Monoclonal antibodies (MAbs) were raised against surface antigens from Pythium sulcatum. The immunogens were prepared from salt extractable cell wall protein to produce monoclonal antibodies. The MAbs showed high specificity to seven P. sulcatum isolates among 26 species of soil‐borne fungi. Weak cross‐reactivities were observed with Pythium aristosporum, Pythium myriotylum, and Pythium zingiberum in indirect enzyme‐linked immunosorbent assay (ELISA), but no reaction was obtained in Western blot analysis. The MAbs recognized glycoproteins in cell wall. Pythium sulcatum was detected in naturally infected carrot tissues and soil using indirect competition ELISA.  相似文献

We investigated the hybrid origin of × Crepidiastrixeris denticulato-platyphylla using RAPDs and ITS sequence data. The putative parents Paraixeris denticulata and Crepidiastrum platyphyllum represent separate species, irrespective of geographical origin. The occurrence of species specific RAPD markers from P. denticulata and C. platyphyllum in × C. denticulato-platyphylla established unambiguously a hybrid origin between the two taxa. This was in line with the occurrence of a combination of morphological characters such as plant habit and floret numbers. The parent taxa differed from each other by 7 nucleotide substitutions and 2 indel events in the ITS region. The hybrids showed sequence additivity and most likely represent F1 plants, with the exception of two plants which were of possible F2 origin, possessing either the ITS sequences of one parent only, or one predominant ITS type. The hybrids occurred in two out of three localities where the parents occurred sympatrically. This fact, together with the short life-span of the plants, suggests that × C. denticulato-platyphylla exists as a result of repeated, frequent hybridization between the parent species.  © 2006 The Linnean Society of London, Botanical Journal of the Linnean Society , 2006, 151 , 333–343.  相似文献

1. Detecting hot spots of litter decomposition will promote understanding of litter processing in a heterogeneous system. To identify hot spots of leaf breakdown within a headwater stream reach, we examined the difference in leaf breakdown rate among four types of litter patches, one that formed in riffles and three that formed in pools (middle, alcove, edge), in different seasons. 2. Middle patches showed the highest breakdown rate in some seasons; the rate in middle patches was 1.5–4 times higher than in the other patches. Thus middle patches can be regarded as hot spots of leaf breakdown in the study reach. This result contrasted with other studies showing higher breakdown rate in riffles than in pools. 3. Significant relationships between abundance of caddisfly shredders and breakdown rate were observed in seasons when the rate differed among patch types. Greater abundance of Lepidostoma seems to be responsible for middle patches being hot spots of leaf breakdown. 4. It is expected that when the proportion of leaves retained in middle patches within a reach is higher, the breakdown rate of the entire reach will be increased. Clarifying how the proportion of leaves retained on middle patches within a reach varies temporally and spatially would improve our understanding of leaf breakdown in headwater streams.  相似文献

In the last two decades, the relationship between diversity and stability/ecosystem functioning has been widely discussed and has become a central issue in ecology. Here, we assessed the relationship between wetland plant diversity and community resilience after a disturbance. Our study area was located in the Upper Paraná River floodplain (Brazil). An experiment was carried out in situ (18 1 m × 1 m plots with richness varying from 1 to 18 species). In each plot, we recorded the number of species, total per cent vegetation cover and per cent age cover of each species. The above‐ground biomass of wetland plants was removed, simulating a disturbance by animal trampling or an extreme flood. The recovery of vegetation was monitored over 3 months. According to a linear regression, the recovery of wetland plants was positively correlated with diversity. Comparisons with plots containing monocultures of one of the dominant species (Polygonum stelligerum) suggested that this species did not overyield in mixed cultures. Thus, our experiments indicate that the higher resilience in richer plots after a disturbance is mainly due to the fact that species have different resource use requirements (complementarity effect) and not due to the presence of a single, more productive species. Our experiment carried out in a more real condition (in situ) showed that biodiversity is important to wetland functioning and stability, paralleling the results obtained in laboratory and mesocosms experiments. These results also suggest that the loss of plant diversity in our study area could compromise community recovery following strong disturbances.  相似文献

Peanut/maize intercropping is a sustainable and effective agroecosystem that evidently enhances the Fe nutrition of peanuts in calcareous soils. So far, the mechanism involved in this process has not been elucidated. In this study, we unravel the effects of phytosiderophores in improving Fe nutrition of intercropped peanuts in peanut/maize intercropping. The maize ys3 mutant, which cannot release phytosiderophores, did not improve Fe nutrition of peanut, whereas the maize ys1 mutant, which can release phytosiderophores, prevented Fe deficiency, indicating an important role of phytosiderophores in improving the Fe nutrition of intercropped peanut. Hydroponic experiments were performed to simplify the intercropping system, which revealed that phytosiderophores released by Fe‐deficient wheat promoted Fe acquisition in nearby peanuts and thus improved their Fe nutrition. Moreover, the phytosiderophore deoxymugineic acid (DMA) was detected in the roots of intercropped peanuts. The yellow stripe1‐like (YSL) family of genes, which are homologous to maize yellow stripe 1 (ZmYS1), were identified in peanut roots. Further characterization indicated that among five AhYSL genes, AhYSL1, which was localized in the epidermis of peanut roots, transported Fe(III)–DMA. These results imply that in alkaline soil, Fe(III)–DMA dissolved by maize might be absorbed directly by neighbouring peanuts in the peanut/maize intercropping system.  相似文献

柞蚕是一种野外饲养的经济昆虫,主要分布在我国东北部山区。柞蚕以蛹滞育越冬,其蚕蛹个大、容易固定、保存时间长、无须饲养、容易运输等优点。利用柞蚕蛹作为生物反应器生产外来蛋白质,可以大规模机械化生产,减少操作上的烦琐和劳动力。本文利用柞蚕核型多角体病毒(AnpeNPV)作为基因表达载体,在柞蚕培养细胞(AnPe细胞)和柞蚕蛹中成功地表达了β-半乳糖苷酶基因(LacZ),并利用AnPe细胞筛选、纯化获得了AnpeLacZ重组病毒。该重组病毒的β-半乳糖苷酶产量,在TC-100(含10%FBS)培养的AnPe细胞中最高酶活性为感染后第12天的40.9 units/ml,在SF-900Ⅱ培养的AnPe细胞中最高酶活性为感染后第18天的59.9 units/ml,后者表达量稍高,但时间滞后。AnpeLacZ在5℃保存了7个月的柞蚕蛹中,感染后第15天酶活性达到最高,雌蛹14.3 units/g,雄蛹11.7 units/g,雌蛹比雄蛹略高。结果显示,柞蚕核型多角体病毒和柞蚕蛹可以作为一个可以机械化大规模生产的新型杆状病毒基因表达载体系统开发和利用。  相似文献

The nuclear DNA contents of atypical mesothelial cells from five patients who had an eosinophilic pleural effusion (EPE) were studied by the use of DAPI (4′,6-diamidino-2-phenylindole dihydrochloride) DNA staining. Analysis of the nuclear DNA content revealed a polyploid pattern, with a major peak in the tetraploid region. Using an immunocytochemical technique, the atypical mesothelial cells showed a positive reaction for cytokeratin. In contrast carcinoembryonic antigen (CEA) was always negative in these cells. It is suggested that the atypical mesothelial cells with EPE had a higher rate of proliferation than did the normal mesothelial cells.  相似文献
