The study of placental transport is confounded by species differences, gestational differences, and a permanent non-steady state. The histology of the interhemal membrane, the degree of steric hindrance to diffusional exchange, the discrimination of electric charge, the flow pattern, and the transfer of intact gamma globulins are all species specific. We suggest the creation of order based on the ultrastructure of the placental barrier. The gas diffusion capacity of the membrane changes during gestation. This change is initially due to an increase in placental mass, but the gas diffusion capacity continues to increase after placental growth has ceased, because of changes in surface area and thickness of the membrane. The non-steady state of fetal weight is accompanied by an uptake of nutrients far in excess of what is necessary to satisfy oxygen consumption. Measurements of venous and arterial cord blood chemistry are sufficient to distinguish the fraction of nutrient uptake that is dedicated to catabolism from the fraction that is dedicated to fetal growth.  相似文献

The reassociation rates of repair replicated DNA of two human lymphoblastoid cell lines, the WIL2-A3 ‘normal’ line and the RAJI line of Burkitt's lymphoma, were examined using the DNA/DNA ‘C0t’ hybridization technique. The cells were treated with methyl methanesulfonate (MMS), an alkylating agent and mutagen, to induce the repair.The incorporated repair replication radioactivity in highly repetitive sequences of WIL2-A3 cell DNA reassociates as expected for a randomly distributed incorporation. The reassociation of repair radioactivity in sequences of fewer numbers of copies, however, is less than expected for a random distribution. It is less than that occurring for semiconservatively synthesized DNA of WIL2-A3 cells co-incubated with the repair labeled DNA as an internal control.The observed difference could be due to an over-representation of repair replication radioactivity in DNA sequences with fewer copies. It is unlikely to be due to residual alkali labile damage resulting from MMS treatment, since a similar difference was not observed when semiconservatively labeled DNA from cells which had been treated with MMS for the same time and at the same concentration as in the repair experiments was substituted for repair replicated DNA in the reassociation reactions. Other possible causes of the apparent difference in the reassociation rates observed are discussed.  相似文献

Tyzzer's disease was induced experimentally in nonimmunosuppressed, weanling Mystromys albicaudatus by oral inoculation with Bacillus piliformis spores. Focal areas of necrosis bordered by cells containing B. piliformis were observed first in the tunica muscularis of the intestine and in the periportal region of the liver 3 days post-inoculation, in the ventricular myocardium 7 days post-inoculation and in the brainstem 9 days post-inoculation. Healing in the tunica muscularis, liver and myocardium was accompanied by granuloma formation. The findings indicate that Mystromys are susceptible to B. piliformis infection. This is, to our knowledge, the first time brain lesions have been reported in any species following oral inoculation with B. piliformis. Tyzzer's disease should be considered as a possible diagnosis in Mystromys with hepatoenteritis, myocarditis, or indications of central nervous system disorders.  相似文献
