Background
Overcoming boundaries is crucial for incursion of alien plant species and their successful naturalization and invasion within protected areas. Previous work showed that in Kruger National Park, South Africa, this process can be quantified and that factors determining the incursion of invasive species can be identified and predicted confidently. Here we explore the similarity between determinants of incursions identified by the general model based on a multispecies assemblage, and those identified by species-specific models. We analyzed the presence and absence of six invasive plant species in 1.0×1.5 km segments along the border of the park as a function of environmental characteristics from outside and inside the KNP boundary, using two data-mining techniques: classification trees and random forests.Principal Findings
The occurrence of Ageratum houstonianum, Chromolaena odorata, Xanthium strumarium, Argemone ochroleuca, Opuntia stricta and Lantana camara can be reliably predicted based on landscape characteristics identified by the general multispecies model, namely water runoff from surrounding watersheds and road density in a 10 km radius. The presence of main rivers and species-specific combinations of vegetation types are reliable predictors from inside the park.Conclusions
The predictors from the outside and inside of the park are complementary, and are approximately equally reliable for explaining the presence/absence of current invaders; those from the inside are, however, more reliable for predicting future invasions. Landscape characteristics determined as crucial predictors from outside the KNP serve as guidelines for management to enact proactive interventions to manipulate landscape features near the KNP to prevent further incursions. Between species, variation in sperm size has been related to male–female coevolution and male–male competition. In contrast, variation within species is poorly understood. A particular case of intraspecific sperm-size variation occurs in sperm-heteromorphic species, where males produce distinct sperm morphotypes, usually only one of which is fertile. This allows to investigate sperm size variation under different selection regimes. Nonfertile morphotypes, whose role is aside from fertilization, may have other functions, and this may be reflected by changes in developmental processes and a different phenotype compared to fertile sperm. We show that the intraspecific coefficient of variation in sperm length is up to four times lower for fertile than nonfertile morphotypes across 150 sperm-heteromorphic species (70 butterfly, 71 moth, 9 diopsid fly species). This is in agreement with a previous study on 11 species in the Drosophila obscura group. Significantly lower variation in fertile than nonfertile sperm morphometry may result from fertilization-related selection for optimal sperm size, novel functions of nonfertile sperm, or from tighter control of fertile sperm development. In the genus Petunia, distinct pollination syndromes may have evolved in association with bee-visitation (P. integrifolia spp.) or hawk moth-visitation (P. axillaris spp). We investigated the extent of congruence between floral fragrance and olfactory perception of the hawk moth Manduca sexta. Hawk moth pollinated P. axillaris releases high levels of several compounds compared to the bee-pollinated P. integrifolia that releases benzaldehyde almost exclusively. The three dominating compounds in P. axillaris were benzaldehyde, benzyl alcohol and methyl benzoate. In P. axillaris, benzenoids showed a circadian rhythm with an emission peak at night, which was absent from P. integrifolia. These characters were highly conserved among different P. axillaris subspecies and P. axillaris accessions, with some differences in fragrance composition. Electroantennogram (EAG) recordings using flower-blends of different wild Petunia species on female M. sexta antennae showed that P. axillaris odours elicited stronger responses than P. integrifolia odours. EAG responses were highest to the three dominating compounds in the P. axillaris flower odours. Further, EAG responses to odour-samples collected from P. axillaris flowers confirmed that odours collected at night evoked stronger responses from M. sexta than odours collected during the day. These results show that timing of odour emissions by P. axillaris is in tune with nocturnal hawk moth activity and that flower-volatile composition is adapted to the antennal perception of these pollinators. 相似文献

