Molecular heterogeneity of adherens junctions   总被引:53,自引:39,他引:14       下载免费PDF全文
We describe here the subcellular distributions of three junctional proteins in different adherens-type contacts. The proteins examined include vinculin, talin, and a recently described 135-kD protein (Volk, T., and B. Geiger, 1984, EMBO (Eur. Mol. Biol. Organ.) J., 10:2249-2260). Immunofluorescent localization of the three proteins indicated that while vinculin was ubiquitously present in all adherens junctions, the other two showed selective and mutually exclusive association with either cell-substrate or cell-cell adhesions. Talin was abundant in focal contacts and in dense plaques of smooth muscle, but was essentially absent from intercellular junctions such as intercalated disks or adherens junctions of lens fibers. The 135-kD protein, on the other hand, was present in the latter two loci and was apparently absent from membrane-bound plaques of gizzard or from focal contacts. Radioimmunoassay of tissue extracts and immunolabeling of cultured chick lens cells indicated that the selective presence of talin and of the 135-kD protein in different cell contacts is spatially regulated within individual cells. On the basis of these findings it was concluded that adherens junctions are molecularly heterogeneous and consist of at least two major subgroups. Contacts with noncellular substrates contain talin and vinculin but not the 135-kD protein, whereas their intercellular counterparts contain the latter two proteins and are devoid of talin. The significance of these results and their possible relationships to contact-induced regulation of cell behavior are discussed.  相似文献

Although nutrient stress is known to alter partitioning between shoots and roots, the physiological basis for the phenomenon is unresolved. Experiments were conducted to examine assimilation of 15NO3 by N-stressed plants and to determine whether apparent changes in assimilation in the root contributed to alterations in whole-plant partitioning of reduced-N. Tobacco plants (Nicotiana tabacum L. cv. NC 2326) were exposed to a low concentration of NO3? in solution (80 μM) for 9 days to effect a N-stress response. Exposure of plants to 1000 μM15NO3? for 12 h on selected days revealed that roots of N-stressed plants developed an increased capacity to absorb NO3?, and accumulation of reduced-15N in the root increased to an even greater extent. When plants were exposed to 80 or 1000 μM15NO3? in steady-state, 15NO3? uptake over a 12 h period was noticeably restricted at the lower concentration, but a larger proportion of the absorbed 15N still accumulated as reduced-15N in the root. The alteration in reduced-15N partitioning was maintained in N-stressed plants during the subsequent 3-day “chase” period when formation of insoluble reduced-15N in the root was quantitatively related to the disappearance of 15NO3? and soluble reduced-15N. The results indicate that increased assimilation of absorbed NO3?, in the root may contribute significantly to the altered reduced-N partitioning which occurs in N-stressed plants.  相似文献

Roots of decapitated maize seedlings (Zea mays L.) were exposed for 12 hours to 1.0 millimolar KNO3 (98.5 atom per cent 15N) in the presence and absence (control) of 0.1 millimolar p-fluorophenylalanine (FPA), an analog of the amino acid phenylalanine. FPA decreased nitrate uptake but had little effect on potassium uptake. In contrast, accumulation of both ions in the xylem exudate was greatly restricted. The proportion of reduced 15N-nitrogen that was translocated at each time was also restricted by FPA. These observations are interpreted as indicating that synthesis of functional protein(s) is required for nitrate uptake and for transport of potassium, nitrate, and reduced-15N from xylem parenchyma cells into xylem elements. The effect of FPA on nitrate reduction is less clear. Initially, FPA limited nitrate reduction more than nitrate uptake, but by 8 hours the cumulative reduction of entering nitrate was similar (~35%) in both control and FPA-treated roots. A relationship between nitrate uptake and nitrate reduction is implied. It is suggested that nitrate influx regulates the proportion of nitrate reductase in the active state, and thereby regulates concurrent nitrate reduction in decapitated maize seedlings.  相似文献

The influence of nitrogen stress on net nitrate uptake resulting from concomitant 15NO3 influx and 14NO3 efflux was examined in two 12-day-old inbred lines of maize. Plants grown on 14NO3 were deprived of nitrogen for up to 72 hours prior to the 12th day and then exposed for 0.5 hour to 0.15 millimolar nitrate containing 98.7 atom% 15N. The nitrate concentration of the roots declined from approximately 100 to 5 micromolar per gram fresh weight during deprivation, and 14NO3 efflux was linearly related to root nitrate concentration. Influx of 15NO3 was suppressed in nitrogen-replete plants and increased with nitrogen deprivation up to 24 hours, indicating a dissipation of factors suppressing influx. Longer periods of nitrogen-deprivation resulted in a decline in 15NO3 influx from its maximal rate. The two inbreds differed significantly in the onset and extent of this decline, although their patterns during initial release from influx suppression were similar. Except for plants of high endogenous nitrogen status, net nitrate uptake was largely attributable to influx, and genetic variation in the regulation of this process is implied.  相似文献

