Liposome-mediated delivery of DNA to carrot protoplasts 总被引:1,自引:0,他引:1
The encapsulation of DNA within liposomes and subsequent fusion of the liposomes with carrot (Daucus carota L.) protoplasts were examined to determine optimum conditions for effective liposome-mediated delivery of DNA to protoplasts. Escherichia coli [3H]DNA could be encapsulated with 50% efficiency using encapsulation volumes as low as 0.5 ml. Incorporation of liposome-encapsulated [3H]DNA by carrot protoplasts increased linearly for 2.5 h, and increasing the ratio of protoplasts to liposomes increased the total amount of radioactive label incorporated within the protoplasts. Liposome-mediated incorporation of [3H]DNA by protoplasts increased over a range of polyethylene glycol concentrations up to 20%, but Ca2+ did not increase liposome-mediated incorporation when present in the liposome-protoplast incubation mixture. Optimum incorporation was observed when the pH of the liposome-protoplast incubation medium was decreased to 4.8. Encapsulation experiments using DNA of the plasmid pBR322 indicated that an average of 200–1,000 intact copies of pBR322 were sequestered within each nucleus after liposome delivery. 相似文献

Summary Computer simulation was used to compare Hallauer's cyclic single cross selection (CSCS) with reciprocal recurrent selection (RRS). Three epistatic and three non-epistatic models with 60 loci determining a single character provided the genetic base. The rate of advance over seven cycles was always greater for CSCS than for RRS on either a cycle or generation basis. No genetic conditions were found where CSCS failed to respond. The advantages of CSCS increases as the proportion of non-additive genetic variance increases. Genetic advance of the hybrid population was shown to result from the joint effects of an average change in gene frequency and complementary effects (nicking) of selection. Nicking effects accounted for most of the advance for some starting conditions. RRS generally had higher selection limits with no epistasis or low gene frequency of the dominant allele. CSCS generally had higher limits with epistasis or high frequency of the dominant allele. We suggested that CSCS begin with divergent genetic populations and strong selection intensity for three cycles. The plasmalemma and hyaline ectoplasm together constitute the sensory and motor organ of macrophages. The purpose of this study was to isolate this cell fraction in order to analyze it biochemically and functionally. Brief sonification of warmed rabbit lung macrophages caused release of heterodisperse hyaline blebs and filopodia, which were easily collected by differential centrifugation. Viewed in the electron microscope, these structures consisted of membrane-bounded sacs principally containing actin filaments. Some contained secondary lysosomes. They were enriched threefold over whole cell homogenates in specific adenylate cyclase activity and in trichloroacetic-acid-precipitable (125)I when derived from cells labeled with 125(I) by means of a lactoperoxidase-catalyzed reaction. These markers were found to have identical isopycnic densitites when macrophage homogenates were subjected to sedimentation in a focusing sucrose density gradient system, and these markers had densities distinct from those of other cytoplasmic organelles. These markers were therefore assumed to be associated with macrophage plasma membranes. The specific β- glucuronidase activity of the bleb fraction was similar to that of homogenates, but the blebs had considerably lower specific succinic dehydrogenase activity and RNA content, and DNA was undetectable. Electrophoresis of blebs solubilized in sodium dodecyl sulfate on polyacrylamide gels revealed polypeptides co-migrating with macrophage actin-binding protein, myosin, and actin; blebs also had EDTA-activated adenosine triphosphatase activity characteristic of myosin. The concentrations of actin-binding protein and myosin were higher in blebs than in cells or cytoplasmic extracts, whereas actin concentrations were similar (relative to extracts) or only slightly greater (than in cells). Blebs and intact cells had high lactate dehydrogenase activities in the presence but not the absence of Triton X-100. Blebs and cells oxidased 1-[(14)C]glucose, and the rate of glucose oxidation was increased substantially in the presence of latex beads. We conclude that intact sacs of plasmalemma encasing contractile proteins and cytoplasmic enzymes can be isolated from macrophages. They are enriched in myosin and actin-binding protein, indicating that the contractile apparatus is regulated in the cell periphery. These structures have the capacity to respond to environmental signals. We suggest the name "podosomes" for them because of their resemblance to macrophage pseudopodia. We propose that podosome formation results from rapid dissolution of the cortical gel when the membrane is in an actively extended configuration. 相似文献

