The ergosterol biosynthesis pathway is well understood in Saccharomyces cerevisiae, but currently little is known about the pathway in plant‐pathogenic fungi. In this study, we characterized the Fusarium graminearum FgERG4 gene encoding sterol C‐24 reductase, which catalyses the conversion of ergosta‐5,7,22,24‐tetraenol to ergosterol in the final step of ergosterol biosynthesis. The FgERG4 deletion mutant ΔFgErg4‐2 failed to synthesize ergosterol. The mutant exhibited a significant decrease in mycelial growth and conidiation, and produced abnormal conidia. In addition, the mutant showed increased sensitivity to metal cations and to various cell stresses. Surprisingly, mycelia of ΔFgErg4‐2 revealed increased resistance to cell wall‐degrading enzymes. Fungicide sensitivity tests revealed that ΔFgErg4‐2 showed increased resistance to various sterol biosynthesis inhibitors (SBIs), which is consistent with the over‐expression of SBI target genes in the mutant. ΔFgErg4‐2 was impaired dramatically in virulence, although it was able to successfully colonize flowering wheat head and tomato, which is in agreement with the observation that the mutant produces a significantly lower level of trichothecene mycotoxins than does the wild‐type progenitor. All of these phenotypic defects of ΔFgErg4‐2 were complemented by the reintroduction of a full‐length FgERG4 gene. In addition, FgERG4 partially rescued the defect of ergosterol biosynthesis in the Saccharomyces cerevisiae ERG4 deletion mutant. Taken together, the results of this study indicate that FgERG4 plays a crucial role in ergosterol biosynthesis, vegetative differentiation and virulence in the filamentous fungus F. graminearum.  相似文献

Esophageal carcinoma, with a increasing incidence, is one of the most aggressive carcinomas in gastrointestinal tract. Epidemiologic studies demonstrate an association of oral pathogens with multiple diseases, including rheumatoid arthritis, cardiovascular diseases, diabetes, and gastrointestinal malignancies. Nevertheless, a causal relationship between oral pathogens and esophageal squamous cell carcinoma (ESCC) has not been elucidated. Here, we found that Porphyromonas was significantly enriched in the saliva of patients with ESCC, compared with that in normal human. In vitro studies showed that Porphyromonas gingivalis (P. gingivalis) promoted the proliferation and motility of ESCC cells, as evidenced by up regulated expression of key molecules implicated in NF-κB signaling pathway. These findings, for the first time, demonstrated a role of oral pathogens in inducing ESCC tumorigenesis and metastasis, which might involve regulation of NF-κB signaling pathway.  相似文献

Seven new polyhydroxypregnane glycosides, named cynotophyllosides P–V, together with three known analogs were isolated from the roots of Cynanchum otophyllum C.K.Schneid . Their structures were elucidated by a variety of spectroscopic techniques, as well as acid‐catalyzed hydrolysis. All isolates were tested for their immunological activities in vitro against Con A‐ and LPS‐induced proliferation of mice splenocytes. Immunoenhancing (for 1 , 9 ) and immunosuppressive (for 2 ) activities were observed. Furthermore, cynotophylloside R ( 3 ) showed immunomodulatory as it enhanced the proliferation of splenocytes in low concentration and suppressed immune cells in concentration more than 1.0 μg/ml.  相似文献

Food waste (FW) management is a global conundrum because of the rapid population growth and growing economic activity. Currently, incineration and landfill are still the main means for FW management, while their environmental sustainability and economic viability have been in question. Recently, the biological processes including anaerobic digestion, aerobic composting, bioethanol fermentation, feed fermentation etc. have attracted increasing interest with the aims for energy and resource recovery from FW. However, these biological approaches have inherent drawbacks, and cannot provide a comprehensive solution for future FW management. Therefore, this review attempts to offer a critical and holistic analysis of current biotechnologies for FW management with the focus on the challenges and solutions forward. The biological approaches towards future FW management should be able to achieve both environmental sustainability and economic viability. In this instance, the concept of zero solid discharge-driven resource recovery has thus been put forward. According to which, several innovative biological processes for FW management are further elucidated with critical analysis on their engineering feasibility and environmental sustainability. It turns out that is an urgent need for turning current single task-orientated bioprocess to an integrated biological process with multiple tasks of concurrent recovery of water, resource and energy together with zero-solid discharge.  相似文献

The structural, elastic and electronic properties of Co7M6 (M?=?W, Mo, Nb) μ phases were investigated by first-principles calculations based on the density functional theory (DFT). The calculated cohesive energy indicates that Co7M6 (M?=?W, Mo, Nb) μ phases are thermodynamically stable. Besides, Co7W6 owns a higher structural stability than that of Co7Mo6 and Co7Nb6. The obtained elastic constant demonstrates that Co7M6 (M?=?W, Mo, Nb) are mechanically stable. With Voigt-Reuss-Hill (VRH) approximation, the elastic bulk modulus (B), shear modulus (G), Young's modulus (E) and Poisson's ratio (ν) were derived. The ductility and plasticity as well as the elastic anisotropy of the three phases were discussed in details. Finally, the density of states and charge density difference were also analysed to reveal the underlying mechanism of structural stability and the elastic properties.  相似文献
