Endogenous pararetroviruses (EPRVs) represent a new class of dispersed repetitive DNA in plants. The genomes of many Nicotiana species and other solanaceous plants are rich in EPRVs. Distinct EPRV families are present in N. sylvestris ( Ns ) and in N. tomentosiformis ( Nto ), the two diploid progenitors of allotetraploid N. tabacum . Nicotiana EPRVs represent an interesting type of repetitive sequence to analyse in polyploids because of their potential impact on plant fitness and the epigenetic architecture of plant genomes. The Ns EPRV family appears identical in N. sylvestris and N. tabacum , indicating little change has occurred in either species since polyploid formation. By contrast, the Nto EPRV family is larger in N. tomentosiformis than in N. tabacum , suggesting either preferential elimination from the polyploid genome or specific accumulation in the diploid genome following polyploidization. The lability of Nto EPRVs might be enhanced by a frequent association with gypsy retrotransposons. Although some EPRVs are probably benign, others are potentially pathogenic or, conversely, determinants of virus resistance. Normally quiescent EPRVs can be reactivated and cause symptoms of infection in hybrids of species that differ in their EPRV content. EPRVs that furnish immunity to the free virus exemplify the selective value of so-called 'junk' DNA. The complete nucleotide sequences of two Suberites domuncula cDNAs and one Sycon raphanus cDNA, all encoding ubiquitin, have been determined. One cDNA from S. domuncula codes for polyubiquitin with four tandemly repeated monomeric units and the second cDNA encodes ubiquitin fused to a ribosomal protein of 78 amino acids (aa). S. domuncula possesses at least one additional polyubiquitin gene, from which the last two monomers were also sequenced. All analysed genes from S. domuncula encode identical ubiquitin proteins, with only one aa difference (Ala 19) to the human/higher animals ubiquitin (Pro 19). Ubiquitin in S. domuncula is identical with the ubiquitin found in another Demospongia, Geodia cydonium. The cDNA from S. raphanus encodes polyubiquitin with seven tandemly repeated units. All these gene monomers code for the same ubiquitin, which differs from the human/higher animals ubiquitin only at position 24 (Asp in Sycon, Glu in others). However, ubiquitin from S. raphanus (Calcarea) shows two aa differences (positions 19 and 24), when compared with the ubiquitin sequences from the two Demospongiae. In a phylogenetic tree constructed by multiple sequence alignment of all sponge ubiquitin gene monomers so far identified, all monomers from the same species cluster together, with the clear exception of the monomer from S. domuncula ribosomal protein fusion gene. This monomer branches off first from the tree and forms a separate line; this gives evidence for a very ancient split of ubiquitin-ribosomal-protein fusion genes from polyubiquitin encoding genes and their long separate coexistence in eukaryotes. The ubiquitin extension protein from S. domuncula is 78 aa long, displays all characteristics of 76–81 aa long ribosomal fusion proteins and shows 78% identity in the first 73 aa with the human S27a protein. However, its C-terminal sequence: 69-GLTYVYKKSD-78 is more similar to the plant consensus (69-GLTYVYQ/NK-76), than to the higher animal consensus (69-CLTYCFNK-76). This protein isolated from a sponge, belonging to the phylogenetically oldest multicellular animals, the Porifera, branches off first from the phylogenetic tree of metazoan ubiquitin extension proteins of the small ribosomal subunits.  相似文献

ABSTRACT. Developmental processes in multicellular organisms require structural elements, such as adhesion molecules, to stabilize cells at functional positions. In vertebrates, a series of extracellular matrix proteins, e.g. fibronectin and laminin are involved in cell adhesion. These proteins contain Arg-Gly-Asp [RGD] at their binding sites. Here we show that at concentrations above 2 mM the peptide GRG D SPK, comprising the tripeptide RGD (Arg-Gly-Asp), prevents the adhesiveness of cells of the marine amoeba Neopar-amoeba aestuarina. In addition, elevated levels of GRG D SPK cause cells to alter their shapes from those with digitiform subpseudopodia to rounded cells with small lobed pseudopodia. These cells detach from the substratum. These results are specific for the RGD sequence, because incubation in GRG E SPK solution at the same concentrations had no effect on cell attachment or structure. From these data we suggest that the structural adhesion molecules identified in vertebrates shows amino acid homologies with those found in unicellular protozoa.  相似文献

