On the basis of allozyme and cultivation data, and of additional herbarium material, a taxonomic and nomenclatural revision ofTaraxacum sect.Piesis A.J. Richards exKirschner et?těpánek is provided. The section is made up of halophilous, sexually reproducing taxa. InT. stenocephalum Boiss. etKotschy,T. pindicum Kirschner et?těpánek, sp. nov., andT. perenne Kirschner et?těpánek, sp. nov., a tetraploid chromosome number has been recorded, representing the only known case of sexuality at the tetraploid level in the genus. The complex ofT. stenocephalum, includes some geographically and morphologically extreme populations treated as subspecies: subsp.gumusanicum (Soest)Kirschner et?těpánek, comb. nov., subsp.magnum Kirschner et?těpánek, subsp. nov., and subsp.daralagesicum (Schischk.)Kirschner et?těpánek, comb. nov. In addition toT. bessarabicum (Hornem.)Hand.-Mazz., a widely distributed Eurasian species,T. stenocephalum, a complex centred in Transcaucasia and Anatolia, andT. pachypodum H. Involvement of mammalian mitochondrial glycerophosphate dehydrogenase (mGPDH, EC 1.1.99.5) in reactive oxygen species (ROS) generation was studied in brown adipose tissue mitochondria by different spectroscopic techniques. Spectrofluorometry using ROS-sensitive probes CM-H2DCFDA and Amplex Red was used to determine the glycerophosphate- or succinate-dependent ROS production in mitochondria supplemented with respiratory chain inhibitors antimycin A and myxothiazol. In case of glycerophosphate oxidation, most of the ROS originated directly from mGPDH and coenzyme Q while complex III was a typical site of ROS production in succinate oxidation. Glycerophosphate-dependent ROS production monitored by KCN-insensitive oxygen consumption was highly activated by one-electron acceptor ferricyanide, whereas succinate-dependent ROS production was unaffected. In addition, superoxide anion radical was detected as a mGPDH-related primary ROS species by fluorescent probe dihydroethidium, as well as by electron paramagnetic resonance (EPR) spectroscopy with DMPO spin trap. Altogether, the data obtained demonstrate pronounced differences in the mechanism of ROS production originating from oxidation of glycerophosphate and succinate indicating that electron transfer from mGPDH to coenzyme Q is highly prone to electron leak and superoxide generation.  相似文献

Goal, Scope and Background The energy systems included in the ecoinvent database v1.1 describe the situation around year 2000 of Swiss and Western European power plants and boilers with the associated energy chains. The addressed nuclear systems concern Light Water Reactors (LWR) with mix of open and closed fuel cycles. The system model ‘Natural Gas’ describes production, distribution, and combustion of natural gas. Methods Comprehensive life cycle inventories of the energy systems were established and cumulative results calculated within the ecoinvent framework. Swiss conditions for the nuclear cycle were extrapolated to major nuclear countries. Long-term radon emissions from uranium mill tailings have been estimated with a simplified model. Average natural gas power plants were analysed for different countries considering specific import/export of the gas, with seven production regions separately assessed. Uncertainties have been estimated quantitatively. Results and Discussion Different radioactive emission species and wastes are produced from different steps of the nuclear cycle. Emissions of greenhouse gases from the nuclear cycle are mostly from the upstream chain, and the total is small and decreasing with increasing share of centrifuge enrichment. The results for natural gas show the importance of transport and low pressure distribution network for the methane emissions, whereas energy is mostly invested for production and long-distance pipeline transportation. Because of significant differences in power plant efficiencies and gas supply, country specific averages differ greatly. Conclusion The inventory describes average worldwide supply of nuclear fuel and average nuclear reactors in Western Europe. Although the model for nuclear waste management was extrapolated from Swiss conditions, the ranges obtained for cumulative results can represent the average in Europe. Emissions per kWh electricity are distributed very differently over the natural gas chain for different species. Modern combined cycle plants show better performance for several burdens like cumulative greenhouse gas emissions compared to average plants. Recommendation and Perspective Comparison of country-specific LWRs or LWR types on the basis of these results is not recommended. Specific issues on different strategies for the nuclear fuel cycle or location-specific characteristics would require extension of analysis. Results of the gas chain should not be directly applied to areas other than those modelled because emission factors and energy requirements may differ significantly. A future update of inventory data should reconsider production and transport from Russia, as it is a major producer and exporter to Europe. The calculated ranges of uncertainty factors in ecoinvent provide useful information but they are more indications of uncertainties rather than strict 95% intervals, and should therefore be applied carefully.  相似文献

A collection of 34 lactococcal strains were characterized using the polymerase chain reaction (PCR) for the acmA gene, and for the 16S rDNA gene, and DNA fingerprinting methods for randomly amplified polymorphic DNA (RAPD) and repetitive extragenic palindrome-PCR (rep-PCR). PCR experiments corroborated the genotypic identification of Lactococcus lactis strains by RAPD; rep-PCR did not distinguish between L. lactis subspecies. In some cases, phenotypic classification of L. lactis subspecies did not correlate with genotypic characterization.  相似文献

In termites, juvenile hormone plays a key role in soldier differentiation. To better understand the evolutionary origin of the soldiers, we studied the external and inner morphology of pseudergate-soldier intercastes and neotenic-soldier intercastes formed artificially by the application of juvenile hormone analogue in Prorhinotermes simplex. A majority of these intercastes had a soldier phenotype, whereas the inner anatomy had an intermediary form between two castes or a form specific to intercastes. Our experiments showed that traits of neotenics and soldiers can be shared by the same individuals, although such individuals do not exist naturally in P. simplex, and they have not been reported in other species but in some Termopsidae. Our results reinforce the hypothesis that soldiers may have emerged from soldier neotenics during the evolution of termites.  相似文献