The gene man5K encoding the mannanase Man5K from Clostridium cellulolyticum was cloned alone or as an operon with the gene cipC1 encoding a truncated scaffoldin (miniCipC1) of the same origin in the solventogenic Clostridium acetobutylicum. The expression of the heterologous gene(s) was under the control of a weakened thiolase promoter Pthl. The recombinant strains of the solventogenic bacterium were both found to secrete active Man5K in the range of milligrams per liter. In the case of the strain expressing only man5K, a large fraction of the recombinant enzyme was truncated and lost the N-terminal dockerin domain, but it remained active towards galactomannan. When man5K was coexpressed with cipC1 in C. acetobutylicum, the recombinant strain secreted almost exclusively full-length mannanase, which bound to the scaffoldin miniCipC1, thus showing that complexation to the scaffoldin stabilized the enzyme. The secreted heterologous complex was found to be functional: it binds to crystalline cellulose via the carbohydrate binding module of the miniscaffoldin, and the complexed mannanase is active towards galactomannan. Taken together, these data show that C. acetobutylicum is a suitable host for the production, assembly, and secretion of heterologous minicellulosomes. 相似文献

Seed weight is a crucial plant life history trait, determining establishment success and dispersal ability. Especially in
stressful environments, larger seeds may be selected at the expense of seed number, because larger seeds have a better chance
of giving rise to an established offspring. We tested the hypotheses that between related species-pairs and among populations
of single species a similar trend for increasing seed weight with increasing altitude should be present. Firstly, we measured
seed weights from 29 species-pairs, with one species occurring in lowland areas and a congeneric species from high altitudes.
Seeds of the alpine species were 28±8% larger than seeds from lowland species (P<0.01). Compared to the related lowland species, 55% of the alpine species had heavier seeds, 3% (one species) had lighter,
and 41% had seeds of approximately equal weight. Secondly, we compared seed weights among populations of four species from
different habitats and with different life histories. Seeds from between 11 and 34 populations per species were sampled along
altitudinal gradients of 800–1,500 m (ca. 800 m in Scabiosa lucida, ca. 1,000 m in Saxifraga oppositifolia, ca. 1,000 m in Epilobium fleischeri, and ca. 1,500 m in Carex flacca). In all the four species, we found no indication for heavier seeds at higher altitudes. Our results indicate a selection
pressure for species with heavier seeds at higher altitude, but the trend does not seem to operate across all cases. Phylogenetic
constraints may limit the correlation among altitude and seed weight, operating particularly against selection for larger

Floral scent emission and pollinator attraction in two species of <Emphasis Type="Italic">Gymnadenia</Emphasis> (Orchidaceae) 总被引:4,自引:0,他引:4
We investigated scent composition and pollinator attraction in two closely related orchids, Gymnadenia conopsea (L.) R.Br. s.l. and Gymnadenia odoratissima (L.) Rich. in four populations during the day and night. We collected pollinators of both species using hand nets and sampled floral odour by headspace sorption. We analysed the samples by gas chromatography with mass spectrometry to identify compounds and with electroantennographic detection to identify compounds with physiological activity in pollinators. In order to evaluate the attractiveness of the physiologically active compounds, we carried out trapping experiments in the field with single active odour substances and mixtures thereof. By collecting insects from flowers, we caught eight pollinators of G. conopsea, which were members of four Lepidoptera families, and 37 pollinators of G. odoratissima, from five Lepidopteran families. There was no overlap in pollinator species caught from the two orchids using nets. In the scent analyses, we identified 45 volatiles in G. conopsea of which three (benzyl acetate, eugenol, benzyl benzoate) were physiologically active. In G. odoratissima, 44 volatiles were identified, of which seven were physiologically active (benzaldehyde, phenylacetaldehyde, benzyl acetate, 1-phenyl-2,3-butandione, phenylethyl acetate, eugenol, and one unknown compound). In field bioassays using a mixture of the active G. odoratissima compounds and phenylacetaldehyde alone we caught a total of 25 moths, some of which carried Gymnadenia pollinia. A blend of the active G. conopsea volatiles placed in the G. odoratissima population did not attract any pollinators. The two orchids emitted different odour bouquets during the day and night, but G. odoratissima showed greater temporal differences in odour composition, with phenylacetaldehyde showing a significant increase during the night. The species differed considerably in floral odour emission and this differentiation was stronger in the active than non-active compounds. This differentiation of the two species, especially in the emission of active compounds, appears to have evolved under selection for attraction of different suites of Lepidopteran pollinators. 相似文献

