Systematic conservation planning efforts typically focus on protecting current patterns of biodiversity. Climate change is poised to shift species distributions, reshuffle communities, and alter ecosystem functioning. In such a dynamic environment, lands selected to protect today's biodiversity may fail to do so in the future. One proposed approach to designing reserve networks that are robust to climate change involves protecting the diversity of abiotic conditions that in part determine species distributions and ecological processes. A set of abiotically diverse areas will likely support a diversity of ecological systems both today and into the future, although those two sets of systems might be dramatically different. Here, we demonstrate a conservation planning approach based on representing unique combinations of abiotic factors. We prioritize sites that represent the diversity of soils, topographies, and current climates of the Columbia Plateau. We then compare these sites to sites prioritized to protect current biodiversity. This comparison highlights places that are important for protecting both today's biodiversity and the diversity of abiotic factors that will likely determine biodiversity patterns in the future. It also highlights places where a reserve network designed solely to protect today's biodiversity would fail to capture the diversity of abiotic conditions and where such a network could be augmented to be more robust to climate-change impacts. 相似文献

RBM5/LUCA-15/H37 is a nuclear SR-related RNA binding protein with the ability to modulate both apoptosis and the cell cycle, and retard tumour formation. How RBM5 functions to carry out these, potentially interrelated, biological activities is unknown. Since reversible phosphorylation has been shown to play an important role in the regulation of SR protein function, apoptosis and cell cycle control, in an attempt to elucidate the underlying mechanisms regulating RBM5 function, the phosphorylation status of RBM5 was investigated. Whole cell lysate from growing cell cultures was treated with the broad phosphatase spectrum of CIP, resulting in a decrease in the molecular mass of RBM5. A similar decrease in molecular mass, of a subset of RBM5 proteins, was observed during growth factor deprivation, in a manner consistent with partial dephosphorylation of RBM5. Molecular mass increased upon growth factor addition, demonstrating that this apoptosis-associated alteration in molecular mass was a reversible process. Immunoprecipitation and mutagenesis experiments strongly suggested that phosphotyrosines are not present in RBM5 under normal growth conditions, and that serine 69 is phosphorylated, but not by Akt kinase. Taken together, these results suggest that reversible phosphorylation of RBM5 is a mechanism capable of regulating RBM5 participation in modulating apoptosis, and perhaps tumour suppression. 相似文献

Lake biodiversity is an incomplete indicator of exogenous forcing insofar as it ignores underlying deformations of community structure. Here, we seek a proxy for deformation in a network of diatom assemblages comprising 452 species in 273 lakes across China. We test predictions from network theory that nodes of similar type will tend to self‐organize in an unstressed system to a positively skewed frequency distribution of nodal degree. The empirical data reveal shifts in the frequency distributions of species associations across regions, from positive skew in lakes in west China with a history of low human impacts, to predominantly negative skew amongst lakes in highly disturbed regions in east China. Skew values relate strongly to nutrient loading from agricultural activity and urbanization, as measured by total phosphorus in lake water. Reconstructions through time show that positive skew reduces with temporal intensification of human impacts in the lake and surrounding catchments, and rises as lakes recover from disturbance. Our study illustrates how network parameters can track the loss of aquatic assemblage structure in lakes associated with human pressures. 相似文献

Hypotheses for explaining plant invasions have focused on a variety of factors that may influence invasion success, including propagule pressure, interactions of the introduced species with the biotic, abiotic, or disturbance properties of the new ecosystem, or the genetic characteristics of the invader itself. Evaluating the relative importance of these factors has been difficult because for most invaders key information about the introduced population or the introduction event is not available. We propose that natural experiments using model species is an important tool to test multiple invasion hypotheses at the same time, providing a complementary approach to meta-analysis and literature review. By focusing on a single candidate species, Pinus contorta, we explore several attributes that we propose constitute a good model, including: (a) intentional and relatively well documented introduction into a wide range of environments and countries across the world during the past century, where invasion success or failure has already occurred, (b) conspicuous growth form that simplifies assessment of growth rates, and comparisons across native and introduced ecosystems around the world, and, (c) documented and replicated variability of introduction intensity, genetic characteristics of the introduced populations, contrasting biotic communities present at sites of introduction, and abiotic conditions within and across introduced ecosystems. We propose that identifying model species with these characteristics will provide opportunities to disentangle the relative importance of different mechanisms hypothesized to influence invasion success, and thereby advance the field of invasion ecology. 相似文献

