The effect of two quinoline-N-oxide derivatives (2-(4′-nitrostyryl)-quinoline-l-oxide (2-NSQO) and 4-(4′-nitrostyryl)-quinoline-1-oxide (4-NSQO)) on modulation of microsomal NADPH oxidoreductase activity, nicotinamide coenzyme concentrations, and induction of apoptosis in K562 human erythroleukemia cells has been. 4-NSQO at the concentration of (10 μM) and 2-NSQO (10 μM) inhibited the activity of microsomal NADPH cytochrome c reductases in tumor cells by 15 and 50% respectively. Treatment of cells with these compounds for two days resulted in the activation of caspase-9 and caspase-3, the increase in ethidium bromide and 4′,6-diamidino-2-phenylindole (DAPI) fluorescence upon DNA binding, and induction of apoptosis. The latter was preceded by the decrease in intracellular nicotinamide coenzyme concentrations. The results obtained allow considering 4-NSQO (and its structural analogs) as perspective compounds for further experimental studies as antitumor agent with low toxic effect on tissues of an organism. 相似文献

Pseudomonas’ IncP-7 plasmids play significant role in the environmental biodegradative potential and sometimes carry antibiotic-resistance genes. Rms148 plasmid was used as archetypal P-7 plasmid in microbiological incompatibility studies for more then 30 years. However, the structure of its basic replicon was not described until now; furthermore, the phylogenetic relationships between all known plasmids within the IncP-7 group have not yet been studied. In the course of our study, we have constructed two pairs of primers to amplify the main components of the region of the initiation of P-7 replication, and the subsequent screening of repA intragenic polymorphism was performed using the laboratory collection of IncP-7 plasmids. The minimal replicon of Rms148 was determined and its nucleotide sequence was found to be 81–83% identical to repA-oriV of known P-7 plasmids and is considered to fall into a separate clade of the corresponding phylogenetic tree. Additionally, repA group members seem to be more conservative than the putative oriV region. The estimated amino acid sequence and predicted secondary and tertiary structures of Rms148 RepA protein allowed us to make the assumption that the initiation of replication in plasmids of the P-7 incompatibility group is described by the same model as for the unclassified cryptic plasmid pPS10. 相似文献

Series of phthalocyanines of zirconium containing lysine, citric, nonanoic acid residues and dibenzolylmethane groups as out-of-plane ligands are firstly studied as inhibitors of fibrillogenesis using cyanine-based fluorescent inhibitory assay. It was shown that studied phthalocyanines at concentration of 20μM inhibited aggregation reaction on 38.5-57.6% and inhibitory activity of phthalocyanines depended on the chemical nature of out-of-plane ligand. For the most active compound PcZrLys(2) (zirconium phthalocyanine containing lysine fragment) the efficient inhibitor concentration was estimated to be 37μM. AFM studies have shown that in the presence of PcZrLys(2) the inhibition of fibrils formation and formation of spherical oligomeric aggregates took place. Due to the ability of phthalocyanines to decrease efficiently protein aggregation into the amyloid fibrils, modification of phthalocyanine molecules via out-of-plane substitutions was proposed as approach for design of anti-fibrillogenic agents with required properties. 相似文献

The effects of mutations and genetic background on the mating activity of male and receptivity of female Drosophila melanogaster have been studied at different population densities. Population density, as well as its combinations with other factors, significantly affects mating behavior of D. melanogaster. There are two distinct trends in the effect of this factor on mating behavior: the maximum larval overpopulation may cause either a significant suppression of the behaviors studied or an increase in their expressivity. In Drosophila polytene chromosomes, regions of intercalary heterochromatin are scattered throughout the euchromatic arms. Here, we present
data on the first fine analysis of the individual intercalary heterochromatin region, 75C1-2, located in the 3L chromosome.
By using electron microscopy, we demonstrated that this region appears as three closely adjacent condensed bands. Mapping
of the region on the physical map by means of the chromosomal rearrangements with known breakpoints showed that the length
of the region is about 445 kb. Although it seems that the SUUR protein binds to the whole 75C1-2 region, the proximal part
of the region is fully polytenized, so the DNA underreplication zone is asymmetric and located in the distal half of the region.
Finally, we speculate that intercalary heterochromatin regions of Drosophila polytene chromosomes are organized into three different types with respect to the localization of the underreplication zone. 相似文献
