ABSTRACT. We developed a method to study the DNA synthetic cycles of Entamoeba histolytica and Entamoeba invadens by flow cytometry (FCM) based on a preparative procedure to reduce both high levels of natural fluorescence and non-specific adsorption of fluorochromes. We modeled G1, S, and G2 phases as a series of overlapping Gaussian curves. Both E. histolytica and E. invadens displayed G1, S, and G2 proportions that are consistent with eukaryotic cell populations in exponential or stationary growth phase. Exponential phase E. histolytica populations contained a hypodiploid subset with a mass of about 20% less than the diploid value which we estimate by FCM to be 24 × 10-14 g DNA/cell. Exponential phase E. invadens populations contained a hypodiploid subset with a mass of about 6% less than the diploid value which we estimate by FCM to be 30 × 10-14 g DNA/cell.  相似文献

A new species of bucculaticid moth, Bucculatrix hamaboella sp. nov. (Host plant: Hibiscus hamabo , Malvaceae) is described from Wakayama Prefecture, Japan. The feeding habit of the new species is unique in that: (i) the young larva is a leaf miner forming a long red linear mine but in the later instars the larva becomes a stem borer; (ii) later instar larvae undergo double molts within a cocoonet (molting cocoon); and (iii) penultimate and final instars appear on the surface of the leaf as non-feeding stages. The external non-feeding larvae of B. hamaboella undergoing double molts within one cocoonet are considered to be an abbreviated form of the external feeding instars of other bucculatricids typically making first and second cocoonets, undergoing a single molt within each cocoonet. On the basis of morphological characters, this species is related to the species of Sections I and II (Host: Asteraceae) of Braun (1963), rather than to the species of Section VIII (Host: Malvaceae).  相似文献

The present study describes the morphology of the sternum V gland of three caddisfly species, Stenopsyche marmorata Navas, Eubasilissa regina (McLachlan) and Nemotaulius admorsus (McLachlan), each of which belongs to a different family of the order Trichoptera, using light and scanning electron microscopy. In both sexes of these three species, the gland orifices are located on the sides of the sternum V as crescent-shaped slits, and are connected with the glandular tissue via cuticular gland ducts. The shapes of glands differ greatly among species; a slender ampullar form in S. marmorata , a flattened saccular form (horseshoe shape) in E. regina and a kidney shape in N. admorsus . The glands are composed of four essential components: large secretory cells, small reservoir cells, the lining of the reservoir and the gland duct. In S. marmorata and E. regina , additional components, muscle fibers, are present around the small reservoir cells. The secretory cells covering the whole outer surface of the gland are very large, and form many bunches in S. marmorata and E. regina , but do not form them in N. admorsus . The small reservoir cells lie inside the layer of the secretory cells and are tightly connected with the cuticular lining of the reservoir. The linings become thick cuticular ducts near the gland orifices. Histological features suggest that the secretory cells of the sternum V gland of Trichoptera belong to the type of class 3 cells in insect epidermal glands.  相似文献

Tyrosinase-related protein 1 (TRP1) maps to the brown locus in mice. Although the specific function of TRP1 has been in dispute, mutations in its structural gene result in the formation of brown rather than black melanin. We have investigated the melanogenic function of TRP1 by using immune-affinity purification of the protein and also by using transfection of its gene into fibroblasts to study its characteristics. We show that TRP1 has the ability to oxidize DHICA, a melanogenic intermediate derived from DOPAchrome. In addition, TRP1 has the ability to interact with tyrosinase and significantly stabilize the latter's catalytic function.  相似文献

Analyses of papers published in General and Comparative Endocrinologyfrom 1961 to 1981 were made from several aspects. Number ofpapers published has increased year after year. Molluscan endocrinologypapers have increased and overtaken annelid publications. Duringthe past 20 yr studies using crustaceans and insects have comprised25–50% of all endocrine research on invertebrate groups.Papers on protochordates, elasmobranchs, and cyclostomes arefew but constant in number, indicating that these three groupsare important from phylogenic aspects. Studies on birds andteleosts have increased, while those on amphibians have decreased.Biochemical techniques were employed in 50% of all publicationsin this journal. Radioimmunoassay has replaced autoradiography.Immunohistochemistry increased in popularity. Studies on thepars distalis and gonads are predominant among all endocrineorgans. Research at the molecular level is decreasing, but isincreasing at the organismal level. Research at the organismallevel may be characteristic of comparative endocrinology. Largelythrough the initiative of Aubrey Gorbman, General and ComparativeEndocrinology was first published in 1961, and the InternationalCommittee of the Symposium on Comparative Endocrinology wasformed in 1957. Gorbman has produced many students. His originaland important contributions are in diversified areas: thyroidphysiology and its evolution, hormones and the brain and behavior,cyclostome endocrinology and neurosecretory systems in lowervertebrates. In addition, he has influenced investigators throughhis ideas of evolution. He has had particular impact on thedevelopment of comparative endocrinology in Japan.  相似文献

There are only a few reports on the relative biological effectiveness (RBE) of thermal neutrons and 10B(n,α)7Li reactions either in vitro or in vivo. The data in this paper summarize almost all previously published in vitro data. Because only a few reactors are available for biomedical purposes, it is difficult to make a comparison of data from experiments using the same kind of radiation, and also to make a comparison of data from experiments using the different kinds of radiations. However, it is indispensable for boron neutron capture therapy to make a radiobiological analysis. More intensive study, including repair process and oxygen effect, is necessary for establishing the fundamental basis of the clinical application of boron neutron capture therapy.  相似文献