The adapter protein ADAP (FYB/SLAP-130) provides a critical link between T cell receptor (TCR) signaling and cell adhesion via the activation of integrins. The C-terminal 70 residues of ADAP show homology to SH3 domains; however, conserved residues of the fold are absent. An alignment and annotation of this domain has therefore been elusive. We have solved the three-dimensional structure of the ADAP C-terminal domain by NMR spectroscopy and show that it represents an altered SH3 domain fold. An N-terminal, amphipathic helix makes extensive contacts to residues of the regular SH3 domain fold, and thereby a composite surface with unusual surface properties is created. We propose this SH3 domain variant to be classified as a helically extended SH3 domain (hSH3 domain) and show that the ADAP-hSH3 domain can no longer bind conventional proline-rich peptides. 相似文献

E3 ubiquitin ligase Cbl-b plays a crucial role in T cell activation and tolerance induction. However, the molecular mechanism by which Cbl-b inhibits T cell activation remains unclear. Here, we report that Cbl-b does not inhibit PI3K but rather suppresses TCR/CD28-induced inactivation of Pten. The elevated Akt activity in Cbl-b(-/-) T cells is therefore due to heightened Pten inactivation. Suppression of Pten inactivation in T cells by Cbl-b is achieved by impeding the association of Pten with Nedd4, which targets Pten K13 for K63-linked polyubiquitination. Consistent with this finding, introducing Nedd4 deficiency into Cbl-b(-/-) mice abrogates hyper-T cell responses caused by the loss of Cbl-b. Hence, our data demonstrate that Cbl-b inhibits T cell activation by suppressing Pten inactivation independently of its ubiquitin ligase activity. 相似文献

Simultaneous fluorescence in situ hybridisation with 5S and 25S rDNA probes enables the discrimination of a substantial number
of chromosomes of the complement of all diploid and tetraploid Brassica species of the ”U-triangle”, and provides new chromosomal landmarks for the identification of some chromosomes of this genus
which were hitherto indistinguishable. Twelve out of 20 chromosomes can be easily identified in diploid Brassica campestris (AA genome), eight out of 16 in Brassica nigra (BB genome), and six out of 18 in Brassica oleracea (CC genome). Furthermore, just two rDNA markers permit 20 out of 36 chromosomes to be distinguished and assigned to either
the A or B genomes of the allotetraploid Brassica juncea, and 18 out of 38 chromosomes identified and assigned to the A or C genomes of the allotetraploid Brassica napus. The number of chromosomes bearing rDNA sites in the tetraploids is not in all cases simply the sum of the numbers of sites
in their diploid ancestors. This observation is discussed in terms of the phylogeny and variability within the genomes of
the species of this group.

There is no single way to represent a task. Indeed, despite experiencing the same task events and contingencies, different subjects may form distinct task representations. As experimenters, we often assume that subjects represent the task as we envision it. However, such a representation cannot be taken for granted, especially in animal experiments where we cannot deliver explicit instruction regarding the structure of the task. Here, we tested how rats represent an odor-guided choice task in which two odor cues indicated which of two responses would lead to reward, whereas a third odor indicated free choice among the two responses. A parsimonious task representation would allow animals to learn from the forced trials what is the better option to choose in the free-choice trials. However, animals may not necessarily generalize across odors in this way. We fit reinforcement-learning models that use different task representations to trial-by-trial choice behavior of individual rats performing this task, and quantified the degree to which each animal used the more parsimonious representation, generalizing across trial types. Model comparison revealed that most rats did not acquire this representation despite extensive experience. Our results demonstrate the importance of formally testing possible task representations that can afford the observed behavior, rather than assuming that animals’ task representations abide by the generative task structure that governs the experimental design. 相似文献
