The effect of hemorrhagic arterial hypotension on local cerebral glucose metabolism was studied on 33 rats. The mean arterial pressure was set with the aid of a reservoir at 80, 60, 50 and 40 mmHg pressures. Local cerebral glucose utilization was measured with the 14C-2-D-deoxyglucose accumulation autoradiographic technique. Local glucose consumption decreased somewhat in cortical structures when mean arterial pressure was reduced to 60-50 mmHg. Further decrease in mean arterial pressure to 50-40 mmHg caused inhomogeneity of tissue metabolism. Columns and patches of high glucose consumption interchanged with areas of very low glucose consumption in most telencephalic and cerebellar gray matter structures. Brain stem and white matter structures seem to be less sensitive to decreased mean arterial pressure in the range studied. We found a decrease in glucose utilization rather than an increase with decreasing mean arterial pressure down to 60-50 mmHg (in the range of the autoregulation of cerebral circulation). This finding makes it improbable that autoregulation would be connected with elevated anaerobic metabolism of the tissue. Patchy areas and columns of high glucose consumption found at 50-40 mmHg in all probability reflect areas of increased anaerobic metabolism of glucose. Here, circulation was not enough to transport adequate quantity of oxygen to the tissue, but still it transported relative large amount of glucose. Columns and patches of very low glucose consumption should reflect areas, where circulation was inadequate to transport both enough glucose and oxygen.  相似文献

The level of anhydrotetracycline oxygenase (an enzyme catalyzing the penultimate reaction in the biosynthesis of tetracycline) inStreptomyces aureofaciens was substantially influenced by the amount of inorganic phosphate and by the presence of benzyl thiocyanate in the cultivation medium. Phosphate decreased the specific activity of the enzyme, particularly when added to a growing culture. On the other hand, benzyl thiocyanate increased the specific activity of the enzyme. Its effect was most conspicuous in the growth phase. The effect of benzyl thiocyanate was more pronounced in the low-production strain than in the producing variant. Inorganic phosphate and benzyl thiocyanate did not influence the enzyme activityin vitro. Phosphate added to the growing cultures was readily absorbed by the cells. During this time the enzyme synthesis was repressed, derepression occurred only after exhaustion of phosphate from the medium. The stimulatory efect of benzyl thiocyanate on the enzyme synthesis was not reversed by the inorganic phosphate added.  相似文献

In the seeds ofAllium altaicun (Pall.)Reyse a set of enzymes was found, metabolizing choline esters, composed of active choline esterases and choline acetyltransferase. Choline esterase cleaving acetylcholine occurs in five isoenzymes. The enzyme preparation hydrolyses strongly acetylthiocholine and sinapine, but weakly butyrylthiocholine (20%) in comparison with acetylthiocholine. The hydrolysis of the substrates mentioned is inhibited by physostigmine and neostigmine, but it is not inhibited by the specific inhibitor of acetylcholine esterase (BW 284 C51). In addition to hydrolytic activity a strong catalytic activity of choline acetyltransferase was also observed during the synthesis of sinapine from sinapic acid and choline. The detection of the mentioned enzymes in some representatives of theAllium genus indicates that choline esterases are more widely distributed in monocotyledons than previously assumed.  相似文献

FUdR 10?4 M was applied together with [3H] TdR on the growing barley embryos cultivated in the nutrient solution. Samples were fixed 1–4 h after the onset of the treatment and microautoradiograms were prepared. All the mitoses were unlabelled, while more than 150 autoradiographic grains were found above labelled interphase nuclei; no cells that terminated S phase at the onset of the treatment passed through entire G2 and reached mitosis during the 4 h duration of the treatment. Chromosomal fragments in anaphases appeared the first hour after the onset of the treatment. Their frequency increased from the first to the second hour and remained almost constant from the second to the fourth hour. FUdR induces chromosomal fragments in barley root meristems also in G2 phase of the mitotic cycle.  相似文献

Ten taxa of the genusPisum were examined by disc electrophoresis in gels according to Davis and to Reisfeldet al. For evaluation of band patterns the Jaccard Index was applied. The results in both types of gels show thatPisum abyssinicum and especiallyP. fulvum have biochemically a relatively isolated position.Pisum elatius and its subspeciescaspicum andpalestinicum form a subgroup withP. cinereum;P. sativum var.zeylanicum and cv. Jupiter form another subgroup withP. syriacum. Our results are in good agreement with the results of Przybylskaet al. (with the exception ofP. cinereum) and also with immunoelectrophoretic analyses performed by Turkováet al. (1980), with the same exception.  相似文献

Optimal conditions for the submerged cultivation ofStreptomyces glomeratus 3980, producer of the anthracycline antibiotics beromycins, and its variants were sought in media with glucose, soybean meal and salts differing in the content of ammonium sulphate. As compared with the original activity of the strain the antibiotic titre of some variants increased about 12 times on increasing the glucose concentration from 3 to 5 %, or on omitting CaCO3 from the medium (i.e. under conditions leading to an increased production of propionic acid and suppression of production of the melanin-like pigment). In melanin-less variants accumulating propionate even under standard conditions the activity increased about 18–40 times in the medium with 3 % glucose and 0.2 % CaCO3 under conditions of more intensive aeration (i.e. under conditions when no propionic acid accumulated). Individual strains also differed in the requirement for (NH4)2SO4 in the medium, their response to changes of volume of the vegetative inoculum and sensitivity to MgSO4.7H2O. The biosyiithetic activity of all strains was inhibited by the addition of ZnSO4.7H2O or CaCl2 and substitution of glucose with starch, lactose or sucrose.  相似文献
