Quantitative cytological analyses of aging C57BL/6J mouse ovaries revealed that the populations of primordial and growing follicles were nearly exhausted by 13-14 months, the average age of ovulatory failure. Anovulatory animals of this age had, on the average, half the follicle number of their counterparts which were still ovulating. This result suggests that follicular depletion is a factor which contributes to the loss of ovarian cyclicity during aging. However, the considerable overlap of follicle number between the two groups suggests that other, possibly extraovarian, factors also influence the loss of ovulatory function. Although the numbers of follicles recruited for growth was much lower in ovaries from old cycling animals, the number of ova shed cyclically was generally within the range of younger individuals. The observed reduction in incidence of morphological atresia among medium-sized follicles may explain how ovulatory constancy is maintained virtually throughout the cyclical life of the ovary.  相似文献

Binocular neurons with receptive fields about 5 degrees across were recorded just beneath the pia. Most of them responded to dark stimuli in the lower half of their receptive field and to light stimuli above. There was almost no vertical disparity between the left and right fields and the modal value of the horizontal disparity of the population of cells was 1.7 degrees. Because frogs do not verge their eyes it is possible to calculate at what distance the receptive fields through the two eyes are superimposed. This calculation suggests that the neurons are tuned to detect features in the external world about 50 cm away. This is too far for the neurons to be involved in the frog's everyday distance vision. It is more likely that they are concerned with assessing the vertical position of a horizontal surface.  相似文献

Plastids affected by either iojap or chloroplast mutator fail to green, and altered plastids are maternally transmitted to subsequent generations. The ultrastructure of iojap-affected plastids indicates that these plastids contain no ribosomes and are capable of supporting little internal membrane organization in either light or dark-grown plants. Chloroplast mutator-affected plastids of light-grown plants contain some organized internal membrane structures. In dark-grown plants, chloroplast mutator-aftected plastids contain a crystalline prolamellar body, numerous vesicles, and osmiophilic granules. The chloroplast mutator-affecled etioplasts display an abnormal distribution of lamellar membranes; these membranes, rather than radiating in a spokelike pattern from the prolamellar body, are condensed into a portion of the organelle. Light causes disruption of the prolamellar body in chloroplast mutator-affected plastids without promoting the organization of a normal thylakoid membrane system. The effects of iojap and chloroplast mutator are cell autonomous and apparently influence the individual plastid, as evidenced by the persistence of heteroplastidic cells containing normal and affected plastids.  相似文献

Evidence for the occurrence of polyphosphates having apparent chain-lengths ranging from less than 10 to over 5000 orthophosphate units has been found in adult brain as well as in a number of other mammalian tissues which have been examined. There appears to be three times as much polyphosphate in rat brain as there is in rat liver. Adult rat brain appears to contain at least 15 μg of phosphorus as polyphosphate per g of fresh tissue. In addition, neural polyphosphate is extremely labile to catabolic degradation after death whereas hepatic polyphosphate is relatively more stable. The nature of this inorganic polymer was elucidated through use of the technique of 31P nuclear magnetic resonance spectroscopy. Further structural evidence was obtained by application of the isotopic dilution technique to 32P-labelled neural polyphosphate mixed with an abiotically prepared inorganic polyphosphate. The conditions and rates of hydrolysis of the biological polyphosphate and a non-biological polyphosphate were comparable.  相似文献

Enzymic methods of micro assay have been described for four neutral sugars commonly present in glycoproteins and glycolipids. These assays can be applied to glycoprotein hydrolyzates without prior purification of individual sugars.d-Mannose is assayed by first phosphorylating the sugar in the presence of hexokinase and then measuring the formation of ADP by the use of pyruvate kinase and lactic dehydrogenase. This assay is not specific for d-mannose since both d-glucose and d-glucosamine can be phosphorylated by hexokinase. It is possible to remove d-glucosamine prior to hexokinase treatment by passage through a Dowex 50-X8 (H+) column. The effect of d-glucose in the sample can be corrected for by measuring d-glucose with d-glucose-6-phosphate dehydrogenase, an assay which is highly specific for d-glucose.d-Galactose and l-fucose are measured by their respective dehydrogenases. Neither of these dehydrogenases is affected by sugars commonly found in glycoproteins or glycolipids, nor by the presence of a partial acid hydrolyzate of bovine serum albumin. The methods described enable detection of 0.025 μmole of d-mannose, d-glucose, d-galactose, or l-fucose in a glycoprotein digest. The methods can theoretically be made even more sensitive by the use of fluorometric techniques.  相似文献

Differences in the metabolic fate of antithyroid drugs influence the optimal frequency of administration and their therapeutic efficacy. 35S propylthiouracil differed from the 35S imidazoles (carbimazole and methimazole) in the more rapid absorption and excretion and the shorter biological half-life in the plasma of the former. Renal function may have a more important influence on the biological half-life of the drugs than thyroid status. Further work is required to determine the optimal frequency of administration for each compound.  相似文献

The thinnest areas of the capillaries of the choroid plexus and ciliary processes in the eye of the rabbit are characterized by the presence of fenestrae. When various colloidal particles opaque to the electron beam (thorotrast, gold sol, and saccharated iron oxide) were injected into the blood stream, none were found in fenestrae or in areas that might suggest their having passed through fenestrae. The passage of marker particles from the lumen to the surrounding connective tissue does take place on occasion in the areas of thicker walls in the capillaries and venules rather than in the attenuated and fenestrated endothelial walls. The pathway taken by these markers may be either through the cytoplasm of the endothelial cells via membrane-bounded vesicles and vacuoles or through the intercellular spaces of the vessels. An altered aqueous humor (cloudy and plasmoid) was produced by endotoxin injection or by making a draining fistula in rabbit cornea. Both methods gave rise to the same changes in the blood vessels of the ciliary processes. Under such conditions of inflammation the passage of colloidal particles through the thicker walls of the capillaries and venules was greatly increased and occurred primarily as an intercellular passage between the endothelial cells. The attenuated and fenestrated areas of the endothelium of the small capillaries remained unchanged with no particles passing through them. These results on the altered vessels of the ciliary processes parallel those of Majno and Palade (26) on the rat cremaster muscle.  相似文献

Ion Effects on Calcium Accumulation by Cardiac Sarcoplasmic Reticulum   总被引:6,自引:3,他引:3  
The effects of monovalent cations on the active calcium-accumulating ability of cardiac sarcoplasmic reticulum were assessed. Grana prepared in an ion-free system accumulated calcium when ATP and Mg++ were present. Sodium ion and to a lesser extent lithium but not K+ reduced the amount of calcium taken up. The reduction of calcium binding by Na+ is not due to inhibition of uptake but to a rapid release of the radiocalcium bound. The amount of calcium released by sodium does not appear to be enough to explain contraction on the basis of sodium influx into muscle, but may be significant in the regulation of tension.  相似文献
