The study of novel 2-arylpyrazolo[3,4-c]quinolin-4-(hetero)arylamides, designed as human (h) A(3) adenosine receptor antagonists, is reported. The new derivatives are endowed with nanomolar hA(3) receptor affinity and high selectivity versus hA(1), hA(2A) and hA(2B) receptors. Among the (hetero)aroyl residues introduced on the 4-amino group, the 2-furyl and 4-pyridyl rings turned out to be the most beneficial for hA(3) affinity (K(i)=3.4 and 5.0nM, respectively). An intensive molecular docking study to a rhodopsin-based homology model of the hA(3) receptor was carried out to obtain a 'structure-based pharmacophore model' that proved to be helpful for the interpretation of the observed affinities of the new hA(3) pyrazoloquinoline antagonists.  相似文献

This paper covers genetic and biochemical aspects of mitochondrial bioenergetics dysfunction in hereditary neurological disorders associated with complex I defects. Three types of hereditary complex I dysfunction are dealt with: (i) homozygous mutations in the nuclear genes NDUFS1 and NDUFS4 of complex I, associated with mitochondrial encephalopathy; (ii) a recessive hereditary epileptic neurological disorder associated with enhanced proteolytic degradation of complex I; (iii) homoplasmic mutations in the ND5 and ND6 mitochondrial genes of the complex, cohexistent with mutation in the nuclear PINK1 gene in familial Parkinsonism. The genetic and biochemical data examined highlight different mechanisms by which mitochondrial bioenergetics is altered in these hereditary defects of complex I. This knowledge, besides clarifying molecular aspects of the pathogenesis of hereditary diseases, can also provide hints for understanding the involvement of complex I in sporadic neurological disorders and aging, as well as for developing therapeutical strategies.  相似文献

Chromatophores isolated from cells of Rhodobacter sphaeroides exposed to hypertonic solutions were enriched in cardiolipin (CL). Because CL levels are raised by increasing the incubation time of R. sphaeroides in hypertonic solutions, it was possible to isolate chromatophores containing different CL amounts by starting from cells incubated in hypertonic solutions for different times. The functionality and stability of the photosynthetic proteins in chromatophore membranes having different CL levels were investigated. Reaction center (RC) stabilization with respect to thermal denaturation and photoxidative damage was observed by flash photolysis and fluorescence emission experiments in CL-enriched chromatophores. To gain detailed information about the structures of endogenous CLs, this lipid family was isolated and purified by preparative TLC, and characterized by high-resolution mass spectrometry. We conclude that osmotic shock can be used as a tool to modulate CL levels in isolated chromatophores and to change the composition of the RC lipid annulus, avoiding membrane artifacts introduced by the use of detergents.  相似文献

Background  

Burkholderia cenocepacia are opportunistic Gram-negative bacteria that can cause chronic pulmonary infections in patients with cystic fibrosis. These bacteria demonstrate a high-level of intrinsic antibiotic resistance to most clinically useful antibiotics complicating treatment. We previously identified 14 genes encoding putative Resistance-Nodulation-Cell Division (RND) efflux pumps in the genome of B. cenocepacia J2315, but the contribution of these pumps to the intrinsic drug resistance of this bacterium remains unclear.  相似文献

The second internal transcribed spacer (ITS2) of the nuclear ribosomal RNA cluster (rDNA) is significantly smaller in the Cnidaria (120–260 bp) than in the rest of the Metazoa. ITS2 is one of the fastest evolving DNA regions among those commonly used in molecular systematics and has been proposed as a possible barcoding gene for Cnidaria to replace the currently problematic mitochondrial sequences used. We have reviewed the intraspecific and interspecific variation of ITS2 rRNA sequences in the Anthozoa. We have observed that the lower limits of the interspecific DNA divergence ranges very often overlap with intraspecific ranges, and identical sequences from individuals of different species are not rare. This finding can result in problems similar to those encountered with the mitochondrial COI, and we conclude that ITS2 does not prove significantly better than COI for standard taxonomic DNA barcoding in Anthozoa. However, ITS2 appears to be a promising gene in the ecological DNA barcoding of corallivory, where taxonomic accuracy at genus or even family level may represent a significant improvement of current knowledge. We have successfully amplified and sequenced ITS2 from template DNA extracted from foot muscle and from stomach contents of corallivorous gastropods, and from their anthozoan hosts. The small size of cnidarian ITS2 makes it a very easy and efficient tool for ecological barcoding of associations. Ecological barcoding of corallivory is an indispensable approach to the study of the associations in deep water, where direct observation is severely limited by logistics and costs.  相似文献

