Ribonuclease of Bacillus intermedius 7 P. Purification by chromatography on phosphocellulose and several characteristics of the homogeneous enzyme]

A new procedure for isolation of homogenous ribonuclease of Bac. intermedius from a commercial source is described. The yields of 140 mg of RNAse from 200 g of the enzymic powder were attained. The amino acid composition of the enzyme was determined. The RNAse contains neither the sulfhydryl groups nor the disulfide bonds and has only one histidine residue. At the same time the amount of aromatic amino acid residues is relatively high. The enzyme is highly resistant to heat and acid treatment but is less stable in an alkaline solution. The pH optimum of the RNAse for the RNA digestion is 8,5; the temperature optimum for this reaction is 37 degrees. A spectrophotometric method for the RNAse activity assay using polyA as a specific substrate was developed. The purified product provides a suitable starting material for structural studies.     

Participation of the hypothalamus in the pituitary and testicular responses to administration of the antiandrogen 4-nitro-3-trifluoro-methylisobutyranilide]

The effect of the antiandrogen 4-nitro-3-trifluoro-methylisobutyranilide (flutamide, NFBA) on the content of follitropine, lutropine and testosteron in the blood plasma as well as on the testicular activity of the steroid delta 5-3 beta-Ol-dehydrogenase (SD) has been studied in male rats with intact and destroyed hypothalamic median eminence. In sham-operated rats with intact hypothalamus the levels of lutropin, follitropin, testosteron and the enzymatic activity were 5, 2, 4 and 3-4 times as increased, respectively, under the effect of NFBA. After destroyal of the hypothalamic median eminence NFBA treatment did not change follitropin secretion, while lutropin, testosteron plasma levels and SD activity were only 1 1/2-2 times higher, i.e. significantly less as compared to those in sham-operated animals. The data obtained suggest the most important role of the hypothalamus in pituitary and testicular responses to NFBA administration.     

Structure of the B DNA cationic shell as revealed by an X-ray diffraction study of CsDNA. Sequence-specific cationic stabilization of B form DNA

Synchrotron radiation diffraction data for phage T2 CsDNA fibres have been used to determine the co-ordinates of the caesium ions in crystalline B form DNA. The R value is 0.16 for an optimized structure. The caesium ions are distributed equally between the narrow and wide grooves of B DNA and are located close to the dyad axes lying between the planes of adjacent base-pairs. On the wide-groove side the cations are separated from the nearest phosphate atoms by a hydration layer one to two water molecules thick. In the narrow groove the cations are directly co-ordinated to the base atoms and, for six out of ten possible DNA stacking types, form chelation complexes: O-2(Pyr)-Cs+-O-2(Pyr), O-2(Pyr)-Cs+-N-3(Pur) or N-3(Pur)-Cs+-N-3(Pur), which stabilize the B conformation. The steric properties of such complexes as estimated for different base sequences and for different ions are consistent with the structural behaviour of double-helical polynucleotides with different base sequences, as experimentally observed.     

Changes in the activity of glyoxylate cycle enzymes of yeast during hydrocarbon nutrition]

Six strains of Candida guillermondii of different productivity showed a higher isocitrate lyase and malate dehydrogenase activity of cell-free extracts when grown on paraffin than when grown on glucose. In most cases isocitrate dehydrogenase activity was higher on glucose than on paraffin. A positive correlation between isocitrate activity and growth rate was found from studies of the strains of varying growth rate and the cultures cultivated under different conditions (nitrogen content and the presence or absence of biotin or autolysate in the medium).
