The solution conformation of rabbit liver Cd27+-metallothionein-2 was determined by nuclear magnetic resonance (n.m.r.) and distance geometry. The n.m.r. data are based on complete sequence-specific resonance assignments for the polypeptide chain. Reversion of the transformed phenotype of B16 mouse melanoma: involvement of an 83 kd cell surface glycoprotein in specific growth inhibition 总被引:3,自引:0,他引:3
Treating B16 mouse melanoma cells with monoclonal antibody NORM-2 reduces cell growth in tissue culture, agar, and syngeneic mice. We show that the NORM-2 antibody recognizes an integral 83 kd glycoprotein that is mobile in the plane of the plasma membrane of B16 melanoma cells. Expression of the glycoprotein is reduced under conditions that inhibit B16 growth, such as low serum, high cell density, and addition of transforming growth factor-beta. The glycoprotein reappears during S phase, when growth-arrested cells are restimulated. The NORM-2 antigen appears to be involved in growth regulation of B16 melanoma cells both in vitro and in vivo. 相似文献

The impact of different land-use types on species is traditionally estimated by correlating landscape proportions recorded in buffer areas around focal points with species data observed at these sites. If a high proportion of a specific land-use type exists within a small radius, it will be accumulated in larger buffers and may confound the interpretation at larger scales. We sampled freshwater invertebrates in ten streams using cages with artificial substrate and compared the effects of arable land proportions calculated in disc-shaped buffers of increasing radius versus areas calculated from non-overlapping rings of increasing radius. We hypothesize that (1) the accumulative disc-based approach leads to confounding effects across increasing buffer size and that (2) the use of ring-based methods facilitates the identification of relevant scales for conservation measures. The abundance of crustaceans showed a positive relationship with arable land proportions, but Plecoptera abundance and the taxonomic richness of Ephemeroptera and Plecoptera decreased with increasing arable land proportions in the surrounding landscape. Our results further support the presence of confounding effects in disc-based analyses, as correlations between arable land proportions and Crustacea, or Plecoptera, respectively, were affected by the accumulation of small-scale area proportions. The distance at which arable land proportions significantly affected benthic fauna in freshwater streams was consistently shorter if calculated from rings rather than from discs. Although an a priori definition of ring width introduces new challenges, a combined use of disc- and ring-based techniques for the estimation of land-use effects may substantially improve the realization of conservation and protection measures in terrestrial and aquatic systems. 相似文献

Introduction
Large osteochondral defects of the weight-bearing zones of femoral condyles in young and active patients were treated by autologous transfer of the posterior femoral condyle (large osteochondral autogenous transplantation system (MegaOATS)). The technique presented is a sound and feasible salvage procedure to address large osteochondral defects in weight-bearing zones.Methods
Thirty-six patients between July 1996 and December 2000 were included. Thirty-three patients (10 females, 23 males) were evaluated by the Lysholm score and X-ray scans. A random sample of 16 individuals underwent magnetic resonance imaging analysis. The average age at the date of surgery was 34.3 (15 to 59) years, and the mean follow up was 66.4 (46 to 98) months. The mean defect size was 6.2 (2 to 10.5) cm2, in 27 patients affecting the medial femoral condyle and in six patients affecting the lateral femoral condyle. Trauma or osteochondrosis dissecans were pathogenetic in 82%.Results
The Lysholm score in all 33 individuals showed a highly significant increase from a preoperative median 49.0 points to a median 86.0 points (P ≤ 0.001). Twenty-seven patients returned to recreational sports. X-ray scans showed a rounding of the osteotomy edge in 24 patients, interpreted as a partial remodelling of the posterior femoral condyle. Preoperative osteoarthritis in 17 individuals was related to significant lower Lysholm scores (P = 0.014), but progression in 17 patients did not significantly influence the score results (P = 0.143). All 16 magnetic resonance imaging examinations showed vital and congruent grafts.Conclusion
Patients significantly improve in the Lysholm score, in daily-life activity levels and in return to recreational sports. Thirty-one out of 33 patients were comfortable with the results and would undergo the procedure again. The MegaOATS technique is therefore recommended as a salvage procedure for young individuals with large osteochondral defects in the weight-bearing zone of the femoral condyle. 相似文献38.

The sediment of Lake Balaton (Hungary) provides important information about the lake’s history, particularly with regard to
eutrophication. In this study, we used fossil pigment analysis and subfossil Cladocera remains preserved in a dated sediment
core to identify trophic stages from ~250 bc to present. Dates of the most recent eutrophic events are in good agreement with previously published data. In general, the
abundance and diversity of the Cladocera community increased with eutrophication and decreased with oligotrophication. The
sediments of Lake Balaton were characterised by Chydoridae remains, of which Alona species were the most abundant. Of these, Alona quadrangularis and Alona affinis accounted for 40 and 20% of the total Cladocera remains, respectively. The trophic state of Lake Balaton varied between mesotrophic
and eutrophic regimes. Seven different trophic periods were identified in Lake Balaton on the basis of Sedimentary Pigment
Degradation Unit (SPDU) content of the sediment. Eutrophic states were (1) from ~250 to ~30 bc, (3) between ~300 and ~590 ad, (5) between 1834 and 1944 and (7) from the 1960s until present. Mesotrophic states were (2) ~30 bc to ~300 ad, (4) 590–1834, (6) 1944–1960s. Discriminant analysis of the cladoceran data confirmed these historic events, except for the
short mesotrophic episode between 1944 and 1960. The first stage of eutrophication of Lake Balaton (~250 to ~30 bc) was characterised by extensive macrophyte vegetation, as indicated by the increasing abundance of vegetation-associated
Cladocera species (Eurycercus lamellatus, Sida crystallina, Pleuroxus sp.). Intensification of eutrophication was identified since the 1980s, reflected by a high abundance of Bosmina species. The most significant planktivorous fish of Lake Balaton was the Sabre carp (Pelecus cultratus), and when its number decreased, the abundance of Bosmina species increased. Plasmatic immunoreactive trypsin (IRT), thromboxane and trypsin-like enzymatic activity were measured in 117 patients at risk of developing adult respiratory distress syndrome (ARDS) (53 multiple injury, 30 abdominal surgery, 17 acute pancreatitis, 12 burnt and 5 disseminated intravascular coagulation patients). 69 of these patients developed ARDS.Immunoreactive trypsin and thromboxane were measured by radio-immuno-assay and trypsin-like enzymatic activity by spectrophotometry, using a specific chromogenic substrate.Mean IRT value was 675 ng/ml in ARDS and 265 ng/ml in non ARDS patients (p < 0.05). Mean IRT value was 685 ng/ml in sepatic and 170 ng/ml in non septic patients (p < 0.01). An abnormal trypsin-like enzymatic activity was measured in 26 ARDS patients. In 60 patients (37 ARDS and 23 non ARDS), thromboxane appeared in plasma simultaneously or about 24 hours after the beginning of IRT release. The importance of thromboxane release parallels the intensity of IRT. Originating from pancreas, trypsin can appear in plasma either by absortion from gastrointestinal tract or after pancreatic ischemia. 相似文献