Background

Human Papillomavirus (HPV)-16 is a paradigm for “high-risk” HPVs, the causative agents of virtually all cervical carcinomas. HPV E6 and E7 viral genes are usually expressed in these tumors, suggesting key roles for their gene products, the E6 and E7 oncoproteins, in inducing malignant transformation.

Methodology/Principal Findings

By protein-protein interaction analysis, using mass spectrometry, we identified glutathione S-transferase P1-1 (GSTP1) as a novel cellular partner of the HPV-16 E7 oncoprotein. Following mapping of the region in the HPV-16 E7 sequence that is involved in the interaction, we generated a three-dimensional molecular model of the complex between HPV-16 E7 and GSTP1, and used this to engineer a mutant molecule of HPV-16 E7 with strongly reduced affinity for GSTP1.When expressed in HaCaT human keratinocytes, HPV-16 E7 modified the equilibrium between the oxidized and reduced forms of GSTP1, thereby inhibiting JNK phosphorylation and its ability to induce apoptosis. Using GSTP1-deficient MCF-7 cancer cells and siRNA interference targeting GSTP1 in HaCaT keratinocytes expressing either wild-type or mutant HPV-16 E7, we uncovered a pivotal role for GSTP1 in the pro-survival program elicited by its binding with HPV-16 E7.

Conclusions/Significance

This study provides further evidence of the transforming abilities of this oncoprotein, setting the groundwork for devising unique molecular tools that can both interfere with the interaction between HPV-16 E7 and GSTP1 and minimize the survival of HPV-16 E7-expressing cancer cells.  相似文献

Anti-predator behaviour often represents a trade-off between the benefits of reducing predation risk and the drawbacks of limiting access to resources (e.g. food availability, mating and nesting sites). The effectiveness of avoidance behaviour relies on the ability to detect predator cues, which may provide reliable information on predation risk. Using controlled laboratory experiments, we studied the relative importance of visual and chemical cues in the triggering of anti-predator responses in Galaxias maculatus, where Oncorhyncus mykiss was used as the predator. Metabolic cost was also estimated, measured as oxygen consumption. Exposure to different types of predator cue induced diverse behavioural responses in G. maculatus. Detection of the exotic predator, using both visual and chemical stimuli, resulted in reduced G. maculatus swimming activity and changes in respiratory rate.  相似文献

Bacterial proteins belonging to the MocR/GabR family are chimeric proteins incorporating a short N-terminal helix-turn-helix containing domain with DNA-binding properties, and a long C-terminal domain belonging to the superfamily of the pyridoxal-5′-phosphate enzymes of fold type I. The first purpose of this report is to give an overview of the distribution of these factors among the different taxonomical bacterial divisions and to determine the degree of conservation of the main structural features of the PLP binding domain. Complete proteomes of bacteria phyla were scanned with a hidden Markov model representative of the MocR family. Results indicate that presence of MocR factors is heterogeneous even within the single bacterial phylum: some species miss completely the factors, while others possess one or even more regulators. Absence of MocR factors is distinctive of some phyla such as Chlamydiae. The genomic distribution of MocR is, as expected, highly correlated to the size of the genome. At variance, phyla missing MocR regulators generally are characterized by compact genomes, of the order of 1.0–2.0 Mb, such as the case of Mollicutes or Chlamydiae. Apparently, the minimum genome size compatible with the presence of MocR genes is around 2.0–2.5 Mb. Conservation of the residues corresponding to those involved in the interaction with the cofactor pyridoxal-5′-phosphate in the homologous 2-aminoadipate aminotransferase, was analyzed in the multiple sequence alignments of MocR within each phyla considered. In the vast majority of cases, residues are conserved or conservatively replaced. This result suggests that, in most cases, MocR factors preserve at least ability to bind the cofactor and very likely some catalytic abilities.  相似文献