The present study focused on whether it is possible to expand monocytic cells from CD34+ progenitor cells by using macrophage colony-stimulating factor (M-CSF) in the absence and presence of mast cell growth factor
(MGF) and IL-6. It was demonstrated that CD34+ cells differentiate without expansion to functional mature monocytic cells in the presence of M-CSF or combinations of M-CSF
plus IL-6 and MGF. A different response pattern was observed for the number of clonogenic cells. The addition of IL-6 or both
IL-6 and MGF to M-CSF containing cultures resulted in significant higher numbers of colony-forming unit-macrophage (CFU-M)
as tested in clonogenic and3H-thymidine assays. Furthermore, M-CSF plus both IL-6 and MGF appeared to be the most potent combination to preserve the monocytic
precursor in cell suspension culture assays. These results indicate that IL-6 and MGF in conjunction with M-CSF affect CD34+ cells especially at precursor level without distinct effect on the more mature stages. Secondly we studied whether M-CSF
is only critical for the monocytic lineage or also affects dendritic cell (DC) development. Indeed, we were able to culture
CD83+ DC from CD34+ progenitor cells in the presence of M-CSF in conjunction with TNF-α, IL-4, and MGF although their absolute number is almost

A gas chromatographic-electron capture detection (GC-ECD) method for the analysis of deoxynivalenol (DON) in cereals was investigated.
The sample was extracted with a mixture of acetonitrile-water and purified with a MycoSep #225 column. The silylation was
performed with Tri-Sil-TBT reagent, followed by dilution with hexane and a washing step with buffer. By using Tri-Sil-TBT
reagent no double peaks were observed for DON in the gas chromatograms, in comparison with two other silylation reagents TMSI
and Tri-Sil-Z. The use of trichothecolone (TRI) as an internal standard for DON was studied in order to indicate possible
problems in the derivatisation reaction. TRI proved to be a relatively good internal standard for DON in cereal samples, as
well as 1,1-bis-(4-chlorophenyl)-2,2-dichloroethylene (DDE), which was used as a GC standard for ensuring the function of
GC-ECD. During the study, a matrix effect was clearly observed between the cereal matrix-assisted calibration curve and the
calibration curve prepared without cereal matrix. The results of spiked and reference material samples, quantified with the
calibration curve prepared without and with matrix, demonstrated that the matrix affects the results. However, after recovery
correction the results were comparable. The validation results demonstrated that the GC-ECD method for DON analysis in cereals

Regulation of mRNA stability and translation plays a critical role in determining protein abundance within cells. Processing bodies (P‐bodies) are critical regulators of these processes. Here, we report that the Pim1 and 3 protein kinases bind to the P‐body protein enhancer of mRNA decapping 3 (EDC3) and phosphorylate EDC3 on serine (S)161, thereby modifying P‐body assembly. EDC3 phosphorylation is highly elevated in many tumor types, is reduced upon treatment of cells with kinase inhibitors, and blocks the localization of EDC3 to P‐bodies. Prostate cancer cells harboring an EDC3 S161A mutation show markedly decreased growth, migration, and invasion in tissue culture and in xenograft models. Consistent with these phenotypic changes, the expression of integrin β1 and α6 mRNA and protein is reduced in these mutated cells. These results demonstrate that EDC3 phosphorylation regulates multiple cancer‐relevant functions and suggest that modulation of P‐body activity may represent a new paradigm for cancer treatment. 相似文献

Using a longitudinal network analysis approach, we investigate the structural development of the knowledge base of Wikipedia in order to explain the appearance of new knowledge. The data consists of the articles in two adjacent knowledge domains: psychology and education. We analyze the development of networks of knowledge consisting of interlinked articles at seven snapshots from 2006 to 2012 with an interval of one year between them. Longitudinal data on the topological position of each article in the networks is used to model the appearance of new knowledge over time. Thus, the structural dimension of knowledge is related to its dynamics. Using multilevel modeling as well as eigenvector and betweenness measures, we explain the significance of pivotal articles that are either central within one of the knowledge domains or boundary-crossing between the two domains at a given point in time for the future development of new knowledge in the knowledge base. 相似文献

In four studies we tested a new methodological approach to the investigation of evaluation bias. The usage of piecewise growth curve modeling allowed for investigation into the impact of people’s attitudes on their persuasiveness ratings of pro- and con-arguments, measured over the whole range of the arguments’ polarity from an extreme con to an extreme pro position. Moreover, this method provided the opportunity to test specific hypotheses about the course of the evaluation bias within certain polarity ranges. We conducted two field studies with users of an existing online information portal (Studies 1a and 2a) as participants, and two Internet laboratory studies with mostly student participants (Studies 1b and 2b). In each of these studies we presented pro- and con-arguments, either for the topic of MOOCs (massive open online courses, Studies 1a and 1b) or for the topic of M-learning (mobile learning, Studies 2a and 2b). Our results indicate that using piecewise growth curve models is more appropriate than simpler approaches. An important finding of our studies was an asymmetry of the evaluation bias toward pro- or con-arguments: the evaluation bias appeared over the whole polarity range of pro-arguments and increased with more and more extreme polarity. This clear-cut result pattern appeared only on the pro-argument side. For the con-arguments, in contrast, the evaluation bias did not feature such a systematic picture. 相似文献