The in vitro and in vivo effects of copper, zinc, cadmium, nickel, cobalt, and manganese on nitrate reductase, malate dehydro-genase, isocitrate dehydrogenase, and glucose-6-phosphate dehydrogenase of zinc-, copper- and non-resistant populations of Silene cucubalus were investigated. During the in vitro experiments no resistant enzyme could be detected; enzymes of resistant and non-resistant ecotypes had a similar sensibility to all the metals. Nitrate reductase was the most sensitive enzyme. During the in vivo experiments remarkable differences were found. The nitrate reductase and the isocitrate dehydrogenase of the zinc-resistant population were activated when adding zinc to the culture medium, especially the nitrate reductase showed high activities at zinc concentrations where the nitrate reductase of the non-zinc-resistant populations was nearly completely inhibited. The zinc-resistant ecotype had a real need for zinc.  相似文献

Abstract. Propagules of the mangrove, Rhizophora mangle L., were precultivated for 9 months in a greenhouse. The young plants were transferred into unaerated nutrient solutions without and with 200 mol m 3 NaCl and subsequently their growth, their water relations and the photosynthetic properties of their leaves were studied. Growth of the salttreated plants was significantly increased, while the control plants gradually died off after finishing the experiments. The shoot water potential and the stomatal resistance of the leaves were lowered while the chlorophyll contents and the chlorophyll a/b ratio in the leaves of salt-treated plants were increased by NaCl, the net result being an enhanced rate of CO2 assimilation. The leaves of both sets of plants showed diurnal fluctuations in malic acid concentration which were more pronounced in the leaves of salt treated plants which, additionally, were more succulent. However, the plants showed no net CO2 fixation at night, indicating that Rhizophora mangle is a CAM-cycling plant. After 200 d of cultivation without or with NaCl, the Na+, Cl and K+ concentrations in tissues and vacuoles were measured. Energy-dispersive X-ray microprobe analyses on root vacuoles of control plants reveal Na+ preference, on those of salt treated plants a strong K+ preference. Vacuolar K+ concentrations are neither affected by NaCl nor do they vary across the root radius. High vacuolar Na+ and Cl concentrations are found in the hypodermis followed by a stepwise decrease towards the inner root cortex cells. Ion concentrations of the photosynthetically active leaf tissues seem to be regulated by (1) radial filtration across the root cortex: (2) ion exchange of the xlem parenchyma cells: and (3) sequestration of Na+ and Cl in the hypodermal water storage tissue of the leaves.  相似文献

The occurrence of striped colour patterns and of striped/non-striped polymorphism systems among snakes is reviewed from literature data augmented by some personal observations. Among 1367 species, 190 were striped or had striped morphs. Of 11 families, the striped pattern was common mainly among Colubridae, presumably in relation to the active escape behaviour strategy, prevalent in this family. The striped species tended to cluster in a small number of genera. The 40 striped/non-striped polymorphism systems found, fall into five categories, according to the coloration patterns of the alternative morphs: (I) blotched (cryptic); (2) barred (or ringed); (3) plain; (4) melanistic; (5) albinistic. Most polymorphisms are presumably maintained by eco-behavioural trade-offs, depending on the category and on the habitat: The striped morph is presumed more effective in active escape and sometimes also in camouflage; the alternative morph may be more effective in camouflage, in active escape or in thermoregulation. Hence morph frequency depends on the habitat. Striped-albinistic polymorphism in Elaphe climacophora presumably depends on human protection of the albino morph.  相似文献
