached its maximum (92% trypomastigotes) by 24 h. Shortly before their differentiation into trypomastigotes, epimastigotes were found to develop resistance to lysis by the alternative pathway of complement. Furthermore, implantation of stationary-phase (as opposed to log-phase) parasites resulted in the accumulation of large numbers of complement-resistant epimastigotes in the chambers. These observations suggest that epimastigotes pass through a complement-resistant transitional stage before differentiating into trypomastigotes and that transformation may require cell division. In a further series of experiments, epimastigotes recovered 7 h after implantation in mice were found to differentiate into trypomastigotes when cultured in vitro for an additional 17 h at 37°C. This observation indicates that the events which trigger the morphologic transformation of epimastigotes into trypomastigotes can be dissociated operationally from the differentiation process itself.  相似文献

Experiments were conducted to investigate the effect of concentrationof NH4+ in nutrient solution on root assimilation of NO3and to determine whether the NH4+NO3 interaction wasmodified in the presence of K+. Dark-grown, detopped corn seedlings(cv. Pioneer 3369A) were exposed for 8 h to 0.15 mM Ca(NO3)2and varying concentrations of (NH4)2SO4 in the absence or presenceof 0.15 mM K2SO4. The accelerated phase of NO3 uptakeappeared most sensitive to restriction by additions of 0.15mM (NH4)2SO4. In the absence of K+, the restriction increasedonly slightly even when solution (NH4)2SO4, was increased from0.15 mM to 12.5 mM which was accompanied by an increase of NH4+in the tissue from about 7.0 to 35 µmol g–1 fr.wt. of root. Increasing concentrations of solution NH4+ progressivelyinhibited net K+ uptake. At the highest solution NH4+ concentrations,there was an initial net efflux of K+ and no net influx occurredduring the treatment period. The severity of the NH4)SO4 restrictionof NO3 uptake was moderated considerably in the presenceof K+ as long as a net influx of K+ occurred. However, net influxof K+ was not associated with alteration of NH4+ uptake, assimilation,or accumulation in the root tissue. The lack of correlationbetween the severity of restriction of NO3 uptake andendogenous NHJ suggested the restriction resulted from an effectexerted by exogenous NH4+ which tended to saturate at lowersolution NHJ concentrations or by inhibitory factors generatedduring assimilation of NH4+. Several mechanisms were postulatedto account for the moderating influence of K+. In all experiments,root NO3 reduction was restricted by the presence ofambient NH4+. The quantitative decreases in reduction tendedto be less than decreases in NO3 uptake and therefore,could result from inhibition solely of uptake with subsequentlimitation in availability of substrate for the reduction process,but the possibility of a direct effect on reduction could notbe excluded.  相似文献

The Emsian? through early Eifelian Onondaga Limestone of the Appalachian Basin was deposited in a topographic basin and on the carbonate platform which surrounded the basin on the west, north, and northeast. Onondaga strata thin from the platform into the basin. Two sedimentary cycles are present in the sub-Tioga Onondaga of eastern North America. The Edgecliff-Amherstburg represents an interval of transgression, in which epeiric seas spread over much of eastern North America. During the Nedrow-Lucas regression, the interior of the carbonate platform became restricted, resulting in the deposition of evaporites. The Moorehouse-Anderdon transgression continued through the deposition of the Tioga Bentonite, followed by the pre-Speeds-Dundee regression from the craton. Early Eifelian Appalachian Basin Onondaga brachiopod communities, arranged from nearshore to offshore, include the Atrypid-Megakozlowskiella, Atrypid-Levenea, Chonetid, Atlanticocoelia, Ambocoeliid, and Truncalosia Communities. The Onondaga-age Eastern Americas Realm is divided into the Appohimchi Province in the Appalachian Basin and the Michigan Basin-Hudson Bay Lowland Province in the Midwest. The provincial assignment of the James Bay region of Ontario is uncertain; the Eastern Townships of Quebec are near the boundaries both of the two provinces of the Eastern Americas Realm, and of the Eastern Americas Realm and the Old World Realm, the latter realm being probably in the Canadian Maritime Provinces.  相似文献

