Laboratory experiments on Drosophila have often demonstrated increased heritability for morphological and life‐history traits under environmental stress. We used parent–offspring comparisons to examine the impact of humidity levels on the heritability of a physiological trait, resistance to heat, measured as knockdown time at constant temperature. Drosophila melanogaster were reared under standard nonstressful conditions and heat‐shocked as adults at extreme high or low humidity. Mean knockdown time was decreased in the stressful dry environment, but there was a significant sex‐by‐treatment interaction: at low humidity, females were more heat resistant than males, whereas at high humidity, the situation was reversed. Phenotypic variability of knockdown time was also lower in the dry environment. The magnitude of genetic correlation between the sexes at high humidity indicated genetic variation for sexual dimorphism in heat resistance. Heritability estimates based on one‐parent–offspring regressions tended to be higher under desiccation stress, and this could be explained by decreased environmental variance of heat resistance at low humidity. There was no indication that the additive genetic variance and evolvability of heat resistance differed between the environments. The pattern of heritability estimates suggests that populations of D. melanogaster may have a greater potential for evolving higher thermal tolerance under arid conditions.  相似文献

Agonist-induced internalization of G protein-coupled receptors (GPCRs) is an important mechanism for regulating signaling transduction of functional receptors at the plasma membrane. We demonstrate here that both caveolae/lipid-rafts- and clathrin-coated-pits-mediated pathways were involved in agonist-induced endocytosis of the cannabinoid type 1 receptor (CB1R) in stably transfected human embryonic kidney (HEK) 293 cells and that the internalized receptors were predominantly sorted into recycling pathway for reactivation. The treatment of CB1 receptors with the low endocytotic agonist Δ9-THC induced a faster receptor desensitization and slower resensitization than the high endocytotic agonist WIN 55,212-2. In addition, the blockade of receptor endocytosis or recycling pathway markedly enhanced agonist-induced CB1 receptor desensitization. Furthermore, co-expression of phospholipase D2, an enhancer of receptor endocytosis, reduced CB1 receptor desensitization, whereas co-expression of a phospholipase D2 negative mutant significantly increased the desensitization after WIN 55,212-2 treatment. These findings provide evidences for the importance of receptor endocytosis in counteracting CB1 receptor desensitization by facilitating receptor reactivation. Moreover, in primary cultured neurons, the low endocytotic agonist Δ9-THC or anandamide exhibited a greater desensitization of endogenous CB1 receptors than the high endocytotic agonist WIN 55,212-2, CP 55940 or 2-arachidonoyl glycerol, indicating that cannabinoids with high endocytotic efficacy might cause reduced development of cannabinoid tolerance to some kind cannabinoid-mediated effects.  相似文献

The 5' nontranslated region (NTR) and the X tail in the 3' NTR are the least variable parts of the hepatitis C virus (HCV) genome and play an important role in the initiation of RNA synthesis. By using subgenomic replicons of the HCV isolates Con1 (genotype 1) and JFH1 (genotype 2), we characterized the genotype specificities of the replication signals contained in the NTRs. The replacement of the JFH1 5' NTR and X tail with the corresponding Con1 sequence resulted in a significant decrease in replication efficiency. Exchange of the X tail specifically reduced negative-strand synthesis, whereas substitution of the 5' NTR impaired the generation of progeny positive strands. In search for the proteins involved in the recognition of genotype-specific initiation signals, we analyzed recombinant nonstructural protein 5B (NS5B) RNA polymerases of both isolates and found some genotype-specific template preference for the 3' end of positive-strand RNA in vitro. To further address genotype specificity, we constructed a series of intergenotypic replicon chimeras. When combining NS3 to NS5A of Con1 with NS5B of JFH1, we observed more-efficient replication with the genotype 2a X tail, indicating that NS5B recognizes genotype-specific signals in this region. In contrast, a combination of the NS3 helicase with NS5A and NS5B was required to confer genotype specificity to the 5' NTR. These results present the first genetic evidence for an interaction between helicase, NS5A, and NS5B required for the initiation of RNA synthesis and provide a system for the specific analysis of HCV positive- and negative-strand syntheses.  相似文献

