Synopsis We studied the timing of migratory fish spawning in the last dam-free stretch of the upper Paraná River and in Itaipu Reservoir. Eggs were more common in the Amambai and Ivaï Rivers, while larvae predominated in the Paraná River and in Itaipu Reservoir. Both eggs and larvae were more abundant at night. The highest abundance of eggs was in October and that of larvae in November. Migratory species predominated in the Amambai and Paraná Rivers, and non-migratory species in the Ivaï River and Itaipu Reservoir. The predominance of eggs in the upper and middle portions, and larvae in the lower, infer that there are spawning sites in the former and nurseries in the latter. The high nocturnal abundance of eggs is associated with spawning at sunset and that of larvae with feeding, avoidance of predators and nocturnal disorientation. The presence of tributaries such as the Amambai and Ivaí Rivers in the last dam-free stretch of the Paraná River is extremely important to the maintenance of regional fish diversity and fish stocks in both the Paraná River and Itaipu Reservoir.  相似文献

The sequence of paired, a pair-rule gene required for segmentation in Drosophila, is presented. A search for genes with domains homologous to the paired gene was initiated and three homologues from a set of 12 were characterized with respect to temporal or spatial expression and sequence homologies. All four are transcribed in early development, one in the oocyte and during cleavage stages in the form of a gradient. In addition to the prd-specific his-pro repeat, some of the 12 genes contain M-repeats and two new types of homeo boxes not detectable by hybridization with the two known classes of homeo boxes. The observed linking of gene sets through combinations of homologies coding for protein domains is consistent with a general network concept of gene action.  相似文献

The distribution of Risso's dolphin ( Grampus griseus ) was examined with respect to two physiographic variables, water depth and depth gradient (sea floor slope), in the northern Gulf of Mexico, using shipboard and aerial survey data collected from 1992 to 1994. Univariate x2 analyses demonstrated that Risso's dolphins are distributed non-uniformly with respect to both depth and depth gradient. A bivariate analysis of the shipboard data indicated that Risso's dolphins utilize the steep sections of the upper continental slope in the northern Gulf of Mexico. This narrow core habitat is in waters bounded by the 350-m and 975-m1 isobaths with depth gradients greater than 24 m per 1.1 km and consists of only 2% of the surface area of the entire Gulf of Mexico. Sighting rates inside this region were nearly 5 and 6 times the average for the shipboard and aerial surveys, respectively. Of the groups sighted outside this region, 40% (shipboard) and 73% (aerial) were encountered within 5 km of it. Since it is unlikely that the physiography alone can attract dolphins, oceanographic mechanisms that may concentrate prey along the steep upper continental slope are discussed. The implications of this distribution, including potential prey species, foraging strategies, and impacts of proposed mineral exploration and development, are also considered.  相似文献

A high molecular weight fructan was isolated from garlic and the structure determined by enzymatic, chemical and spectroscopic (NMR) methods. It was found that the garlic fructan belongs to the neokestose family. It has a (2 --> 1)-linked beta-D-Fruf backbone with (2 --> 6)-linked beta-D-Fruf side chains. A structural model was postulated for a degree of polymerisation of about 58. This model was substantiated using an endo-inulinase purified from Aspergillus ficuum and by 1H and 13C NMR spectroscopy.  相似文献

Background

Peroxisome proliferator-activated receptor gamma (PPARγ) agonists are clinically used to counteract hyperglycemia. However, so far experienced unwanted side effects, such as weight gain, promote the search for new PPARγ activators.

Methods

We used a combination of in silico, in vitro, cell-based and in vivo models to identify and validate natural products as promising leads for partial novel PPARγ agonists.

Results

The natural product honokiol from the traditional Chinese herbal drug Magnolia bark was in silico predicted to bind into the PPARγ ligand binding pocket as dimer. Honokiol indeed directly bound to purified PPARγ ligand-binding domain (LBD) and acted as partial agonist in a PPARγ-mediated luciferase reporter assay. Honokiol was then directly compared to the clinically used full agonist pioglitazone with regard to stimulation of glucose uptake in adipocytes as well as adipogenic differentiation in 3T3-L1 pre-adipocytes and mouse embryonic fibroblasts. While honokiol stimulated basal glucose uptake to a similar extent as pioglitazone, it did not induce adipogenesis in contrast to pioglitazone. In diabetic KKAy mice oral application of honokiol prevented hyperglycemia and suppressed weight gain.

