Demonstration of 125I-labelled thrombin binding platelet proteins by use of crossed immunoelectrophoresis and autoradiography

A possible receptor for thrombin on the platelet membrane has been identified. Whole platelets were treated with 125I-labelled thrombin followed by washing of the platelets, solubilization in Triton X-100, crossed immunoelectrophoresis and autoradiography. A heavily labelled antigen which migrated slightly more slowly than albumin was observed. No corresponding arc was seen on the same immunoplate when stained with Coomassie brilliant blue, indicating that the antigen possessed weak antigenic properties and/or was present in very small amounts. When 125I-labelled thrombin that had been inactivated by phenylmethylsulphonyl fluoride was used, no such labelled arc was seen. The radiolabelled immunoprecipitate does not represent any of the antigens identified hitherto in the immunoelectrophoretic patterns obtained with platelets or platelet material. The electrophoretic mobility of the antigen was influenced neither by neuraminidase treatment of the platelets prior to the 125I-labelled thrombin exposure nor by inclusion of concanavalin A, wheat-germ lectin or lentil lectin in the gel during the first-dimension electrophoresis. This suggests that the antigen does not represent a glycoprotein. Upon subcellular fractionation the radioactively labelled arc was observed in the cytosol fraction following crossed immunoelectrophoresis and autoradiography. Analysis of the secreted proteins after induction of the release reaction with 125I-labelled thrombin revealed labelling of immunoprecipitates representing thrombospondin, albumin and the 'line' form of platelet factor 4. This confirms that stable complexes of 125I-labelled thrombin and platelet proteins can exist in the presence of Triton X-100 and during electrophoresis.     

Zur Feinstruktur und Histochemie des Kiemendarmes und der „Leber“ von Branchiostoma lanceolatum (Pallas)

Zusammenfassung Leberblindsack, Epibranchialrinne und Endostyl von Branchiostoma lanceolatum wurden enzymhistochemisch und elektronenmikroskopisch untersucht. Folgende Fermente wurden nachgewiesen: alkalische und saure Phosphatase, ATP-ase, Aminopeptidase, -Naphthyl-acetat- und Indoxyl-acetat-Esterase, Glucosaminidase, Arylsulfatase, NADH- und NADPH-Diaphorase. Die Epibranchialrinne erwies sich als besonders enzymreich.Die Epithelzellen des Leber-Blindsackes besitzen basal zahlreiche Fetteinschlüsse und oberhalb des Kerns große Granula, die vermutlich Verdauungsenzyme enthalten. Das Epithel der Epibranchialrinne enthält z.T. basal sehr große Vakuolen sowie apikale Sekretionsgranula.Im Endostyl lassen sich verschiedene Zonen nachweisen. In den esterasereichen Zonen 2 und 4/5 finden sich Hinweise auf Proteinsynthese (granuläres E.R. und Sekretionsgranula, bei denen es sich zum Teil um Schleimballen handelt). Zone 4 ist weiterhin besonders durch ihren Gehalt an Lipideinschlüssen gekennzeichnet. In den an saurer Phosphatase reichen Zonen 1, 3 und 6 sind die zilientragenden Zellen in charakteristischen Säulen angeordnet.

Fine structure and histochemistry of the pharynx and the hepatic caecum of Branchiostoma lanceolatum (Pallas)

Summary Cytochemical and ultrastructural studies have been carried out on the hepatic caecum, hyperpharyngeal groove and endostyle of Branchiostoma lanceolatum. The following enzymes have been demonstrated: acid and alkaline phosphatase, ATP-ase, aminopeptidase, -naphthyl-acetate- and indoxyl-acetate esterase, glucosaminidase, arylsulphatase, NADH-and NADPH-diaphorase. The hyperpharyngeal groove proved to be particularly rich in enzymes.The epithelial cells of the hepatic caecum possess at their bases numerous lipid droplets and in the supranuclear cytoplasm granules which presumably contain digestive enzymes.The epithelium of the hyperpharyngeal groove is characterized by large basal vacuoles and apical secretion granules.The electron microscope confirms the existence of several cell zones in the endostyle. The cells of the esterase rich zones 2 and 4/5 possess a well developed system of granulated E. R. as well as globular inclusions which presumably represent mucus droplets. In zone 4 in addition a great number of lipid inclusions and specific electron dense secretion granules occur. In the acid phosphatase rich zones 1, 3 and 6 the cilia-bearing cells are arranged in typical columns.