Effects of heterogeneous habitat use by cattle on nutrient availability and litter decomposition in soils of an Alpine pasture 总被引:3,自引:1,他引:2
Grazing by free-ranging cattle on Alpine pastures in southern Switzerland creates sharp contrasts in plant species composition between small camp areas, which are grazed intensely and receive most cattle excreta, and surrounding pasture dominated by Nardus stricta, which is only lightly grazed. We hypothesised that these contrasts are maintained by positive feedbacks related to nutrient availability in soil, in that (a) plant material with rapid decomposition and nutrient release decomposes in camp areas and (b) litter decomposition is further stimulated by enhanced nutrient availability in soil. We compared nutrient availability at three camp areas with that in surrounding Nardus vegetation and investigated how the decomposition of plant material from both vegetation types responds to nutrient availability in soil, both in the field (during 14 weeks) and in the laboratory (during 4, 10, and 16 weeks). At all three field sites P availability was significantly enhanced in camp areas, whereas differences in N availability were inconsistent among the three sites. Laboratory incubations indicated that microbial activity after the addition of labile C (cellulose) was limited by P availability in the Nardus vegetation but not in camp areas. The camp-area plant substrate decomposed much faster (81.5% vs. 27.1% ash-free dry mass loss in the field) and released more N and P than the Nardus substrate, which tended to immobilise soil nutrients. However, the decomposition rate of neither substrate was influenced by nutrient availability in soil, both in the field (comparing camp areas and Nardus vegetation) and in the laboratory (comparing incubations with and without N or P fertilisation). We conclude that the contrasting quality of plant substrates contributes to the greater nutrient availability in camp areas (feedback a) but that the latter does not influence the decomposition of in situ plant material (feedback b) because the latter is not nutrient-limited. 相似文献

The ability to express tightly controlled amounts of endogenous and recombinant proteins in plant cells is an essential tool
for research and biotechnology. Here, the inducibility of the soybean heat-shock Gmhsp17.3B promoter was addressed in the moss Physcomitrella patens, using β-glucuronidase (GUS) and an F-actin marker (GFP-talin) as reporter proteins. In stably transformed moss lines, Gmhsp17.3B-driven GUS expression was extremely low at 25 °C. In contrast, a short non-damaging heat-treatment at 38 °C rapidly induced
reporter expression over three orders of magnitude, enabling GUS accumulation and the labelling of F-actin cytoskeleton in
all cell types and tissues. Induction levels were tightly proportional to the temperature and duration of the heat treatment,
allowing fine-tuning of protein expression. Repeated heating/cooling cycles led to the massive GUS accumulation, up to 2.3%
of the total soluble proteins. The anti-inflammatory drug acetyl salicylic acid (ASA) and the membrane-fluidiser benzyl alcohol
(BA) also induced GUS expression at 25 °C, allowing the production of recombinant proteins without heat-treatment. The Gmhsp17.3B promoter thus provides a reliable versatile conditional promoter for the controlled expression of recombinant proteins in

The goal of the study was to monitor the antioxidative effect of stobadine derivative in the conditions of ischemia-reperfusion of laboratory rat kidney tissue. The animals were divided by random selection into 5 groups (n = 10). The treated groups were given stobadine derivate in peroral doses of 5, 10 and 20 mg/kg in 0.5 % solution of Avicel once a day; the placebo group was given only the solution of Avicel. The last group was an intact group (without ischemia-reperfusion and without treatment). After conclusion of medication on the 15th day all animals were subjected to kidney tissue ischemia (60 min.) followed by reperfusion (10 min.). All animals were subsequently exsanquined and single identification of superoxiddismutase, glutathion peroxidase, total antioxidative capacity, and malondialdehyde level in the blood were determined. Kidneys were recovered for histopathological examination. A statistically significant decrease of the superoxiddismutase and statistically significant increase of the glutathione peroxidase catalytic activity in the treated groups compared to the groups of placebo and intact was discovered. There was also a statistically highly significant increase of total antioxidative capacity in the treated groups compared to the groups of placebo and intact. A statistically significant decrease of malondialdehyde level was identified in the treated groups compared to the groups of placebo and intact. The results of biochemical examination show a protective antioxidative effect of stobadine derivative. The results of histopathological examination support this assumption. 相似文献