Macrofungal species richness generally increases with forest continuity as does the richness of so-called ancient forest plant species (AFS). Based on this assumption, we examined the ability of AFS to indicate macrofungal diversity in six study areas covering a range of elevations and environments in the Czech Republic. In total, we used data from 106 sampling plots (2,500 m2 each) distributed over six types of forest stands reflecting different intensities and temporal stages of forest management. Species composition of vascular plants and macrofungi was recorded by a single inventory and regular 2-year monitoring, respectively. In total, we found 71 AFS and 1,413 macrofungal species, of which 150 were red-listed macrofungal species. We documented that AFS show potential for being used in the prediction of macrofungi species richness, including endangered species, at the local scale (α-diversity). Additionally, we found significant differences in macrofungal species richness depending on study area and type of forest management, which did not, however, derogate the effect of AFS. Spatial congruence between species composition of AFS and macrofungi communities (β-diversity) increased with forest age and decreased with intensity of forest management. If we consider the simplicity of monitoring AFS in comparison to regular monitoring of macrofungi, we found a widely usable tool for estimating macrofungal diversity in all dominant types of managed forest in central Europe. However, we should be aware of the limited ability of AFS to capture macrofungal diversity across a broader spatial context (γ-diversity), especially in areas with a low diversity of AFS.  相似文献

A novel antimicrobial peptide, designated macropin (MAC‐1) with sequence Gly‐Phe‐Gly‐Met‐Ala‐Leu‐Lys‐Leu‐Leu‐Lys‐Lys‐Val‐Leu‐NH2, was isolated from the venom of the solitary bee Macropis fulvipes. MAC‐1 exhibited antimicrobial activity against both Gram‐positive and Gram‐negative bacteria, antifungal activity, and moderate hemolytic activity against human red blood cells. A series of macropin analogs were prepared to further evaluate the effect of structural alterations on antimicrobial and hemolytic activities and stability in human serum. The antimicrobial activities of several analogs against pathogenic Pseudomonas aeruginosa were significantly increased while their toxicity against human red blood cells was decreased. The activity enhancement is related to the introduction of either l ‐ or d ‐lysine in selected positions. Furthermore, all‐d analog and analogs with d ‐amino acid residues introduced at the N‐terminal part of the peptide chain exhibited better serum stability than did natural macropin. Data obtained by CD spectroscopy suggest a propensity of the peptide to adopt an amphipathic α‐helical secondary structure in the presence of trifluoroethanol or membrane‐mimicking sodium dodecyl sulfate. In addition, the study elucidates the structure–activity relationship for the effect of d ‐amino acid substitutions in MAC‐1 using NMR spectroscopy. Aim Invasive alien species (IAS) are recognized as major drivers of biodiversity loss, but few causal relationships between IAS and species declines have been documented. In this study, we compare the distribution (Belgium and Britain) and abundance (Belgium, Britain and Switzerland) of formerly common and widespread native ladybirds before and after the arrival of Harmonia axyridis, a globally rapidly expanding IAS. Location Europe Methods We used generalized linear mixed‐effects models (GLMMs) to assess the distribution trends of eight conspicuous and historically widespread and common species of ladybird within Belgium and Britain before and after the arrival of H. axyridis. The distribution data were collated largely through public participatory surveys but verified by a recognized expert. We also used GLMMs to model trends in the abundance of ladybirds using data collated through systematic surveys of deciduous trees in Belgium, Britain and Switzerland. Results Five (Belgium) and seven (Britain) of eight species studied show substantial declines attributable to the arrival of H. axyridis. Indeed, the two‐spot ladybird, Adalia bipunctata, declined by 30% (Belgium) and 44% (Britain) over 5 years after the arrival of H. axyridis. Trends in ladybird abundance revealed similar patterns of declines across three countries. Main conclusion Together, these analyses show H. axyridis to be displacing native ladybirds with high niche overlap, probably through predation and competition. This finding provides strong evidence of a causal link between the arrival of an IAS and decline in native biodiversity. Rapid biotic homogenization at the continental scale could impact on the resilience of ecosystems and severely diminish the services they deliver.  相似文献

Many woody plants have been recently recognized as major invasive species with serious impacts on species diversity and functioning of invaded ecosystems. We evaluated the global invasion success of temperate trees and shrubs with native ranges in central Europe and explored the role of their native distribution and that of biological traits in determining whether they have become invasive in other parts of the world. Of the 94 species forming the source-area species pool, 27 % are known to be invasive in at least one region of the world. Generalized linear models on phylogenetically non-informed analyses revealed that tall woody plants flowering early in the season, and occupying many grid cells in the native range are significantly more likely to become successful invaders than species not possessing these traits. However, other traits can partly reduce the disadvantages resulting from low regional frequencies in the native range and consequent lower probability of them being introduced elsewhere. Species that do not depend for reproduction on another individual, those pollinated by wind and dispersed by animals are likely to become invasive even if they do not have extensive native distributions. However, of these traits only the regional frequency in the native range remained significant when phylogenetic relationships were taken into account. This indicates that the effect of the regional frequency is generic, valid across all woody species, and fine-tuned by advantageous biological traits inherited from common ancestors, shared by groups of phylogenetically-related species. Traits that only appeared significant in the phylogenetically non-informed analyses should be considered as specific for shrubs and trees of central Europe. Mode of reproduction was only significant in the phylogenetically-informed analysis, indicating that reproduction exclusively by seed favours invasiveness of woody species. From the management perspective, however, the predictive value of some traits is not diminished by them being phylogenetically constrained since we are not interested whether the behaviour of particular species is a result of evolutionary history but how we can treat specific cases of invasion.  相似文献