Organic requirements for attachment to glass, elongation, and motility of Entamoeba histolytica, have been determined. the trophozoite, which has been grown axenically only in highly complex media with reduced oxygen tensions, remains rounded and detached when placed in a Tris-HCl buffered solution containing NaCI, KCI, MgCI2, and CaCI2. A maintenance medium in which the amebae could attach to glass, elongate, and remain motile and viable for 12 to 24 h was devised with the addition of cysteine, ascorbic acid, bovine serum albumin, and the vitamin solution of medium NCTC #107. Tris-HCI was the most effective buffer tested and the optimal pH was 6.9 to 7.0. Survival, but not attachment, of the amebae was decreased at osmolalities ranging between 110 and 180 milliosmoles/kg, whereas both functions were decreased above ~260 milliosmoles/kg. Bovine serum albumin, the most effective of the proteins tested, and the vitamin solution helped maintain attachment of some ameba strains, but were not required by other strains. the requirements for cysteine and ascorbic acid were absolute and highly specific. During incubation in the maintenance medium, cell volumes decreased. Sensitivity of the organisms to agglutination by concanavalin A, wheat germ agglutinin, soybean agglutinin and fucose binding protein remained unchanged.  相似文献

Abstract. Modifications in plasma membrane structure and permeability were observed in Chlorella sorokiniana following exposure to 0.2 gm−3(140 p.p.m.) O3 for 30 min. Sixty-eight per cent of the cells were plasmolysed after 15 min O3 exposure with disruption of organelles similar to that previously described in higher plants. Freeze-fracture exposed large areas of plasma membrane in 90% of the control cells and those exposed to O3for short periods. After 20 min O3 90% of the cells cross-fracture, which indicates a change in molecular interactions in the membrane exposed to O3 The earliest observed ultraslructural alteration is an aggregation of particles on the plasma membrane P face, statistically significant after 10 min O3 Changes in 86Rb influx occur during a similar time. After more extended exposure to O3 the plasma membrane P face shows regions of lipid phase transition to the crystalline state.  相似文献

SYNOPSIS. A new design of flow vessel provides a method for continuous culture of P. falciparum in a settled layer of human erythrocytes with a slow flow of culture medium over them. The parasitemia is kept fluctuating from ? 1%, just after addition of fresh erythrocytes. to ? 10%, 2 or 3 days later. Wider attention to Fry's (1947) "final preferendum" paradigmwould facilitate comparative studies of temperature preference(behavioral thermoregulation) among different animal groups.According to Fry's bipartite definition, the final preferendumis that temperature at which preference and acclimation areequal, and to which an animal in a thermal gradient will finallygravitate regardless of its prior thermal experience (acclimation).This paradigm is helpful in distinguishing between acute thermalpreferenda (measured within 2 hr or less after placing an animalin a thermal gradient), which are influenced by acclimationtemperature, and the species-specific final preferendum (measured24–96 hr after placement in the gradient), which is essentiallyindependent of prior acclimation because reacclimation occursduring the gravitation process. The paradigm does not take intoaccount non-thermal acclimatization influences (e.g., season,photoperiod, age, light intensity, salinity, disease, nutrition,pollutants, biotic interactions) which can also affect temperaturepreference. However, a graph of acutely preferred temperaturesversus acclimation temperatures can be employed to determinean equivalent acclimation temperature for any given acclimatizationstate, as a simple means of quantifying acclimatization statesresulting from interactions of many influences. This paradigm,developed for use with fishes, can also be applied to otherectothermic taxa, although it is most easily employed with aquaticorganisms because of the simplicity of specifying aquatic thermalenvironments in terms of water temperature alone. Methodologiesused in studies of behavioral thermoregulation should take theparadigm into account (especially with respect to length oftests) to enhance the comparative value of data across taxa.  相似文献
