Methylmethacrylate (MMA) is the most commonly used embedding medium for sectioning undecalcified bone; however, a number of problems exist with its use in a research laboratory. MMA requires a long infiltration time and temperature control, and it reacts with many polymers. We used Kleer Set resin™ as an alternative embedding medium for sectioning undecalcified bone specimens. Fluorochrome labeled bone specimens were sectioned transversely using a ground section technique and longitudinally on a sledge macrotome. The slides were viewed using both transmitted light and epifluorescence microscopy. High quality sections were obtained using Kleer Set resin™ for both sectioning techniques. We have shown that this new embedding medium is simpler, safer, quicker to use and does not interfere with visualization of fluorochromes.  相似文献

As shown by us previously (van Berkel et al. 1985. J. Biol. Chem. 260: 2694-2699 and van Berkel et al. 1985. J. Biol. Chem. 260: 12203-12207) the clearance of both low density lipoproteins (LDL) and high density lipoproteins (HDL) from the blood can be greatly enhanced by pretreatment of these lipoproteins with a tris-galactosylated cholesterol derivative, which makes these particles recognizable by hepatic galactosyl-receptors. Here we report that intravenous infusion of the (water-soluble) tris-galactosyl-cholesterol in rats caused a dose-dependent decrease of the plasma cholesterol level. This fall was sustained long after termination of the infusion. It was not observed upon infusion of tris-glucosyl-cholesterol. The fall in plasma cholesterol was accompanied by an increase in hepatic cholesterol. Upon injection of rat HDL and LDL labeled in their cholesteryl ester moieties, plasma clearance of label in both lipoproteins was enhanced in rats infused with tris-galactosyl-cholesterol, the stimulation being more pronounced when the label was in HDL. The appearance of label in bile was also enhanced in the rats receiving the compound, again more markedly when the label was given as HDL. Ninety four percent or more of the radioactivity excreted in the bile was in the form of bile salts, with conjugated cholate being the major species in both control and treated rats; 6% or less of the radioactivity in the bile was as free cholesterol. Infusion of tris-galactosyl-cholesterol constitutes a new and defined method of lowering plasma lipoprotein levels by enhancing their uptake in the liver.  相似文献

Isopod crustaceans have developed a variety of feeding strategies, which impact on their various life habits over time. Apart from morphological adaptations and the typical secondary deformations that some parasitic isopods inflict on their host's exoskeleton, traces of feeding behaviours involving isopods on their victim/food are seldom fossilized. Many of these rare occurrences consist of cases for which the degree of association between the isopods and their potential food source is unclear, or the interaction only very briefly explained (recently analysed cases excepted). There are two limiting problems in identifying the biological nature of fossilized associations: (1) direct associations of organisms preserved as ‘imprint’ (as opposed to inclusions in cherts or amber) are shaped by several taphonomic events difficult to identify (such as the time of death, burial and fossilization of the organism); (2) even in modern nature, differences within syn vivo interactions (like parasitism and micropredation) are poorly understood in marine systems. We report the occurrence of isopods associated with ancient chondrichthyans which also represent rare cases of the preservation of several fossil isopods on larger organisms. These organisms are adult electric rays of the genus ?Titanonarke Carvalho from the late Ypresian (Eocene) of the Monte Postale site, Bolca Lagerstätte, Italy. By examining: (1) the involved lineages of rays and isopods; (2) the taphonomy of the association; (3) its environmental context; and (4) biological/adaptive features; we identify this association as a selective case, either of ancient scavenging or of micropredation on specific electric rays by isopods.  相似文献
