Tissues characterized by various degrees of immune privilege are thought to differ in repair processes. This circumstance may be due to mast cells (MCs) that occur in all tissues of the body, secrete a wide range of biologically active compounds, and play an important role in the regulation of repair processes. The present paper reports the results of investigations of morphometric parameters and functional activity of MCs in tissues characterized by various degrees of immune privilege such as the skin and testis. It was shown that MCs migrate into the skin in the early stages after damage, followed by a slow increase in the synthetic activity and degranulation index of MCs for 30 days. In the case of the testis, the MC degranulation index increases immediately after damage, with a pronounced migration of MCs being absent. The stabilization of MC membranes with ketotifen inhibits skin repair; namely, there is no increase in the thickness of the dermis and epidermis and the number of fibroblasts and collagen fibers, along with slowing down of the scar formation. Meanwhile, the MC inactivation contributes to the reparative regeneration of the testis. This circumstance is confirmed by a significant reduction of the number of nonfunctioning tubules and an increase in the number of normal spermatogonia, which are a proliferative pool for all subsequent stages of spermatogenesis. Thereby, the number and the functional state of MCs influences the repair processes in tissues characterized by various degrees of immune privilege.  相似文献

Bacterial amidases and nitrile hydratases can be used for the synthesis of various intermediates and products in the chemical and pharmaceutical industries and for the bioremediation of toxic pollutants. The aim of this study was to analyze the expression of the amidase and nitrile hydratase genes of Rhodococcus erythropolis and test the stereospecific nitrile hydratase and amidase activities on chiral cyanohydrins. The nucleotide sequences of the gene clusters containing the oxd (aldoxime dehydratase), ami (amidase), nha1, nha2 (subunits of the nitrile hydratase), nhr1, nhr2, nhr3 and nhr4 (putative regulatory proteins) genes of two R. erythropolis strains, A4 and CCM2595, were determined. All genes of both of the clusters are transcribed in the same direction. RT-PCR analysis, primer extension and promoter fusions with the gfp reporter gene showed that the ami, nha1 and nha2 genes of R. erythropolis A4 form an operon transcribed from the Pami promoter and an internal Pnha promoter. The activity of Pami was found to be weakly induced when the cells grew in the presence of acetonitrile, whereas the Pnha promoter was moderately induced by both the acetonitrile or acetamide used instead of the inorganic nitrogen source. However, R. erythropolis A4 cells showed no increase in amidase and nitrile hydratase activities in the presence of acetamide or acetonitrile in the medium. R. erythropolis A4 nitrile hydratase and amidase were found to be effective at hydrolysing cyanohydrins and 2-hydroxyamides, respectively.  相似文献

A memristor is a nonlinear element because its current-voltage characteristic is similar to that of a Lissajous pattern for nonlinear systems. This element was postulated recently and researchers are looking for it in different biosystems. We investigated electrical circuitry of red Irish potato tubers (Solanum tuberosum L.). The goal was to discover if potato tubers might have a new electrical component - a resistor with memory. The analysis was based on a cyclic current-voltage characteristic where the resistor with memory should manifest itself. We found that the electrostimulation by bipolar sinusoidal or triangle periodic waves induces electrical responses in the potato tubers with fingerprints of memristors. Tetraethylammonium chloride, an inhibitor of voltage gated K+ channels, transforms a memristor to a resistor in potato tubers. Our results demonstrate that a voltage gated K+ channel in the excitable tissue of potato tubers has properties of a memristor. Uncoupler carbonylcyanide-4-trifluoromethoxy-phenyl hydrazone decreases the amplitude of electrical responses at low and high frequencies of bipolar periodic sinusoidal or triangle electrostimulating waves. The discovery of memristors in plants creates a new direction in the understanding of electrical phenomena in plants.  相似文献

We analyzed, for the first time, both in vitro and in vivo, the effect of very low density lipoprotein (VLDL), or of pure triglycerides, on high-density lipoprotein (HDL)-associated paraoxonase1 (PON1) catalytic activities. Incubation of serum or HDL from healthy subjects with VLDL (0-330 μg protein/mL) significantly decreased serum PON1 lactonase or arylesterase activities by up to 11% or 24%, and HDL-associated PON1 lactonase or arylesterase activities by up to 32% or 46%, respectively, in a VLDL dose-dependent manner. VLDL (0-660μg protein/mL) also inhibited recombinant PON1 (rePON1) lactonase or arylesterase activities by up to 20% or 42%, respectively. Similar inhibitory effect was noted upon rePON1 incubation with pure triglyceride emulsion. Bezafibrate therapy to three hypertriglyceridemic patients (400 mg/day, for one month) significantly decreased serum triglyceride concentration by 67%, and increased serum HDL cholesterol levels by 48%. PON1 arylesterase or paraoxonase activities in the patients' HDL fractions after drug therapy were significantly increased by 86-88%, as compared to PON1 activities before treatment. Similarly, HDL-PON1 protein levels significantly increased after bezafibrate therapy. Finally, bezafibrate therapy improved HDL biological activity, as HDL obtained after drug therapy showed increased ability to induce cholesterol efflux from J774A.1 macrophages, by 19%, as compared to HDL derived before therapy. We thus conclude that VLDL triglycerides inhibit PON1 catalytic activities, and bezafibrate therapy significantly improved HDL-PON1 catalytic and biological activities. ? 2012 International Union of Biochemistry and Molecular Biology, Inc.  相似文献

In Drosophila polytene chromosomes, regions of intercalary heterochromatin are scattered throughout the euchromatic arms. Here, we present data on the first fine analysis of the individual intercalary heterochromatin region, 75C1-2, located in the 3L chromosome. By using electron microscopy, we demonstrated that this region appears as three closely adjacent condensed bands. Mapping of the region on the physical map by means of the chromosomal rearrangements with known breakpoints showed that the length of the region is about 445 kb. Although it seems that the SUUR protein binds to the whole 75C1-2 region, the proximal part of the region is fully polytenized, so the DNA underreplication zone is asymmetric and located in the distal half of the region. Finally, we speculate that intercalary heterochromatin regions of Drosophila polytene chromosomes are organized into three different types with respect to the localization of the underreplication zone.  相似文献

The effects of bis(adenosyl-5)oligophosphates on cation conductance-promoting ATP receptors at the somatic membrane of rat sensory ganglia neurons were investigated. Both bis(adenosyl-5)tetraphosphate and bis(adenosyl-5)pentaphosphate act as partial agonists of ATP receptors at saturating concentration and activate ionic currents of almost one order of magnitude below the amplitude of peak ATP-activated currents. Combined application of these two substances to the neuron produces a considerable decline in the amplitude of ATP-activated ionic currents, competing with ATP for common binding sites.A. A. A quantum-chemical study has been made of the interactions of pheromones of some lepidopteran forest pests (Siberian moth Dendrolimus superans sibiricus Tschetv and gypsy moth Limantria dispar L.) with components of the ambient air, as well as the effect of electromagnetic radiation on the pheromones. It is found that the reactions of pheromones with substances contained in the forest air are irreversible and proceed with liberation of heat. Electromagnetic radiation quite strongly affects the structure of pheromones, whereby the pheromone molecule is activated and can readily enter into reactions.  相似文献
