Spores ofBacillus cereus (strain NCIB 8122) were germinated in a synthetic germination limited medium (GL-medium), which permitted germination but did not make the termination of post-germinative development possible. Incorporation of14C-diaminopimelic acid into the newly formed cell wall was followed in this culture. Morphological changes were studied by optical and electron microscopy. Germination was associated with the usual germination changes,i.e. depolymerization of the “bulky” cortex, differentiation of nuclear structure and mesosomes and ribosomes in the cytoplasm. At this stage the spore protoplast is surrounded by several layers: exosporium, laminated coat with four layers, residual spore wall and the protoplast membrane. During incubation in this limited medium the residual wall layer thickens and the nuclear structure, mesosomes and ribosomes were not more detectable. After enrichment of the GL medium (shift up) the thick-walled cells can form additional cell wall material, elongate and an atypical septum formation can occur. The cell wall material forms local thickenings. On long-term cultivation in the GL medium some of the cells in the GL medium lyze. If, in the course of 3–6 h the cells are transferred from the GL-medium to a solid complex medium (Difco Nutrient Agar) the thickwalled cells are transformed into dividing cells. When the cells are transferred later, their colony-forming ability rapidly decreases. The decrease of viability of the thick-walled cells derived directly from spores after their germination in the limited medium indicates that these cellular forms probably do not represent more stable cellular types that would be of considerable importance for survival of the populat ion of bacilli.  相似文献

Rabbit antibodies against the Mn-dependent DNAse were obtained. The specificity of the anti-DNAse antibody was established by the enzyme inhibition in vitro. The enzyme activity was inhibited by more than 75 and 54% using rabbit antisera and affinity-purified IgG. Localization of the enzyme in the rat hepatocyte nuclei was studied by indirect immunofluorescence.  相似文献

Using stepwise extraction of chromatin from Candida tropicalis by NaCl (0.1-1.0 M) the protein dissociated by 0.3 and 0.6 M NaCl (fractions 0.3 and 0.6) possessing the DNAase activity were obtained. These DNAases are activated by Mg2+ and cause preferential hydrolysis of heat-denaturated DNA. Fraction 0.3 DNAase has a maximum at neutral values of pH (around 7.0) and causes endonucleolytic hydrolysis of DNA. Fraction 0.6 DNAase causes exonucleolytic hydrolysis of DNA but a maximum at alkaline pH (8.0). The properties of isolated chromatin DNAases of Candida tropicalis differ from those of the known DNAases of the yeast Saccharomyces cerevisiae.  相似文献

Degeneration of the tactile cells in epithelium of the cat sinuous hairs after sectioning the infraorbital nerve manifests itself as cytoplasmic vacuolization and induration with electron opaque bodies in it, changes in nuclear configuration and in chromatin density. At all stages of the experiment the tactile cells were practically observed demonstrating various degenerative signs. On the 45th day after sectioning the tactile cells disappeared completely. Structural changes in the tactile cells were preceded by degeneration and complete disappearance of tactile menisci from epithelium of the sinuous hairs. Perhaps, sensitive innervation is necessary to maintain differential status of the tactile cells in epithelium.  相似文献

Organ explants from the canine prostate with and without methylprednisolone pretreatment were cultivated for ten days in Trowell's T-8 medium or medium supplemented with testosterone and/or vitamin A. Upon termination of the experiments, explants were fixed and examined histologically. All glands from the most central section of each explant were graded according to epithelial type, and from these grades the proportion of acini with maintained columnar cells was calculated for each explant. Stromal cellular maintenance was also estimated. While a small proportion of explants from the vitamin-treated and methylprednisolone-pretreated groups showed epithelial maintenance, a combination of these treatments significantly increased such maintenance. Stromal maintenance was enhanced with methylprednisolone pretreatment but not by vitamin A. These results are in accord with the hypothesis that methylprednisolone acts to stabilize the lysosomal membrane, thus protecting tissue against the effects of ischemic shock. In protected explants vitamin A is able to maintain a columnar glandular epithelium. In a subsequent experiment a series of linoleic acid dosages was tested in the presence or absence of vitamin A. In neither case was this fatty acid of value in improving epithelial or stromal maintenance.  相似文献