Conclusion

We identified honokiol as a partial non-adipogenic PPARγ agonist in vitro which prevented hyperglycemia and weight gain in vivo.

General significance

This observed activity profile suggests honokiol as promising new pharmaceutical lead or dietary supplement to combat metabolic disease, and provides a molecular explanation for the use of Magnolia in traditional medicine.  相似文献

We find that both human red blood cells and rabbit skeletal muscle contain a soluble activator which can stimulate (Ca2+ + Mg2+)-ATPase activity. The activator protein from either source can enhance the (Ca2+ + Mg2+)-ATPase of both the red blood cell membrane and the microsomal fraction from skeletal muscle. The data suggest that they are members of the class of Ca2+-binding modulator proteins. A possible physiological role for the skeletal muscle activator protein in the contractile process is discussed.  相似文献

Plasmid-encoded GM-CSF (pGM-CSF) is an adjuvant for genetic vaccines; however, little is known about how pGM-CSF enhances immunogenicity. We now report that pGM-CSF injected into mouse muscle leads to a local infiltration of potential APCs. Infiltrates reached maximal size on days 3 to 5 after injection and appeared in several large discrete clusters within the muscle. Immunohistological studies in muscle sections from mice injected with pGM-CSF showed staining of cells with the macrophage markers CD11b, Mac-3, IA(d)/E(d) and to the granulocyte marker GR-1 from day 1 through day 14. Cells staining with the dendritic cell marker CD11c were detected only on days 3 to 5. Muscles injected with control plasmids did not stain for CD11c but did stain for CD11b, Mac-3, IA(d)/E(d), and GR-1. No staining was observed with the APC activation markers, B7.1 or CD40, or with markers for T or B cells. These findings are consistent with the infiltrating cells in the pGM-CSF-injected muscles being a mixture of neutrophils, macrophages, and immature dendritic cells and suggest that the i.m. APCs may be enhancing immune responses to coinjected plasmid Ags. This hypothesis is supported by data showing that 1) separation of injections with pGM-CSF and Ag-expressing plasmid into different sites did not enhance immune responses and 2) immune enhancement was associated with the presence of CD11c+ cells in the infiltrates. Thus, pGM-CSF enhancement may depend on APC recruitment to the i.m. site of injection.  相似文献

The shallow marine and subaerial sedimentary and hydrothermal rocks of the ~3.48 billion‐year‐old Dresser Formation are host to some of Earth's oldest stromatolites and microbial remains. This study reports on texturally distinctive, spherulitic barite micro‐mineralization that occur in association with primary, autochthonous organic matter within exceptionally preserved, strongly sulfidized stromatolite samples obtained from drill cores. Spherulitic barite micro‐mineralization within the sulfidized stromatolites generally forms submicron‐scale aggregates that show gradations from hollow to densely crystallized, irregular to partially radiating crystalline interiors. Several barite micro‐spherulites show thin outer shells. Within stromatolites, barite micro‐spherulites are intimately associated with petrographically earliest dolomite and nano‐porous pyrite enriched in organic matter, the latter of which is a possible biosignature assemblage that hosts microbial remains. Barite spherulites are also observed within layered barite in proximity to stromatolite layers, where they are overgrown by compositionally distinct (Sr‐rich), coarsely crystalline barite that may have been sourced from hydrothermal veins at depth. Micro‐spherulitic barite, such as reported here, is not known from hydrothermal systems that exceed the upper temperature limit for life. Rather, barite with near‐identical morphology and micro‐texture is known from zones of high bio‐productivity under low‐temperature conditions in the modern oceans, where microbial activity and/or organic matter of degrading biomass controls the formation of spherulitic aggregates. Hence, the presence of micro‐spherulitic barite in the organic matter‐bearing Dresser Formation sulfidized stromatolites lend further support for a biogenic origin of these unusual, exceptionally well‐preserved, and very ancient microbialites.  相似文献