The etiologic agent of Lyme disease, Borrelia burgdorferi, is capable of circumventing the immune defense of a variety of potential vertebrate hosts. Previous work has shown that interaction of host-derived complement regulators, factor H and factor H-like protein 1 (FHL-1), with up to five complement regulator-acquiring surface proteins (CRASPs) expressed by resistant B. burgdorferi sensu lato isolates conferred complement resistance. In addition expression of CRASP-1 is directly correlated with complement resistance of Borrelia species. This work describes the functional characterization of BbCRASP-1 as the dominant factor H and FHL-1-binding protein of B. burgdorferi. The corresponding gene, zs7.a68, is located on the linear plasmid lp54 and is different from factor H-binding Erp proteins that are encoded by genes localized on circular plasmids (cp32). Deletion mutants of BbCRASP-1 were generated, and a high affinity binding site for factor H and FHL-1 was mapped to the C terminus of BbCRASP-1. Similarly, the predominant binding site of factor H and FHL-1 was localized to the short consensus repeat 7. Factor H and FHL-1 maintain their cofactor activity for factor I-mediated C3b inactivation when bound to BbCRASP-1, and factor H is up to 6-fold more efficient in mediating C3b conversion than FHL-1. In conclusion, BbCRASP-1 (i). binds the host complement regulators factor H and FHL-1 with high affinity, (ii). is the key molecule of the complement resistance of spirochetes, and (iii). is distinct from the Erp protein family. Thus, BbCRASP-1 most likely contributes to persistence of B. burgdorferi and to pathogenesis of Lyme disease.  相似文献

The springtail Megaphorura arctica (Onychiuridae: Collembola) inhabits the arctic and sub-arctic parts of the northern hemisphere where it on a seasonal basis will be exposed to severe cold and desiccating conditions. In the present study we compared how traits of stress resistance differed between two populations of M. arctica that were collected at a high arctic site (Spitsbergen) and a sub-arctic site (Akureyri, Iceland) with contrasting thermal environments. In addition we investigated how cold and desiccation affected the phospholipid fatty acid composition of M. arctica from Spitsbergen. The springtails from Spitsbergen were the most cold tolerant and this was linked to an almost three times higher level of trehalose accumulation during cryoprotective dehydration (15% and 5% of tissue dry weight in the Spitsbergen and Iceland populations, respectively). Although cryoprotective dehydration is intimately related to desiccation stress it was shown that M. arctica had a higher mortality when dehydrated over ice (−10 or −20 °C) than when dehydrated at temperatures above 1 °C. Thus, survival was lower after exposure to −10 °C than after exposure to a relative humidity of 91.2% RH at +1 °C although both treatments led to the same level of dehydration. Exposure to both cold (−10 and −20 °C) and desiccation at +1 °C caused significant changes in the phospholipid fatty acid composition with some similarities. These changes included a decrease in average chain length of the fatty acids due primarily to an increase in the phospholipid fatty acids 16:0 and a decrease in 18:3 and 20:4ω6.  相似文献

AbstractThe p53 protein family is the most studied protein family of all. Sequence analysis and structure determination have revealed a high similarity of crucial domains between p53, p63 and p73. Functional studies, however, have shown a wide variety of different tasks in tumor suppression, quality control and development. Here we review the structure and organization of the individual domains of p63 and p73, the interaction of these domains in the context of full-length proteins and discuss the evolutionary origin of this protein family. Immunoreactive dynorphin in the neurointermediate pituitary of rats was found to consist of four different molecular weight forms. The three larger molecular weight forms, with apparent molecular weights of 4800, 3200, and 1700, constituted more than 80% of the total dynorphin immunoreactivity, and each liberated leucine-enkephalin but not alpha-N-acetyl-leucine-enkephalin upon enzymatic treatment with trypsin followed by carboxypeptidase B. Only a minor portion of the smallest dynorphin-related molecular weight form, dynorphin-(1-8), released alpha-N-acetyl-leucine-enkephalin upon enzymatic cleavage. This suggests that the vast majority of dynorphin-related peptides in the rat neurointermediate pituitary is not alpha-N-acetylated. The exceptionally high opiate-like activity of the molecular weight 1700 dynorphin suggests that this dynorphin-related opioid peptide may constitute the major part of opioid activity in the neurointermediate pituitary of rats.  相似文献