Zusammenfassung Im Vergleich zwischen Literatur und eigenem Fall von Bloom-Syndrom werden Erörterungen zum Erbgang sowie zur Diagnostik des Minderwuchses und der Hauterscheinungen angestellt. Es wird Stellung genommen zur Bedeutung von Mangelgeburt, Immunglobulindefizienz und cytogenetischen Befunden und auf gleichartige Chromosomen-anomalien bei Fanconi-Anämie und akuten Leukämien hingewiesen.

Beitrag zur Histopathologie des Halsgrenzstranges bei der Raynaudschen Erkrankung

Zusammenfassung Mit der Bielschowsky-Methode wurden an dem von Herrn Obermedizinalrat Dr. K. Lange operativ entfernten sympathischen Halsganglion einer 41jährigen an Raynaudscher Erkrankung leidenden Patientin schwerste krankhafte Veränderungen festgestellt. Die überwiegende Mehrzahl aller Ganglienzellen ist pathologisch verändert.Die Ganglienzellen weisen vielfach eine außerordentliche Fortsatzdisharmonie auf. Ausgedehnte Wucherungsprozesse im Bereich des pericellulären Hüllplasmodiums lassen sich beobachten.Die großen Fortsätze der Ganglienzellen sind häufig in ihrem Inneren ausgehöhlt oder fallen in Einzelfibrillen auseinander. Andere Fortsätze tragen kugelige Endgebilde, deren Auftreten als pathologische Reizerscheinung zu werten ist.Alle erdenklichen krankhaften Formen von Kernveränderungen an den Ganglienzellen kommen zu Gesicht.Das gehäufte Vorkommen mehrkerniger Ganglienzellen mit den sich an ihnen abspielenden pathologischen Veränderungen stellt einen weiteren bei der Raynaudschen Erkrankung erscheinenden degenerativen Prozeß im sympathischen Ganglion dar.Die Hyperplasie feinster Nervenfäserchen in der Nähe erkrankter Ganglienzellen sowie die sog. Knötchenbildungen sind vielleicht Ausdruck einer Störung in den Wechselwirkungen zwischen Ganglienzellen und ihrem Hüllplasmodium.Akut entzündliche Erscheinungen im bindegewebigen Interstitium des Ganglions und infiltratähnliche Kernansammlungen sind mehrfach anzutreffen.     

Induction of the yeast alpha-specific STE3 gene by the peptide pheromone a-factor

The yeast Saccharomyces cerevisiae exhibits two mating types, a and alpha. Efficient mating of a and alpha cells requires the action of peptide pheromones secreted by each cell type. For example, a cells secrete a-factor, which alters the physiology of alpha cells, thereby preparing those cells for mating. To investigate the mechanism by which the pheromones act on the target cells, we have examined the effect of a-factor on expression of the STE3 gene, a gene which is required for mating by alpha cells and which is expressed only in alpha cells. We have monitored STE3 expression by two assays: RNA production from the chromosomal STE3 locus and beta-galactosidase activity produced from a plasmid-borne STE3-lacZ gene fusion. By both assays we show that a-factor induces a rapid increase in STE3 expression. Induction of STE3 RNA occurs even if protein synthesis is blocked by cycloheximide. Using temperature-sensitive cell division cycle mutants, we have also shown that induction occurs in cells arrested at several discrete positions in the cell cycle. These results demonstrate (1) that induction of STE3 expression by a-factor is a primary response to the pheromone, and (2) that alpha cells are capable of responding to a-factor regardless of their position in the cell cycle.

The extraction of high-molecular-mass DNA from hair shafts

A simple and efficient method is presented for the extraction of DNA from hair shafts. DNA preparations obtained by this approach can be made amenable to restriction enzyme digestion, thereby allowing further molecular biological analysis.