Vancomycin resistance plasmids in enterococci carry the genes vanH and vanA, which encode enzymes catalyzing, respectively, the reduction of 2-keto acids to 2-D-hydroxy acids and the addition of D-hydroxy acids to D-alanine. It has therefore been postulated that resistant cells produce peptidoglycan precursors that terminate in the depsipeptide D-alanine-2-D-hydroxy acid rather than the dipeptide D-alanine-D-alanine, thus preventing vancomycin binding (M. Arthur, C. Molinas, T. D. H. Bugg, G. D. Wright, C. T. Walsh, and P. Courvalin, Antimicrob. Agents Chemother. 36:867-869, 1992). In the present work, a cytoplasmic peptidoglycan precursor was isolated from vancomycin-resistant Enterococcus faecalis and analyzed by mass spectrometry, which suggested the structure UDP-N-acetyl-muramyl-L-Ala-D-Glu-L-Lys-D-Ala-D-lactate.  相似文献

To increase the understanding of poplar and willow perennial woody crops and facilitate their deployment for the production of biofuels, bioproducts, and bioenergy, there is a need for broadscale yield maps. For national analysis of woody and herbaceous crops production potential, biomass feedstock yield maps should be developed using a common framework. This study developed willow and poplar potential yield maps by combining data from a network of willow and poplar field trials and the modeling power of PRISM‐ELM. Yields of the top three willow cultivars across 17 sites ranged from 3.60 to 14.6 Mg ha?1 yr?1 dry weight, while the yields from 17 poplar trials ranged from 7.5 to 15.2 Mg ha?1 yr?1. Relationships between the environmental suitability estimates from the PRISM‐ELM model and results from field trials had an R2 of 0.60 for poplar and 0.81 for willow. The resulting potential yield maps reflected the range of poplar and willow yields that have been reported in the literature. Poplar covered a larger geographic range than willow, which likely reflects the poplar breeding efforts that have occurred for many more decades using genotypes from a broader range of environments than willow. While the field trial data sets used to develop these models represent the most complete information at the time, there is a need to expand and improve the model by monitoring trials over multiple cutting cycles and across a broader range of environmental gradients. Despite some limitations, the results of these models represent a dramatic improvement in projections of potential yield of poplar and willow crops across the United States.  相似文献

Numerous branched plasmodesmata (pd) are present between bundle-sheath cells (BSCs) and specialized companion cells known as intermediary cells (ICs) in the minor-vein phloem of melon (Cucumis melo L.) and squash (Cucurbita pepo L.). These pd were found to be secondary, i.e., they form across existing walls. Sink, sink-source transition, and source tissues were sampled from developing and mature leaves. In sink tissue, IC precursors divide to produce the two to four ICs and associated sieve elements which are present by the time of the sink-source transition. Plasmodesmata along the interface between the IC precursor and adjacent BSCs in sink tissue are unbranched and few in number. Before the leaf tissue undergoes the sink-source transition, the number of pd channels (individual branches of pd) becomes more numerous. This increase in number of pd channels occurs at least in part and perhaps entirely by branching, resulting in more channels on the IC-side than on the BSC-side. In melon there is a 12-fold increase in the number of pd channels within the IC-side of the interface and a corresponding 9-fold increase in pd channels within the BSC-side. Thus, secondary pd form by the time of the sink-source transition and may be involved in phloem loading and photoassimilate export. Use of spectral analysis to test hypotheses on the origin of pinnipeds   总被引:10,自引:4,他引:6  
The evolutionary origin of the pinnipeds (seals, sea lions, and walruses) is still uncertain. Most authors support a hypothesis of a monophyletic origin of the pinnipeds from a caniform carnivore. A minority view suggests a diphyletic origin with true seals being related to the mustelids (otters and ferrets). The phylogenetic relationships of the walrus to other pinniped and carnivore families are also still particularly problematic. Here we examined the relative support for mono- and diphyletic hypotheses using DNA sequence data from the mitochondrial small subunit (12S) rRNA and cytochrome b genes. We first analyzed a small group of taxa representing the three pinniped families (Phocidae, Otariidae, and Odobenidae) and caniform carnivore families thought to be related to them. We inferred phylogenetic reconstructions from DNA sequence data using standard parsimony and neighbor-joining algorithms for phylogenetic inference as well as a new method called spectral analysis (Hendy and Penny) in which phylogenetic information is displayed independently of any selected tree. We identified and compensated for potential sources of error known to lead to selection of incorrect phylogenetic trees. These include sampling error, unequal evolutionary rates on lineages, unequal nucleotide composition among lineages, unequal rates of change at different sites, and inappropriate tree selection criteria. To correct for these errors, we performed additional transformations of the observed substitution patterns in the sequence data, applied more stringent structural constraints to the analyses, and included several additional taxa to help resolve long, unbranched lineages in the tree. We find that there is strong support for a monophyletic origin of the pinnipeds from within the caniform carnivores, close to the bear/raccoon/panda radiation. Evidence for a diphyletic origin was very weak and can be partially attributed to unequal nucleotide compositions among the taxa analyzed. Subsequently, there is slightly more evidence for grouping the walrus with the eared seals versus the true seals. A more conservative interpretation, however, is that the walrus is an early, but not the first, independent divergence from the common pinniped ancestor.   相似文献
